    The role of hRev7, the accessory subunit of hPolĪ¶, in translesion synthesis past DNA damage induced by benzo[a]pyrene diol epoxide (BPDE)

    <p>Abstract</p> <p>Background</p> <p>DNA polymerase zeta (PolĪ¶) is a specialized DNA polymerase that, unlike classical replicative polymerases, is capable of replicating past DNA lesions, i.e. of performing translesion synthesis (TLS). The catalytic subunit of hPolĪ¶, hRev3, has been shown to play a critical role in DNA damage-induced mutagenesis in human cells, but less is known about the role of hRev7, the accessory subunit of hPolĪ¶, in such mutagenesis. To address this question, we recently generated human fibroblasts with very significantly reduced levels of hRev7 protein and demonstrated that hRev7 is required to protect cells from ultraviolet<sub>(254 nm) </sub>(UV) radiation-induced cytotoxicity and mutagenesis (McNally et al., DNA Repair 7 (2008) 597-604). The goal of the present study was to determine whether hRev7 is similarly involved in the tolerance of DNA damage induced by benzo[a]pyrene diol epoxide (BPDE), the reactive form of the widespread environmental carcinogen benzo[a]pyrene.</p> <p>Methods</p> <p>To determine whether hRev7 also plays a role in protecting human cells from the cytotoxicity and mutagenesis induced by benzo[a]pyrene diol epoxide (BPDE), cell strains with reduced hRev7 were compared to their parental strain and a vector control strain for the effect of BPDE on cell survival, induction of mutations, and the ability to progress through the cell cycle.</p> <p>Results</p> <p>The results show that cell strains with reduced hRev7 are more sensitive to the cytotoxic effect of BPDE than the control strains, and progress through S-phase at a slower rate than the control cells following BPDE treatment, indicating that hRev7, and likely hPolĪ¶, is required for efficient bypass of BPDE-induced DNA lesions. However, neither the frequency nor kinds of mutations induced by BPDE in cells with reduced hRev7 differ significantly from those induced in the control strains, suggesting that hPolĪ¶ is not essential for inserting nucleotides opposite BPDE-induced DNA damage.</p> <p>Conclusions</p> <p>Taken together, our results which show that hRev7 is required for TLS past BPDE-induced DNA lesions but that it is not essential for inserting nucleotides opposite such lesions suggest a role for hPolĪ¶ in the extension step of translesion synthesis.</p

    A Deep Search For Faint Galaxies Associated With Very Low-redshift C IV Absorbers: III. The Mass- and Environment-dependent Circumgalactic Medium

    Using Hubble Space Telescope Cosmic Origins Spectrograph observations of 89 QSO sightlines through the Sloan Digital Sky Survey footprint, we study the relationships between C IV absorption systems and the properties of nearby galaxies as well as large-scale environment. To maintain sensitivity to very faint galaxies, we restrict our sample to 0.0015 < z < 0.015, which defines a complete galaxy survey to L > 0.01 L* or stellar mass log M_* > 8 Msun. We report two principal findings. First, for galaxies with impact parameter rho < 1 rvir, C IV detection strongly depends on the luminosity/stellar mass of the nearby galaxy. C IV is preferentially associated with galaxies with log M_* > 9.5 Msun; lower mass galaxies rarely exhibit significant C IV absorption (covering fraction f = 9 +12-6% for 11 galaxies with log M_* < 9.5 Msun). Second, C IV detection within the log M_* > 9.5 Msun population depends on environment. Using a fixed-aperture environmental density metric for galaxies with rho < 160 kpc at z < 0.055, we find that 57+/-12% (8/14) of galaxies in low-density regions (regions with fewer than seven L > 0.15 L* galaxies within 1.5 Mpc) have affiliated C IV absorption; however, none (0/7) of the galaxies in denser regions show C IV. Similarly, the C IV detection rate is lower for galaxies residing in groups with dark-matter halo masses of log Mhalo > 12.5 Msun. In contrast to C IV, H I is pervasive in the CGM without regard to mass or environment. These results indicate that C IV absorbers with log N(C IV) > 13.5 cm^-2 trace the halos of log M_* > 9.5 Msun galaxies but also reflect larger scale environmental conditions.Comment: 26 pages, 13 figures.     Teacher Beliefs about Motivating and Teaching Students to Carry out Engineering Design Challenges: Some Initial Data

    The present study examines middle school teachersā€™ beliefs about seven learning outcomes related to a project that involves developing and examining the effects of a set of engineering design modules constructed for use by middle school math and science teachers. Overall, the teachers involved in the intervention appear to believe they have the instructional skills, professional development, and resources to carry out the modules. Teachers from all of the schools (both intervention and comparison schools) for the most part valued the outcomes as important. Results of the study indicate that, although teachers believe they value and can obtain most of these outcomes; beliefs vary by school and other factors. One area where teachers do not seem strongly efficacious in some schools is that of fostering intrinsic motivation in their students. Teachers in one of the schools where the modules were implemented did not feel their students were capable of becoming intrinsically motivated. The implications for implementing engineering education in middle school of these beliefs and other attitudes are discussed

