    Romantic and sexual behavior in young adolescents : repeated surveys in a population-based cohort

    Adverse outcomes of teenage sexual activity are common in the United Kingdom. We used a computer-assisted interview to ask young adolescents aged 11-12 years (N = 6856) and 12 -13 years (N = 6801) who were part of the Avon Longitudinal Study of Parents and Children about romantic and intimate behaviors. 24% of 11-12 year olds and 41% of 12-13 year olds reported holding hands and 17% and 32% respectively reported having been kissed on the mouth. A minority of 12-13 year olds reported sexual behavior including intercourse. The majority of participants who reported intimate sexual behavior did not regret it. Boys reported all activities more than girls and were less likely to express regret. Our findings suggest that the timing and content of sex education should take into account the frequency with which young people are experimenting with sexual behaviors

    A Birth Cohort Analysis to Study Dog Walking in Adolescence Shows No Relationship with Objectively Measured Physical Activity.

    Physical inactivity during childhood and adolescence is a serious health concern. There are few studies of the activity undertaken by adolescents when walking with the family dog, and the effect of this on objectively measured physical activity levels. Objective measures of physical activity using accelerometers were recorded at age 11-12, 13-14, and 15-16 years in the Avon Longitudinal Study of Parents and Children (ALSPAC) (ALSPAC, UK) birth cohort during the 2000s. Family pet ownership was collected retrospectively using a questionnaire at age 18 years, for the ages 7, 11, 13, and 15 years. In addition, approximate frequency per week of walks undertaken with dogs were also reported. Multilevel, multivariable modeling was used to investigate the relationship between dog ownership and dog walking status, and physical activity outcomes. There were a total of 4,373 complete data observations for use in 2,055 children. Reported participation in dog walking tended to increase during adolescence, as did dog ownership. The majority of who own dogs reported walking them either 2-6 times/week (range 39-46%) or never (range 27-37%). A small minority (7-8%) reported walking their dog every day. Most reported never walking any other dog either (94-87%). We found no evidence for an association between dog ownership or reported dog walking, and objectively measured physical activity (counts per minute, P = 0.3, or minutes of moderate-to-vigorous physical activity, P = 0.7) during adolescence. This study provides no evidence to support a relationship between adolescent dog ownership and physical activity, and demonstrates the importance of using objective activity measures and considering dog walking rather than just dog ownership

    An overview of systematic reviews of complementary and alternative therapies for fibromyalgia using both AMSTAR and ROBIS as quality assessment tools

    Abstract Background Fibromyalgia (FM) is a chronic, debilitating pain disorder. Dissatisfaction with conventional medicine can lead people with FM to turn to complementary and alternative medicine (CAM). Two previous overviews of systematic reviews of CAM for FM have been published, but they did not assessed for risk of bias in the review process. Methods Five databases Medline, Embase, AMED (via OVID), Web of Science and Central were searched from their inception to December 2015. Reference lists were hand-searched. We had two aims: the first was to provide an up-to-date and rigorously conducted synthesis of systematic reviews of CAM literature on FM; the second was to evaluate the quality of the available systematic review evidence using two different tools: AMSTAR (Shea et al. BMC Med Res Methodol 15; 7:10, 2007) and a more recently developed tool ROBIS (Whiting et al. J Clin Epidemiol 69:225-34, 2016) specifically designed to assess risk of bias in systematic reviews. Any review that assessed one of eight CAM therapies for participants diagnosed with FM was considered. The individual studies had to be randomised controlled trials where the intervention was compared to placebo, treatment as usual or waitlist controls to be included. The primary outcome measure was pain, and the secondary outcome measure was adverse events. Results We identified 15 reviews that met inclusion criteria. There was low-quality evidence that acupuncture improves pain compared to no treatment or standard treatment, but good evidence that it is no better than sham acupuncture. The evidence for homoeopathy, spinal manipulation and herbal medicine was limited. Conclusions Overall, five reviews scored 6 or above using the AMSTAR scale and the inter-rater agreement was good (83.6%), whereas seven reviews achieved a low risk of bias rating using ROBIS and the inter-rater agreement was fair (60.0%). No firm conclusions were drawn for efficacy of either spinal manipulation or homoeopathy for FM. There is limited evidence for topical Capsicum, but further research is required. There is some evidence to support the effectiveness of acupuncture for FM, but further high-quality trials are needed to investigate its benefits, harms and mechanisms of action, compared with no or standard treatment.     Estimate of the Spontaneous Mutation Rate in Chlamydomonas reinhardtii

    The nature of spontaneous mutations, including their rate, distribution across the genome, and fitness consequences, is of central importance to biology. However, the low rate of mutation has made it difficult to study spontaneous mutagenesis, and few studies have directly addressed these questions. Here, we present a direct estimate of the mutation rate and a description of the properties of new spontaneous mutations in the unicellular green alga Chlamydomonas reinhardtii. We conducted a mutation accumulation experiment for ∼350 generations followed by whole-genome resequencing of two replicate lines. Our analysis identified a total of 14 mutations, including 5 short indels and 9 single base mutations, and no evidence of larger structural mutations. From this, we estimate a total mutation rate of 3.23 × 10(−10)/site/generation (95% C.I. 1.82 × 10(−10) to 5.23 × 10(−10)) and a single base mutation rate of 2.08 × 10(−10)/site/generation (95% C.I., 1.09 × 10(−10) to 3.74 × 10(−10)). We observed no mutations from A/T → G/C, suggesting a strong mutational bias toward A/T, although paradoxically, the GC content of the C. reinhardtii genome is very high. Our estimate is only the second direct estimate of the mutation rate from plants and among the lowest spontaneous base-substitution rates known in eukaryotes

    Interpretable Survival Analysis for Heart Failure Risk Prediction

    Survival analysis, or time-to-event analysis, is an important and widespread problem in healthcare research. Medical research has traditionally relied on Cox models for survival analysis, due to their simplicity and interpretability. Cox models assume a log-linear hazard function as well as proportional hazards over time, and can perform poorly when these assumptions fail. Newer survival models based on machine learning avoid these assumptions and offer improved accuracy, yet sometimes at the expense of model interpretability, which is vital for clinical use. We propose a novel survival analysis pipeline that is both interpretable and competitive with state-of-the-art survival models. Specifically, we use an improved version of survival stacking to transform a survival analysis problem to a classification problem, ControlBurn to perform feature selection, and Explainable Boosting Machines to generate interpretable predictions. To evaluate our pipeline, we predict risk of heart failure using a large-scale EHR database. Our pipeline achieves state-of-the-art performance and provides interesting and novel insights about risk factors for heart failure
