    The Lived Experiences of the Aging of Hong Kong Chinese Baby Boomers Residing in the United States

    Rapid aging of the majority and minority populations is occurring in the United States. Diverse aspects of aging, concerns of the aging of the baby boomer generation, and the heterogeneities among aging persons from diverse races and ethnicities have been examined in detail; however, there is a void in the literature that offers information about the aging of Chinese baby boomers. This qualitative study used a hermeneutic-phenomenological method to investigate the aging experiences of Hong Kong Chinese baby boomers who are residing in the United States. The findings of this study will possibly enhance the literature about the cultural parameters that relate to these persons\u27 aging. The information can benefit counselors whose work involves this population. The results of this study are consistent, highlighting Hong Kong baby boomers\u27 sensitivities to health issues, their perceived physiological changes and change in personality attributes, their desires to strengthen relationships with families and friends and to practice preventive healthcare, their concerns about losing independence, and persons that are important to their aging journeys such as children, spouses, and friends. From an existential perspective based on lived experiences (specifically corporal, temporal, spatial, and relational), the participants\u27 subjective somatic experiences reflected some negative changes such as decline in memory and a lower level of energy. Their temporal sensibilities motivated them to do things that are desirable and meaningful. Their subjective sense of space would be the settings where they nurture their relationships and maintain their active lifestyles. Their relationships seemed to be marked by their longing to see their children often and to grow old with their spouses and friends. The findings indicate that these participants are negotiating their aging identities, autonomy, generativity, and integrity. The factors that protect these baby boomers, such as positive attitudes and sound health, are consistent with the adaptive means that relate to their aging. The importance of this investigation culminates in the cultural meanings of these findings, as they can enrich the counselor education knowledge base. Counselors must develop awareness and understanding of these meanings and integrate them into their work with baby boomer clients of Chinese descent

    Short-term photochemical and biological unreactivity of macrophyte-derived dissolved organic matter in a subtropical shallow lake

    Macrophytes have been associated with low bacterial metabolism in the littoral zones of lake Mangueira, but an explanation for this pattern is largely unknown. In this study, macrophyte-derived DOM was incubated in situ for the measurement of the effect of grazers, bacteria, and light on its degradation in three experiments. The water was separated in bulk, bacterial, and control (+ HgCl2) fractions and exposed to or hidden from sunlight for 120 h. Unchange in bacterial variables in the bulk fraction suggested a combined control of radiation and grazing on bacteria. Light treatment increased bacterial density but not biomass and biovolume, while bacterial density decreased in the dark. Significant fading of water color in the bacterial fraction only occurred after light exposure, indicating a complementary pathway of light and bacteria. DOC and the Abs250 : 365 ratio did not change with incubation, indicating no net change of DOC pool and reactivity. Due to continuous carbon loading from macrophytes and low UV irradiance, the very low rates of DOM degradation provide the mechanistic explanation for the observed impacts of macrophytes in lake’s carbon metabolism in littoral zones

    Neurofascin-155 IgM autoantibodies in patients with inflammatory neuropathies

    Objectives Recently, IgG autoantibodies against different paranodal proteins have been detected and this has led to important advances in the management of inflammatory neuropathies. In contrast, not much is known on IgM autoantibodies against paranodal proteins. Methods In the present study, we screened a large cohort of patients (n=140) with inflammatory neuropathies for IgM autoantibodies against neurofascin-155, neurofascin-186 or contactin-1. Results IgM autoantibodies against neurofascin-155 were detected by ELISA in five patients, four with inflammatory demyelinating polyradiculoneuropathy (CIDP) and one with Guillain-Barre syndrome (GBS), and were confirmed by ELISA-based preabsorption experiments and Western blot. Titres ranged from 1:100 to 1:400. We did not detect IgM anti-neurofascin-186 or anti-contactin-1 antibodies in this cohort. All patients presented with distally accentuated tetraparesis and hypesthesia. Remarkably, tremor was present in three of the patients with CIDP and occurred in the patients with GBS after the acute phase of disease. Nerve conduction studies revealed prolonged distal motor latencies and F wave latencies. Nerve biopsies showed signs of secondary axonal damage in three of the patients, demyelinating features in one patient. Teased fibre preparations did not demonstrate paranodal damage. Conclusion In summary, IgM neurofascin-155 autoantibodies may be worth testing in patients with inflammatory neuropathies. Their pathogenic role needs to be determined in future experiments

