An orientation-space super sampling technique for six-dimensional diffraction contrast tomography

Diffraction contrast tomography (DCT) is an X-ray full-field imaging technique that allows for the non-destructive three-dimensional investigation of polycrystalline materials and the determination of the physical and morphological properties of their crystallographic domains, called grains. This task is considered more and more challenging with the increasing intra-granular deformation, also known as orientation-spread. The recent introduction of a sixdimensional reconstruction framework in DCT (6D-DCT) has proven to be able to address the intra-granular crystal orientation for moderately deformed materials. The approach used in 6D-DCT, which is an extended sampling of the six-dimensional combined position-orientation space, has a linear scaling between the number of sampled orientations, which determine the orientation-space resolution of the problem, and computer memory usage. As a result, the reconstruction of more deformed materials is limited by their high resource requirements from a memory and computational point of view, which can easily become too demanding for the currently available computer technologies. The plasma membrane calcium/calmodulin-dependent calcium ATPase (PMCA) (Shull, G.E., and J. Greeb. 1988. J. Biol. Chem. 263:8646–8657; Verma, A.K., A.G. Filoteo, D.R. Stanford, E.D. Wieben, J.T. Penniston, E.E. Strehler, R. Fischer, R. Heim, G. Vogel, S. Mathews, et al. 1988. J. Biol. Chem. 263:14152–14159; Carafoli, E. 1997. Basic Res. Cardiol. 92:59–61) has been proposed to be a regulator of calcium homeostasis and signal transduction networks of the cell. However, little is known about its precise mechanisms of action. Knock-out of (mainly neuronal) isoform 2 of the enzyme resulted in hearing loss and balance deficits due to severe inner ear defects, affecting formation and maintenance of otoconia (Kozel, P.J., R.A. Friedman, L.C. Erway, E.N. Yamoah, L.H. Liu, T. Riddle, J.J. Duffy, T. Doetschman, M.L. Miller, E.L. Cardell, and G.E. Shull. 1998. J. Biol. Chem. 273:18693–18696). Here we demonstrate that PMCA 4b is a negative regulator of nitric oxide synthase I (NOS-I, nNOS) in HEK293 embryonic kidney and neuro-2a neuroblastoma cell models. Binding of PMCA 4b to NOS-I was mediated by interaction of the COOH-terminal amino acids of PMCA 4b and the PDZ domain of NOS-I (PDZ: PSD 95/Dlg/ZO-1 protein domain). Increasing expression of wild-type PMCA 4b (but not PMCA mutants unable to bind PDZ domains or devoid of Ca2+-transporting activity) dramatically downregulated NO synthesis from wild-type NOS-I. A NOS-I mutant lacking the PDZ domain was not regulated by PMCA, demonstrating the specific nature of the PMCA–NOS-I interaction. Elucidation of PMCA as an interaction partner and major regulator of NOS-I provides evidence for a new dimension of integration between calcium and NO signaling pathways

What does the labour market look like in 20 years? The enquiry combines qualitative future scenarios and mathematical processes to provide a qualified prognosis. It also challenges what politicians, employers and labourers need to do in order to overcome the demographic change and to persist in the international competition. With its investigation of 44 industrial sectors, 88 occupational groups and 27 specialisms within professional education, the report offers a detailed view, which goes far beyond any previous outlooks. The results show that Germany will change from a service economy to becoming a knowledge economy, with new positions available in corporate services, financial services as well as education, health and social services. Employment positions will be lost within the manufacturing industry, trade, transport and public services. X-ray orientation microscopy using topo-tomography and multi-mode diffraction contrast tomography

Polycrystal orientation mapping techniques based on full-field acquisition schemes like X-ray Diffraction Contrast Tomography and certain other variants of 3D X-ray Diffraction or near-field High Energy Diffraction Microscopy enable time efficient mapping of 3D grain microstructures. The spatial resolution obtained with this class of monochromatic beam X-ray diffraction imaging approaches remains typically below the ultimate spatial resolution achievable with X-ray imaging detectors. Introducing a generalised reconstruction framework enabling the combination of acquisitions with different detector pixel size and sample tilt settings provide a pathway towards 3D orientation mapping with a spatial resolution approaching the one of state of the art X-ray imaging detector systems

The third long-term forecast by Economix estimates the implications of the recent influx of refugees on the German labour market as well as the qualification structure among the employees. At the same time, it forecasts the opportunities and risks of digitalisation. All analyses come to the conclusion that the integration of refugees into the labour market represents the primary task of the coming five to ten years. Indeed, the immigration of refugees rejuvenates the population. However, it also causes a decline in the qualification structure of labour supply. The authors emphasise both the necessity of extensive and systematic further education programs for all employees as well as the importance of developing educational services for refugees and encouraging them to participate in vocational education. The digitalisation scenario shows considerable increases in growth and employment as a consequence of the use of digital technologies. However, this positive development requires a high acceptance of digital progress, the improvement of digital skills, the support of the fourth industrial revolution to ensure the survival of the technological leadership of the capital goods industry, and the demand for products which require the implementation and use of digital technologies.
