    Combining GWAS and Population Genomic Analyses to Characterize Coevolution in a Legume-rhizobia Symbiosis

    The mutualism between legumes and rhizobia is clearly the product of past coevolution. However, the nature of ongoing evolution between these partners is less clear. To characterize the nature of recent coevolution between legumes and rhizobia, we used population genomic analysis to characterize selection on functionally annotated symbiosis genes as well as on symbiosis gene candidates identified through a two-species association analysis. For the association analysis, we inoculated each of 202 accessions of the legume host Medicago truncatula with a community of 88 Sinorhizobia (Ensifer) meliloti strains. Multistrain inoculation, which better reflects the ecological reality of rhizobial selection in nature than single-strain inoculation, allows strains to compete for nodulation opportunities and host resources and for hosts to preferentially form nodules and provide resources to some strains. We found extensive host by symbiont, that is, genotype-by-genotype, effects on rhizobial fitness and some annotated rhizobial genes bear signatures of recent positive selection. However, neither genes responsible for this variation nor annotated host symbiosis genes are enriched for signatures of either positive or balancing selection. This result suggests that stabilizing selection dominates selection acting on symbiotic traits and that variation in these traits is under mutation-selection balance. Consistent with the lack of positive selection acting on host genes, we found that among-host variation in growth was similar whether plants were grown with rhizobia or N-fertilizer, suggesting that the symbiosis may not be a major driver of variation in plant growth in multistrain contexts

    Optimisation of estrogen receptor subtype-selectivity of a 4-Aryl-4H-chromene scaffold previously identified by virtual screening

    4-Aryl-4H-Chromene derivatives have been previously shown to exhibit anti-proliferative, apoptotic and anti-angiogenic activity in a variety of tumor models in vitro and in vivo generally via activation of caspases through inhibition of tubulin polymerisation. We have previously identified by Virtual Screening (VS) a 4-aryl-4H-chromene scaffold, of which two examples were shown to bind Estrogen Receptor α and ÎČ with low nanomolar affinity and \u3c20-fold selectivity for α over ÎČ and low micromolar anti-proliferative activity in the MCF-7 cell line. Thus, using the 4-aryl-4H-chromene scaffold as a starting point, a series of compounds with a range of basic arylethers at C-4 and modifications at the C3-ester substituent of the benzopyran ring were synthesised, producing some potent ER antagonists in the MCF-7 cell line which were highly selective for ERα (compound 35; 350-fold selectivity) or ERÎČ (compound 42; 170-fold selectivity)

    LIFE in a ZOO: Henri Lefebvre and the (social) production of (abstract) space in Liverpool

    Building on recent critical contributions towards conceptualising neighbourhood change as socially produced and politically ‘performed’, this paper takes a closer look at the work of Henri Lefebvre to understand the production of urban space as a deeply political process. A common critical characterisation of neighbourhood change—occurring through a grand Lefebvrean struggle between ‘abstract space-makers’ and ‘social space-makers’—is critically examined through an in-depth historical case study of the Granby neighbourhood in Liverpool. Here, these forces are embodied respectively in technocratic state-led comprehensive redevelopment, notably Housing Market Renewal and its LIFE and ZOO zoning models; and in alternative community-led rehabilitation projects such as the Turner Prize-winning Granby Four Streets Community Land Trust. By tracing the surprisingly intimate interactions and multiple contradictions between these apparently opposing spatial projects, the production of neighbourhood is shown to be a complex, often violent political process, whose historical trajectories require disentangling in order to understand how we might construct better urban futures

    In this issue of the Policy Review Section , Stephen Hall and Brendan Nevin of the Centre for Urban and Regional Studies, University of Birmingham, review the experiences of the first three rounds of the Single Regeneration Budget and against that background consider the next steps in the development of regeneration policy. In the second article, Peter Lee also of the Centre for Urban and Regional Studies, examines the policy implications of the recent changes in the methods for identifying deprived areas following the introduction of the Index for Local Deprivation. In the third article, Peter Newman of the School of Built Environment, University of Westminster, and Gilles Verpraet of the Centre d'Analyses et d'Interventions Sociologiques, Paris, set out the findings of a comparative European research network which examined the nature and experience of public-private partnerships in urban governance.
