    Phosphorus and nitrogen limitation of primary production in a simulated estuarine gradient

    The transition between phosphorus limitation of primary production in freshwater and nitrogen limitation in seawater was examined along an estuarine gradient simulated in 4 large 13 m3 enclosures connected in a series and containing pelagic and benthic subsystems. Nominal salinities of 0, 5, 10 and 25 ppt were maintained by exchanging appropriate volumes of water between enclosures. River water, which served as a freshwater endmember, was naturally high in N relative to P, while the oceanic endmember (water from Narragansett Bay, RI, USA) was low in N relative to P. Production in the water column was supported by external inputs and recycled nutrients. Bioassays, inorganic nutrient concentrations and N:P ratios of the seston and inorganic nutrients indicated that phosphorus was limiting at 0, 5 and 10 ppt, while nitrogen was limiting at 25 ppt. Coincident with this shift in limiting nutrient was a shift in the N:P ratio of nutrient supply from greater than the Redfield ratio of 16 to less than 16. External inputs established relative rates of supply in each enclosure. The relative proportion of N and P in external inputs was largely a function of the hydrodynamic mixing of fresh (high N, low P) and salt water (low N, high P) endmembers. At the scale of the estuarine segment or enclosure, neither recycled inputs from the benthos and water column, nitrogen fixation nor internal losses of N and P to sedimentation and/or denitrification materially altered relative supply rates, despite a hydrodynamic residence time of 27 d

    The aim of this study is to understand the implications of asynchronous approvals for genetically modified organisms (GMOs) that are imported to the European Union for use within animal feed products, specifically with regard to the EU livestock sector, as well as upon the upstream and downstream economic industries related to it. Asynchronous approval refers to the situation in which there is a delay in the moment when a genetically modified (GM) event – modifying a specific trait of a plant or animal – is allowed to be used in one country in comparison to another country. In the perspective of this study, the asynchronous GMO approvals concern the use of GM varieties of plants that are approved in the countries which supply them to the EU, in one form or another of feed material, before these are approved by the EU

    Epigenetic Suppression of Transgenic T-cell Receptor Expression via Gamma-Retroviral Vector Methylation in Adoptive Cell Transfer Therapy

    Transgenic T-cell receptor (TCR) adoptive cell therapies recognizing tumor antigens are associated with robust initial response rates, but frequent disease relapse. This usually occurs in the setting of poor long-term persistence of cells expressing the transgenic TCR, generated using murine stem cell virus (MSCV) y-retroviral vectors. Analysis of clinical transgenic adoptive cell therapy products in vivo revealed that despite strong persistence of the transgenic TCR DNA sequence over time, its expression was profoundly decreased over time at the RNA and protein levels. Patients with the greatest degrees of expression suppression displayed significant increases in DNA methylation over time within the MSCV promoter region, as well as progressive increases in DNA methylation within the entire MSCV vector over time. These increases in vector methylation occurred independently of its integration site within the host genomes. These results have significant implications for the design of future viral-vector gene engineered adoptive cell transfer therapies

    Twisted Classical Poincar\'{e} Algebras

    We consider the twisting of Hopf structure for classical enveloping algebra U(g^)U(\hat{g}), where g^\hat{g} is the inhomogenous rotations algebra, with explicite formulae given for D=4D=4 Poincar\'{e} algebra (g^=P4).(\hat{g}={\cal P}_4). The comultiplications of twisted UF(P4)U^F({\cal P}_4) are obtained by conjugating primitive classical coproducts by F∈U(c^)⊗U(c^),F\in U(\hat{c})\otimes U(\hat{c}), where c^\hat{c} denotes any Abelian subalgebra of P4{\cal P}_4, and the universal R−R-matrices for UF(P4)U^F({\cal P}_4) are triangular. As an example we show that the quantum deformation of Poincar\'{e} algebra recently proposed by Chaichian and Demiczev is a twisted classical Poincar\'{e} algebra.     Effective supergravity descriptions of superstring cosmology

    This text is a review of aspects of supergravity theories that are relevant in superstring cosmology. In particular, it considers the possibilities and restrictions for `uplifting terms', i.e. methods to produce de Sitter vacua. We concentrate on N=1 and N=2 supergravities, and the tools of superconformal methods, which clarify the structure of these theories.     The role for saxagliptin within the management of type 2 diabetes mellitus: an update from the 2010 European Association for the Study of Diabetes (EASD) 46th annual meeting and the American Diabetes Association (ADA) 70th scientific session

