μChIP—a rapid micro chromatin immunoprecipitation assay for small cell samples and biopsies
Chromatin immunoprecipitation (ChIP) is a powerful technique for studying protein–DNA interactions. Drawbacks of current ChIP assays however are a requirement for large cell numbers, which limits applicability of ChIP to rare cell samples, and/or lengthy procedures with limited applications. There are to date no protocols for fast and parallel ChIPs of post-translationally modified histones from small cell numbers or biopsies, and importantly, no protocol allowing for investigations of transcription factor binding in small cell numbers. We report here the development of a micro (μ) ChIP assay suitable for up to nine parallel quantitative ChIPs of modified histones or RNA polymerase II from a single batch of 1000 cells. μChIP can also be downscaled to monitor the association of one protein with multiple genomic sites in as few as 100 cells. μChIP is applicable to small fresh tissue biopsies, and a cross-link-while-thawing procedure makes the assay suitable for frozen biopsies. Using μChIP, we characterize transcriptionally permissive and repressive histone H3 modifications on developmentally regulated promoters in human embryonal carcinoma cells and in osteosarcoma biopsies. μChIP creates possibilities for multiple parallel and rapid transcription factor binding and epigenetic analyses of rare cell and tissue samples

Survey of Period Variations of Superhumps in SU UMa-Type Dwarf Novae. II: The Second Year (2009-2010)
As an extension of the project in Kato et al. (2009, arXiv:0905.1757), we
collected times of superhump maxima for 61 SU UMa-type dwarf novae mainly
observed during the 2009-2010 season. The newly obtained data confirmed the
basic findings reported in Kato et al. (2009): the presence of stages A-C, as
well as the predominance of positive period derivatives during stage B in
systems with superhump periods shorter than 0.07 d. There was a systematic
difference in period derivatives for systems with superhump periods longer than
0.075 d between this study and Kato et al. (2009). We suggest that this
difference is possibly caused by the relative lack of frequently outbursting SU
UMa-type dwarf novae in this period regime in the present study. We recorded a
strong beat phenomenon during the 2009 superoutburst of IY UMa. The close
correlation between the beat period and superhump period suggests that the
changing angular velocity of the apsidal motion of the elliptical disk is
responsible for the variation of superhump periods. We also described three new
WZ Sge-type objects with established early superhumps and one with likely early
superhumps. We also suggest that two systems, VX For and EL UMa, are WZ
Sge-type dwarf novae with multiple rebrightenings. The O-C variation in OT
J213806.6+261957 suggests that the frequent absence of rebrightenings in very
short-Porb objects can be a result of sustained superoutburst plateau at the
epoch when usual SU UMa-type dwarf novae return to quiescence preceding a
rebrightening. We also present a formulation for a variety of Bayesian

Young at risk-people in Maputo City, Mozambique, present a high willingness to participate in HIV trials: Results from an HIV vaccine preparedness cohort study
Introduction: Vaccine efficacy testing requires engagement of willing volunteers with high disease incidence. We evaluated factors associated with willingness to participate in potential future HIV vaccine trials in Maputo, Mozambique.
Methods: Adults aged 18–35 years without HIV and who reported at least two sexual partners in the 3 months prior to screening were enrolled into a 24-month observational study. They were asked at screening and exit if they would be willing to participate in a theoretical HIV vaccine study. Bivariate and multivariate logistic regression analyses were done between willingness to participate, demographic, sexual behavior, and motivational factors for screening visit data. Logistic regression with generalized estimating equations (GEE) was used to estimate odds ratios (ORs) and 95% confidence intervals (CIs) for factors potentially associated with willingness to participate for data from both visits.
Results: A total of 577 participants without HIV were eligible, including 275 (48%) women. The mean age was 22.2 (SD ± 3.9) years. At screening 529 (92%) expressed willingness to participate and the proportion remained stable at 378 (88%) of the 430 participants retained through the exit visit (p = 0.209). Helping the country (n = 556) and fear of needles (n = 26) were the top motive and barrier for willingness to participate, respectively. Results from the GEE binary logistic regression (screening visit and exit visit) showed that wanting to learn how to avoid risk behaviors (aOR 3.33, 95% CI: 1.61–6.86) and feeling protected against HIV infection (aOR 2.24, 95% CI: 1.07–4.7) were associated with willingness to participate in HIV vaccine studies.
Conclusion: The majority of our study population in Mozambique expressed willingness to participate in a theoretical HIV vaccine trial. Participation in a HIV vaccine trial was seen as a way to contribute to the fight against HIV but was associated with some unrealistic expectations such as protection against HIV. This reinforces the need for continuous mobilization and awareness of potential participants to HIV vaccine trial
Survey of Period Variations of Superhumps in SU UMa-Type Dwarf Novae
We systematically surveyed period variations of superhumps in SU UMa-type
dwarf novae based on newly obtained data and past publications. In many
systems, the evolution of superhump period are found to be composed of three
distinct stages: early evolutionary stage with a longer superhump period,
middle stage with systematically varying periods, final stage with a shorter,
stable superhump period. During the middle stage, many systems with superhump
periods less than 0.08 d show positive period derivatives. Contrary to the
earlier claim, we found no clear evidence for variation of period derivatives
between superoutburst of the same object. We present an interpretation that the
lengthening of the superhump period is a result of outward propagation of the
eccentricity wave and is limited by the radius near the tidal truncation. We
interpret that late stage superhumps are rejuvenized excitation of 3:1
resonance when the superhumps in the outer disk is effectively quenched. Many
of WZ Sge-type dwarf novae showed long-enduring superhumps during the
post-superoutburst stage having periods longer than those during the main
superoutburst. The period derivatives in WZ Sge-type dwarf novae are found to
be strongly correlated with the fractional superhump excess, or consequently,
mass ratio. WZ Sge-type dwarf novae with a long-lasting rebrightening or with
multiple rebrightenings tend to have smaller period derivatives and are
excellent candidate for the systems around or after the period minimum of