    Abstract: In this investigation, late adolescents' (N = 629) ego identity status (e.g., identity achievement, identity diffusion, identity moratorium, and identity foreclosure), cognitive processing style, and self-reported use of relational aggression and social aggression were measured in order to assess potential relationships among these constructs. Four separate models were used to test these hypotheses, and the results showed support for some but not all the four hypotheses. In this sample, it appears that individuals with high levels of cognitive sophistication who lack social maturity by which to resolve relationship problems were more likely to use social aggression than those with lower levels of cognitive processing skills or with higher levels of emotional maturity

    PET imaging to non-invasively study immune activation leading to antitumor responses with a 4-1BB agonistic antibody

    BACKGROUND: Molecular imaging with positron emission tomography (PET) may allow the non-invasive study of the pharmacodynamic effects of agonistic monoclonal antibodies (mAb) to 4-1BB (CD137). 4-1BB is a member of the tumor necrosis factor family expressed on activated T cells and other immune cells, and activating 4-1BB antibodies are being tested for the treatment of patients with advanced cancers. METHODS: We studied the antitumor activity of 4-1BB mAb therapy using [(18) F]-labeled fluoro-2-deoxy-2-D-glucose ([(18) F]FDG) microPET scanning in a mouse model of colon cancer. Results of microPET imaging were correlated with morphological changes in tumors, draining lymph nodes as well as cell subset uptake of the metabolic PET tracer in vitro. RESULTS: The administration of 4-1BB mAb to Balb/c mice induced reproducible CT26 tumor regressions and improved survival; complete tumor shrinkage was achieved in the majority of mice. There was markedly increased [(18) F]FDG signal at the tumor site and draining lymph nodes. In a metabolic probe in vitro uptake assay, there was an 8-fold increase in uptake of [(3)H]DDG in leukocytes extracted from tumors and draining lymph nodes of mice treated with 4-1BB mAb compared to untreated mice, supporting the in vivo PET data. CONCLUSION: Increased uptake of [(18) F]FDG by PET scans visualizes 4-1BB agonistic antibody-induced antitumor immune responses and can be used as a pharmacodynamic readout to guide the development of this class of antibodies in the clinic

    Re-evaluation of histological diagnoses of malignant mesothelioma by immunohistochemistry

    <p>Abstract</p> <p>Background</p> <p>In order to provide reliable tissue material for malignant mesothelioma (MM) studies, we re-evaluated biopsies and autopsy material from 61 patients with a diagnosis of MM from the period of 1980-2002.</p> <p>Methods</p> <p>Basic positive (Calretinin, EMA, Podoplanin, Mesothelin) and negative (CEA, Ber-Ep4) immunohistochemical (IHC) marker reactions were determined. If needed, more markers were used. Histological diagnoses were made by three pathologists. Survival data were calculated.</p> <p>Results</p> <p>49 cases (80%) were considered being MM by a high degree of likelihood, five more cases possible MM. Of the remaining seven cases, three were diagnosed as adenocarcinoma, three as pleomorphic lung carcinoma, in one peritoneal case a clear entity diagnosis could not be given. One of the possible MM cases and two of the lung carcinoma cases had this already as primary diagnoses, but were registered as MM.</p> <p>With a sensitivity of 100%, Calretinin and CEA were the most reliable single markers. The amount of MM cells with positive immunoreactivity (IR) for Podoplanin and Mesothelin showed most reliable inverse relation to the degree of atypia.</p> <p>In the confirmed MM cases, there had been applied either no IHC or between one and 18 markers.</p> <p>The cases not confirmed by us had either lacked IHC (n = 1), non-specific markers were used (n = 4), IR was different (n = 1), or specific markers had not shown positive IR in the right part of the tumour cells (n = 3).</p> <p>46 of the 49 confirmed and three of the not confirmed cases had been diagnosed by us as most likely MM before IHC was carried out.</p> <p>Conclusions</p> <p>In order to use archival tissue material with an earlier MM diagnosis for studies, histopathological re-evaluation is important. In possible sarcomatous MM cases without any positive IR for positive MM markers, radiology and clinical picture are essential parts of diagnostics. IHC based on a panel of two positive and two negative MM markers has to be adapted to the differential diagnostic needs in each single case. New diagnostic tools and techniques are desirable for cases where IHC and other established methods cannot provide a clear entity diagnosis, and in order to improve MM treatment.</p
