    Feature Tracking Cardiac Magnetic Resonance via Deep Learning and Spline Optimization

    Feature tracking Cardiac Magnetic Resonance (CMR) has recently emerged as an area of interest for quantification of regional cardiac function from balanced, steady state free precession (SSFP) cine sequences. However, currently available techniques lack full automation, limiting reproducibility. We propose a fully automated technique whereby a CMR image sequence is first segmented with a deep, fully convolutional neural network (CNN) architecture, and quadratic basis splines are fitted simultaneously across all cardiac frames using least squares optimization. Experiments are performed using data from 42 patients with hypertrophic cardiomyopathy (HCM) and 21 healthy control subjects. In terms of segmentation, we compared state-of-the-art CNN frameworks, U-Net and dilated convolution architectures, with and without temporal context, using cross validation with three folds. Performance relative to expert manual segmentation was similar across all networks: pixel accuracy was ~97%, intersection-over-union (IoU) across all classes was ~87%, and IoU across foreground classes only was ~85%. Endocardial left ventricular circumferential strain calculated from the proposed pipeline was significantly different in control and disease subjects (-25.3% vs -29.1%, p = 0.006), in agreement with the current clinical literature.Comment: Accepted to Functional Imaging and Modeling of the Heart (FIMH) 201

    Structure of the regulatory hyaluronan binding domain in the inflammatory leukocyte homing receptor CD44

    Adhesive interactions involving CD44, the cell surface receptor for hyaluronan, underlie fundamental processes such as inflammatory leukocyte homing and tumor metastasis. Regulation of such events is critical and appears to be effected by changes in CD44 N-glycosylation that switch the receptor "on" or "off" under appropriate circumstances. How altered glycosylation influences binding of hyaluronan to the lectin-like Link module in CD44 is unclear, although evidence suggests additional flanking sequences peculiar to CD44 may be involved. Here we show using X-ray crystallography and NMR spectroscopy that these sequences form a lobular extension to the Link module, creating an enlarged HA binding domain and a formerly unidentified protein fold. Moreover, the disposition of key N-glycosylation sites reveals how specific sugar chains could alter both the affinity and avidity of CD44 HA binding. Our results provide the necessary structural framework for understanding the diverse functions of CD44 and developing novel therapeutic strategies

    How do I sound to me? Perceived changes in communication in Parkinson's disease

    Objective: To examine self and carer perceived changes in communication associated with Parkinson's disease and relate these to speech intelligibility, gender, age and other disease measures. Design: Cross-sectional survey of a hospital- and community-based sample of 176 people with Parkinson's disease and their carers using a questionnaire based on semantic differential techniques. Participants: One hundred and four people with Parkinson's disease with no history of communication difficulties prior to onset of their Parkinson's disease and 45 primary carers who returned completed questionnaires. Main outcome measures: Differences in ratings for `before' the onset of Parkinson's disease versus present status. Results: There was a strong perception of negative impact on communication between `before' and `now', irrespective of age and gender and largely independent of disease severity and duration, intelligibility and cognitive status. Activities of daily living (assessed by Unified Parkinson's Disease Rating Scale (UPDRS) II) and depression rating scale scores had the strongest association with change (adjusted R 2 0.27). There was a significant correlation between the rank order of perceived change in features examined in people with Parkinson's disease versus their carers, though in general carers rated change as having less impact. Conclusions: Parkinson's disease exercises a strong influence on communication even before apparent alterations to intelligibility or motor status

    The future of image-guided radiotherapy-is image everything?

    MR-based image-guided (IG) radiotherapy via all-in-one MR treatment units (MR-linacs) is one of the hottest topics in contemporary radiotherapy research. From ingenious engineering solutions to complex physical problems, researchers have developed machines with the promise of superior image quality, and all the advantages this may confer. Benefits include better tumour visualisation, online adaptation and the potential for image biomarker-based personalised RT. However, it is important to remember that the technical challenges are real. In many instances, they are skillfully managed rather than abolished, a point illustrated by the wide variety of MR-linac designs. The proposed benefits also deserve careful inspection. Better visibility of the primary tumour on an IG scan cannot be bad, but does not automatically equate to better IG, which often depends on a more generalised match to daily anatomy. MR-linac will undoubtedly be a rich milieu to search for IMBs, but these will need to be carefully validated, and similar work with CT-based biomarkers using existing, cheaper, and more widely available hardware is currently ongoing. Online adaptation is an attractive concept, but practicalities are complex, and more work is required to understand which patients will benefit from plan adaptation, and when. Finally, the issue of cost cannot be overlooked, nor can the research community's responsibilities to global healthcare inequalities. MR-linac is an exciting and ingenious technology, which merits both investment and research. It may not, however, have the future to itself

    Precocial juvenile lizards show adult level learning and behavioural flexibility

In precocial species, young are born at a more advanced developmental stage in need of less or no parental care and brain development is mostly completed at the time of birth. We therefore predicted early maturation of learning ability in precocial species. We used a series of visual discrimination and reversal stages to investigate the ability of the precocial eastern blue-tongue lizard, Tiliqua scincoides scincoides, a long-lived Australian lizard species with slow-developing young, to respond to changes in stimulus relevance and test for behavioural flexibility. To test whether age affects learning in this species, we compared juveniles (23–56 days) with adults (sexually mature, at least 2 years). In accordance with our expectations, adults and juveniles performed similarly well in all stages, suggesting that juveniles of this precocial species learn at adult levels from an early age. Both age classes performed well during reversals showing good behavioural flexibility. This is the first study in lizards to directly compare juvenile and adult behavioural flexibility.     Face pareidolia in products : the effect of emotional content on attentional capture, eagerness to explore, and likelihood to purchase

    Face‐like configurations can be perceived in everyday products. This perceptual phenomenon is known as face pareidolia. However, few studies have investigated the perception of pareidolic emotion in such products and the effect it could have on consumer behaviour. Therefore, in this study, across two experiments, we test the extent to which participants perceive core human emotions in products with pareidolic configurations (Experiment 1), and how this affects key consumer metrics (i.e., likely attentional capture, eagerness to explore, likelihood to purchase; Experiment 2). The findings show that these products do elicit the full range of affective content, with variation in perceived emotional intensity. Products with ‘happy’, ‘angry’ and ‘surprise’ configurations were likely to capture attention/promote product exploration, but only ‘happy’ products retained this advantage for purchasing decisions. Individual differences in mood and level of loneliness predicted likely engagement with these products. The theoretical and practical implications of these findings are discussed
