    Evaluation of synthetic vascular grafts in a mouse carotid grafting model

    Current animal models for the evaluation of synthetic grafts are lacking many of the molecular tools and transgenic studies available to other branches of biology. A mouse model of vascular grafting would allow for the study of molecular mechanisms of graft failure, including in the context of clinically relevant disease states. In this study, we comprehensively characterise a sutureless grafting model which facilitates the evaluation of synthetic grafts in the mouse carotid artery. Using conduits electrospun from polycaprolactone (PCL) we show the gradual development of a significant neointima within 28 days, found to be greatest at the anastomoses. Histological analysis showed temporal increases in smooth muscle cell and collagen content within the neointima, demonstrating its maturation. Endothelialisation of the PCL grafts, assessed by scanning electron microscopy (SEM) analysis and CD31 staining, was near complete within 28 days, together replicating two critical aspects of graft performance. To further demonstrate the potential of this mouse model, we used longitudinal non-invasive tracking of bone-marrow mononuclear cells from a transgenic mouse strain with a dual reporter construct encoding both luciferase and green fluorescent protein (GFP). This enabled characterisation of mononuclear cell homing and engraftment to PCL using bioluminescence imaging and histological staining over time (7, 14 and 28 days). We observed peak luminescence at 7 days post-graft implantation that persisted until sacrifice at 28 days.     Two New Candidate Planets in Eccentric Orbits

    Doppler measurements of two G-type main-sequence stars, HD210277 and HD168443, reveal Keplerian variations that imply the presence of companions with masses (M sin i) of 1.28 and 5.04 M_Jup and orbital periods of 437 d and 58 d, respectively. The orbits have large eccentricities of e=0.45 and e=0.54, respectively. All 9 known extrasolar planet candidates with a=0.2-2.5 AU have orbital eccentricities greater than 0.1, higher than that of Jupiter (e=0.05). Eccentric orbits may result from gravitational perturbations imposed by other orbiting planets or stars, by passing stars in the dense star-forming cluster, or by the protoplanetary disk. Based on published studies and our near-IR adaptive optics images, HD210277 appears to be a single star.     A fast radio burst with a low dispersion measure

    Fast radio bursts (FRBs) are millisecond pulses of radio emission of seemingly extragalactic origin. More than 50 FRBs have now been detected, with only one seen to repeat. Here we present a new FRB discovery, FRB 110214, which was detected in the high latitude portion of the High Time Resolution Universe South survey at the Parkes telescope. FRB 110214 has one of the lowest dispersion measures of any known FRB (DM = 168.9±\pm0.5 pc cm−3^{-3}), and was detected in two beams of the Parkes multi-beam receiver. A triangulation of the burst origin on the sky identified three possible regions in the beam pattern where it may have originated, all in sidelobes of the primary detection beam. Depending on the true location of the burst the intrinsic fluence is estimated to fall in the range of 50 -- 2000 Jy ms, making FRB 110214 one of the highest-fluence FRBs detected with the Parkes telescope. No repeating pulses were seen in almost 100 hours of follow-up observations with the Parkes telescope down to a limiting fluence of 0.3 Jy ms for a 2-ms pulse. Similar low-DM, ultra-bright FRBs may be detected in telescope sidelobes in the future, making careful modeling of multi-beam instrument beam patterns of utmost importance for upcoming FRB surveys.Comment: 8 pages, 3 figures, accepted for publication in MNRA

    Actuation of Micro-Optomechanical Systems Via Cavity-Enhanced Optical Dipole Forces

    We demonstrate a new type of optomechanical system employing a movable, micron-scale waveguide evanescently-coupled to a high-Q optical microresonator. Micron-scale displacements of the waveguide are observed for milliwatt(mW)-level optical input powers. Measurement of the spatial variation of the force on the waveguide indicates that it arises from a cavity-enhanced optical dipole force due to the stored optical field of the resonator. This force is used to realize an all-optical tunable filter operating with sub-mW control power. A theoretical model of the system shows the maximum achievable force to be independent of the intrinsic Q of the optical resonator and to scale inversely with the cavity mode volume, suggesting that such forces may become even more effective as devices approach the nanoscale.Comment: 4 pages, 5 figures.     Ferrets exclusively synthesize Neu5Ac and express naturally humanized influenza A virus receptors

    Mammals express the sialic acids ​N-acetylneuraminic acid (​Neu5Ac) and ​N-glycolylneuraminic acid (​Neu5Gc) on cell surfaces, where they act as receptors for pathogens, including influenza A virus (IAV). ​Neu5Gc is synthesized from ​Neu5Ac by the enzyme cytidine monophosphate-N-acetylneuraminic acid hydroxylase (CMAH). In humans, this enzyme is inactive and only ​Neu5Ac is produced. Ferrets are susceptible to human-adapted IAV strains and have been the dominant animal model for IAV studies. Here we show that ferrets, like humans, do not synthesize ​Neu5Gc. Genomic analysis reveals an ancient, nine-exon deletion in the ferret CMAH gene that is shared by the Pinnipedia and Musteloidia members of the Carnivora. Interactions between two human strains of IAV with the sialyllactose receptor (sialic acid—α2,6Gal) confirm that the type of terminal sialic acid contributes significantly to IAV receptor specificity. Our results indicate that exclusive expression of ​Neu5Ac contributes to the susceptibility of ferrets to human-adapted IAV strains

    Proper Motions in the Galactic Bulge: Plaut's Window

    A proper motion study of a field of 20' x 20' inside Plaut's low extinction window (l,b)=(0 deg,-8 deg), has been completed. Relative proper motions and photographic BV photometry have been derived for ~21,000 stars reaching to V~20.5 mag, based on the astrometric reduction of 43 photographic plates, spanning over 21 years of epoch difference. Proper motion errors are typically 1 mas/yr and field dependent systematics are below 0.2 mas/yr. Cross-referencing with the 2MASS catalog yielded a sample of ~8,700 stars, from which predominantly disk and bulge subsamples were selected photometrically from the JH color-magnitude diagram. The two samples exhibited different proper-motion distributions, with the disk displaying the expected reflex solar motion as a function of magnitude. Galactic rotation was also detected for stars between ~2 and ~3 kpc from us. The bulge sample, represented by red giants, has an intrinsic proper motion dispersion of (sigma_l,sigma_b)=(3.39, 2.91)+/-(0.11,0.09) mas/yr, which is in good agreement with previous results, and indicates a velocity anisotropy consistent with either rotational broadening or tri-axiality. A mean distance of 6.37^{+0.87}_{-0.77} kpc has been estimated for the bulge sample, based on the observed K magnitude of the horizontal branch red clump. The metallicity [M/H] distribution was also obtained for a subsample of 60 bulge giants stars, based on calibrated photometric indices. The observed [M/H] shows a peak value at [M/H]~-0.1 with an extended metal poor tail and around 30% of the stars with supersolar metallicity. No change in proper motion dispersion was observed as a function of [M/H]. We are currently in the process of obtaining CCD UBVRI photometry for the entire proper-motion sample of ~21,000 stars.Comment: Submitted to AJ April 17th 2007.