    Saxagliptin is a potent, selective DPP4 inhibitor. Highlights from abstracts presented at the 2010 meetings of the European Association for the Study of Diabetes and the American Diabetes Association include studies and analyses that shed light on the promising role for saxagliptin within the management of type 2 diabetes mellitus. Data show that saxagliptin combination therapy improves HbA1c levels compared with placebo, particularly in patients with high HbA1c at baseline, long duration of disease, low baseline creatinine clearance, and low homeostasis model assessment 2 ÎČ-cell function at baseline. These efficacy benefits are achieved without any increase in hypoglycemia or other adverse events. The study results also show that the saxagliptin plus metformin combination is a good candidate for initial therapy in drug-naĂŻve patients treated for as long as 72 weeks. Survey data presented confirm that hypoglycemia (and fear of hypoglycemia) is a barrier to patients' acceptance of diabetes treatment, limiting its efficacy. Therefore, therapies such as saxagliptin that have a low risk of hypoglycemia may be more acceptable to patients in helping them to achieve glycemic control and to optimize their quality of life. In patients with renal impairment, for whom metformin is contraindicated, saxagliptin monotherapy is a promising option for antidiabetic management as, when given at a reduced dose, it is well-tolerated with a safety profile similar to that of placebo

    Water Abundance of Dunes in Gale Crater, Mars From Active Neutron Experiments and Implications for Amorphous Phases

    We report the water abundance of Bagnold Dune sand in Gale crater, Mars by analyzing active neutron experiments using the Dynamic Albedo of Neutrons instrument. We report a bulk water‐equivalent‐hydrogen abundance of 0.68 ± 0.15 wt%, which is similar to measurements several kilometers away and from those taken of the dune surface. Thus, the dune is likely dehydrated throughout. Furthermore, we use geochemical constraints, including bulk water content, to develop compositional models of the amorphous fraction for which little information is known. We find the amorphous fraction contains ∌26‐ to 64‐wt% basaltic glass and up to ∌24‐wt% rhyolitic glass, suggesting at least one volcanic source for the dune material. We also find a range of hydrated phases may be present in appreciable abundances, either from the incorporation of eroded aqueously altered sediments or the direct alteration of the dune sand

    Application of unsupervised chemometric analysis and self-organising feature map (SOFM) for the classification of lighter fuels

    A variety of lighter fuel samples from different manufacturers (both unevaporated and evaporated) were analysed using conventional gas chromatography-mass spectrometry (GC-MS) analysis. In total 51 characteristic peaks were selected as variables and subjected to data pre-processing prior to subsequent analysis using unsupervised chemometric analysis (PCA and HCA) and a SOFM artificial neural network. The results obtained revealed that SOFM acted as a powerful means of evaluating and linking degraded ignitable liquid sample data to their parent unevaporated liquids

    A survey among dermatologists: diagnostics of superficial fungal infections - what is used and what is needed to initiate therapy and assess efficacy?

    BACKGROUND: Superficial fungal infections are common. It is important to confirm the clinical diagnosis by mycological laboratory methods before initiating systemic antifungal treatment, especially as antifungal sensitivity and in vitro susceptibility may differ between different genera and species. For many years, the gold standard for diagnosis of superficial fungal infections has been direct fungal detection in the clinical specimen (microscopy) supplemented by culturing. Lately, newer molecular based methods for fungal identification have been developed. OBJECTIVE: This study was initiated to focus on the current usage of mycological diagnostics for superficial fungal infections by dermatologists. It was designed to investigate whether it was necessary to differentiate between initial diagnostic tests and those used at treatment follow-up in specific superficial fungal infections. METHODS: An online questionnaire was distributed among members of the EADV mycology Task Force and other dermatologists with a special interest in mycology and nail disease. RESULTS: The survey was distributed to 62 dermatologists of whom 38 (61%) completed the whole survey, 7 (11%) partially completed and 17 (27%) did not respond. Nearly, all respondents (82-100%) said that ideally they would use the result of direct microscopy (or histology) combined with a genus/species directed treatment of onychomycosis, dermatophytosis, Candida- and Malassezia-related infections. The majority of the dermatologists used a combination of clinical assessment and direct microscopy for treatment assessment and the viability of the fungus was considered more important at this visit than when initiating the treatment. Molecular based methods were not available for all responders. CONCLUSION: The available diagnostic methods are heterogeneous and their usage differs between different practices as well as between countries. The survey confirmed that dermatologists find it important to make a mycological diagnosis, particularly prior to starting oral antifungal treatment in order to confirm the diagnose and target the therapy according to genus and species
