Clinical dilemma and systemic treatment strategy of triple-negative breast cancer in the elderly

    Get PDF
There is no specific biomarker for triple-negative breast cancer (TNBC), and chemotherapy remains as backbone but with limited efficacy and more side effects. 10%-21% TNBC are elderly patients with high prevalence of concomitant cardio-cerebrovascular and renal complications, which may lead to intolerance of chemotherapy. How to properly treat these elderly TNBC patients becomes a big challenge during daily clinical practice. To date, few clinical trials specifically focus on elderly TNBC patients, thus no enough evidence-based safety and efficacy data are provided to support the proper therapy to this population. There are also challenges and controversies in the diagnosis and treatment. Elderly TNBC patients have special age-related disease characteristics and high non-cancer related mortality. Therefore, it is very important to balance survival benefit and quality of life during the treatment. This paper summarized data of the epidemiology, tumor biological behavior, current diagnosis and treatment status and the huge unmet medical needs of elderly TNBC patients, and explored the benefits of novel antibody-drug conjugate (ADC), to provide certain guidance on systemic treatment strategies for elderly TNBC patients

Ankyrin Repeats of ANKRA2 Recognize a PxLPxL Motif on the 3M Syndrome Protein CCDC8

SummaryPeptide motifs are often used for protein-protein interactions. We have recently demonstrated that ankyrin repeats of ANKRA2 and the paralogous bare lymphocyte syndrome transcription factor RFXANK recognize PxLPxL/I motifs shared by megalin, three histone deacetylases, and RFX5. We show here that that CCDC8 is a major partner of ANKRA2 but not RFXANK in cells. The CCDC8 gene is mutated in 3M syndrome, a short-stature disorder with additional facial and skeletal abnormalities. Two other genes mutated in this syndrome encode CUL7 and OBSL1. While CUL7 is a ubiquitin ligase and OBSL1 associates with the cytoskeleton, little is known about CCDC8. Binding and structural analyses reveal that the ankyrin repeats of ANKRA2 recognize a PxLPxL motif at the C-terminal region of CCDC8. The N-terminal part interacts with OBSL1 to form a CUL7 ligase complex. These results link ANKRA2 unexpectedly to 3M syndrome and suggest novel regulatory mechanisms for histone deacetylases and RFX7

Exploring the Sustainability of Urban Leisure Agriculture in Shanghai

Leisure agriculture is an essential part of urban agriculture in Shanghai. However, sustainable development for urban leisure agriculture has reached a critical point. In this paper, we attempted to analyze the sustainability status of 22 urban leisure farms in Shanghai using the IDEA (Indicateurs de Durabilité des Exploitations Agricoles) method for sustainability indicators. From this analysis, we found out that farms’ average sustainability scores were 25.72 on the agroecological scale, 32.5 on the socioterritorial scale, and 46.5 on the economic scale. This proved that urban leisure agriculture in Shanghai has high sustainability at the economic scale, followed by the socioterritorial scale and the agroecological scale. However, the overall sustainability of urban leisure agriculture in Shanghai was low, which indicates that Shanghai’s urban agriculture still needs to be strengthened for sustainability. Thus, this paper concludes with some policy recommendations for the future development of urban leisure agriculture in Shanghai

The Impact of Carbon Emissions Trading on the Profitability and Debt Burden of Listed Companies

The high-quality development of China’s green finance is driven by the aims of reaching peak carbon and carbon neutrality, leading to an important impact of emissions trading policies on the profitability and debt burden of China’s listed companies. This paper uses propensity score matching (PSM) and the difference in differences (DID) method to study the impact of carbon emissions trading policies on the profitability and debt burden of listed companies in seven carbon emissions trading pilots from 2010 to 2019. The empirical evidence shows that the implementation of carbon emissions trading policies has increased the profitability and reduced the debt burden of listed companies: (1) Mechanism test shows that this policy can increase the profitability and debt burden of state-owned listed companies by increasing innovation investment. (2) The negative effects of this policy on the profitability and debt burden of state-owned listed companies are more obvious than that of the others. (3) The policy has also significantly reduced the debt burden of competitive companies and the profitability of monopolies. (4) The policy yields a more intense reduction in the debt burden of listed companies in high-carbon emission industries. Finally, this paper suggests that listed companies’ participation in the carbon emissions trading policy be expanded and that monopoly conditions among listed companies are reduced
