First Results from the Arcminute Cosmology Bolometer Array Receiver
We review the first science results from the Arcminute Cosmology Bolometer
Array Receiver (ACBAR); a multifrequency millimeter-wave receiver optimized for
observations of the Cosmic Microwave Background (CMB) and the
Sunyaev-Zel'dovich (SZ) effect in clusters of galaxies. ACBAR was installed on
the 2 m Viper telescope at the South Pole in January 2001 and the results
presented here incorporate data through July 2002. We present the power
spectrum of the CMB at 150 GHz over the range \ell = 150 - 3000 measured by
ACBAR as well as estimates for the values of the cosmological parameters within
the context of Lambda-CDM models. We find that the inclusion of Omega_Lambda
greatly improves the fit to the power spectrum. We also observe a slight excess
of small-scale anisotropy at 150 GHz; if interpreted as power from the SZ
effect of unresolved clusters, the measured signal is consistent with CBI and
BIMA within the context of the SZ power spectrum models tested.Comment: To be published in the proceedings of "The Cosmic Microwave

Risk-reducing hysterectomy and bilateral salpingo-oophorectomy in female heterozygotes of pathogenic mismatch repair variants: a Prospective Lynch Syndrome Database report
Purpose To determine impact of risk-reducing hysterectomy and bilateral salpingo-oophorectomy (BSO) on gynecological cancer incidence and death in heterozygotes of pathogenic MMR (path_MMR) variants. Methods The Prospective Lynch Syndrome Database was used to investigate the effects of gynecological risk-reducing surgery (RRS) at different ages. Results Risk-reducing hysterectomy at 25 years of age prevents endometrial cancer before 50 years in 15%, 18%, 13%, and 0% of path_MLH1, path_MSH2, path_MSH6, and path_PMS2 heterozygotes and death in 2%, 2%, 1%, and 0%, respectively. Risk-reducing BSO at 25 years of age prevents ovarian cancer before 50 years in 6%, 11%, 2%, and 0% and death in 1%, 2%, 0%, and 0%, respectively. Risk-reducing hysterectomy at 40 years prevents endometrial cancer by 50 years in 13%, 16%, 11%, and 0% and death in 1%, 2%, 1%, and 0%, respectively. BSO at 40 years prevents ovarian cancer before 50 years in 4%, 8%, 0%, and 0%, and death in 1%, 1%, 0%, and 0%, respectively. Conclusion Little benefit is gained by performing RRS before 40 years of age and premenopausal BSO in path_MSH6 and path_PMS2 heterozygotes has no measurable benefit for mortality. Combining Asian and European genome-wide association studies of colorectal cancer improves risk prediction across racial and ethnic populations
Polygenic risk scores (PRS) have great potential to guide precision colorectal cancer (CRC) prevention by identifying those at higher risk to undertake targeted screening. However, current PRS using European ancestry data have sub-optimal performance in non-European ancestry populations, limiting their utility among these populations. Towards addressing this deficiency, we expand PRS development for CRC by incorporating Asian ancestry data (21,731 cases; 47,444 controls) into European ancestry training datasets (78,473 cases; 107,143 controls). The AUC estimates (95% CI) of PRS are 0.63(0.62-0.64), 0.59(0.57-0.61), 0.62(0.60-0.63), and 0.65(0.63-0.66) in independent datasets including 1681-3651 cases and 8696-115,105 controls of Asian, Black/African American, Latinx/Hispanic, and non-Hispanic White, respectively. They are significantly better than the European-centric PRS in all four major US racial and ethnic groups (p-values < 0.05). Further inclusion of non-European ancestry populations, especially Black/African American and Latinx/Hispanic, is needed to improve the risk prediction and enhance equity in applying PRS in clinical practice
Mum or bub? Which influences breastfeeding loyalty
The need for social marketing research in the area of breastfeeding is highlighted by the failure of campaigns to increase breastfeeding rates over the past two decades in developed countries. This is despite evidence of the health benefits of longer breastfeeding duration to both baby and mother, and the high levels of expenditure on these campaigns. Whilst past campaign approaches typically focus on baby-oriented factors, breastfeeding is a complex behaviour that for many women involves barriers that influence their commitment to continued breastfeeding. Using social marketing, this research investigates the role of mother-centred factors on loyalty to breastfeeding. A sample of 405 Australian women completed an online survey. The data were analysed using structural equation modelling, which revealed that mother-oriented, rather than baby-oriented, factors influence attitudinal and behavioural loyalty to breastfeeding
Kinetics and intracellular pathways required for major histocompatibility complex II–peptide loading and surface expression of a fluorescent hapten-protein conjugate in murine macrophage
A fluorescent antigen, FITC10BSA, that is sensitive to several of the biochemical processes involved in antigen processing was constructed. In combination with both flow cytometry and subcellular fractionation, the unique probe provided new details regarding the kinetics and intracellular pathways involved in antigen processing in murine macrophage. These studies suggested that macrophage utilized multiple vesicles as opposed to a few specific organelles for major histocompatibility complex (MHC) type II–peptide loading and transport. Although newly formed MHC II–peptide complexes were detected in cathepsin D-positive, lysosomal associated membrane glycoprotein (LAMP-1)-positive lysosomes, MHC II–peptide loading also occurred in transferrin receptor-positive endosomes. Interestingly, MHC II-fluoresceinated complexes were only observed in transferrin receptor-positive organelles as opposed to MHC II-unlabelled peptide complexes which were detected in traditional early lysosomal compartments. More importantly, MHC II–peptide complexes were monitored in light transferrin receptor-positive fractions following their initial appearance in dense endosomal/lysosomal fractions. Control experiments suggested that these complexes represented intermediates in the process of migrating to the cell surface through a retrograde pathway within the macrophage
Biomechanical Comparison of Same Size Transfacet Screws Versus Pedicle Screws Across the L5-S1 Native Disc
Objective: There is resurgence in facet screw use; however, there is a paucity of data comparing the stability of transfacet pedicle screws (TFPS) to standard pedicle screw-rod constructs at L5-S1. The authors aim to compare the stabilizing potential of TFPS compared to pedicle screws of the same length and diameter at L5-S1 with an intact native disc. Methods: Flexibility of human cadaveric lumbar spine segments was biomechanically tested in vitro to provide a comparison of two types of posterior stabilization across L5-S1. Fourteen cadaveric spine specimens (T12-S1) were tested in intact condition, then after, surgical procedures performed at L5-S1 using 5.0 Ă— 40 mm transfacet pedicle screws (Group #1) and same sized pedicle screw-rod construct (Group #2). Specimens were studied using standard non-destructive pure moment flexibility tests. Results: Transfacet pedicle screws (TFPS) allowed less motion in all loading modes than bilateral pedicle screw-rod (BPSR) construct, with a significantly smaller range of motion allowed during extension and axial rotation (p \u3c 0.05). Transfacet pedicle screws allow less lateral bending than BPSR compared to the intact specimen, although the magnitude of the difference was less than during flexion or extension. Conclusion: Bilateral transfacet screws provided better immediate postoperative stability in vitro intact posterior element facets without osteophytes than equivalent-sized unilateral or a bilateral standard pedicle screw at L5-S1. These are limited to a biomechanical application as larger sized pedicle screws are used in the clinical setting. Further biomechanical and clinically relevant studies are warranted to verify these points