    Vascular Health in American Football Players: Cardiovascular Risk Increased in Division III Players

    Studies report that football players have high blood pressure (BP) and increased cardiovascular risk. There are over 70,000 NCAA football players and 450 Division III schools sponsor football programs, yet limited research exists on vascular health of athletes. This study aimed to compare vascular and cardiovascular health measures between football players and nonathlete controls. Twenty-three athletes and 19 nonathletes participated. Vascular health measures included flow-mediated dilation (FMD) and carotid artery intima-media thickness (IMT). Cardiovascular measures included clinic and 24 hr BP levels, body composition, VO2 max, and fasting glucose/cholesterol levels. Compared to controls, football players had a worse vascular and cardiovascular profile. Football players had thicker carotid artery IMT (0.49 Ā± 0.06 mm versus 0.46 Ā± 0.07 mm) and larger brachial artery diameter during FMD (4.3 Ā± 0.5 mm versus 3.7 Ā± 0.6 mm), but no difference in percent FMD. Systolic BP was significantly higher in football players at all measurements: resting (128.2 Ā± 6.4 mmHg versus 122.4 Ā± 6.8 mmHg), submaximal exercise (150.4 Ā± 18.8 mmHg versus 137.3 Ā± 9.5 mmHg), maximal exercise (211.3 Ā± 25.9 mmHg versus 191.4 Ā± 19.2 mmHg), and 24-hour BP (124.9 Ā± 6.3 mmHg versus 109.8 Ā± 3.7 mmHg). Football players also had higher fasting glucose (91.6 Ā± 6.5 mg/dL versus 86.6 Ā± 5.8 mg/dL), lower HDL (36.5Ā±11.2 mg/dL versus 47.1Ā±14.8 mg/dL), and higher body fat percentage (29.2Ā±7.9% versus 23.2Ā±7.0%).     The COS-Dwarfs Survey: The Carbon Reservoir Around sub-L* Galaxies

    We report new observations of circumgalactic gas from the COS-Dwarfs survey, a systematic investigation of the gaseous halos around 43 low-mass z ā‰¤\leq 0.1 galaxies using background QSOs observed with the Cosmic Origins Spectrograph. From the projected 1D and 2D distribution of C IV absorption, we find that C IV absorption is detected out to ~ 0.5 Rvir_{vir} of the host galaxies. The C IV absorption strength falls off radially as a power law and beyond 0.5 Rvir_{vir}, no C IV absorption is detected above our sensitivity limit of ~ 50-100 mAĖš\AA. We find a tentative correlation between detected C IV absorption strength and star formation, paralleling the strong correlation seen in highly ionized oxygen for L~L* galaxies by the COS-Halos survey. The data imply a large carbon reservoir in the CGM of these galaxies, corresponding to a minimum carbon mass of ā‰³\gtrsim 1.2Ɨ106\times 10^6 MāŠ™M_\odot out to ~ 110 kpc. This mass is comparable to the carbon mass in the ISM and more than the carbon mass currently in stars of these galaxies. The C IV absorption seen around these sub-L* galaxies can account for almost two-thirds of all WrW_r> 100 mAĖš\AA C IV absorption detected at low z. Comparing the C IV covering fraction with hydrodynamical simulations, we find that an energy-driven wind model is consistent with the observations whereas a wind model of constant velocity fails to reproduce the CGM or the galaxy properties.Comment: 18 Pages, 11 Figures, ApJ 796 13

    Unraveling the Complexities of DNA-Dependent Protein Kinase Autophosphorylation

    DNA-dependent protein kinase (DNA-PK) orchestrates DNA repair by regulating access to breaks through autophosphorylations within two clusters of sites (ABCDE and PQR). Blocking ABCDE phosphorylation (by alanine mutation) imparts a dominant negative effect, rendering cells hypersensitive to agents that cause DNA double-strand breaks. Here, a mutational approach is used to address the mechanistic basis of this dominant negative effect. Blocking ABCDE phosphorylation hypersensitizes cells to most types of DNA damage (base damage, cross-links, breaks, and damage induced by replication stress), suggesting that DNA-PK binds DNA ends that result from many DNA lesions and that blocking ABCDE phosphorylation sequesters these DNA ends from other repair pathways. This dominant negative effect requires DNA-PK's catalytic activity, as well as phosphorylation of multiple (non-ABCDE) DNA-PK catalytic subunit (DNA-PKcs) sites. PSIPRED analysis indicates that the ABCDE sites are located in the only contiguous extended region of this huge protein that is predicted to be disordered, suggesting a regulatory role(s) and perhaps explaining the large impact ABCDE phosphorylation has on the enzyme's function. Moreover, additional sites in this disordered region contribute to the ABCDE cluster. These data, coupled with recent structural data, suggest a model whereby early phosphorylations promote initiation of nonhomologous end joining (NHEJ), whereas ABCDE phosphorylations, potentially located in a ā€œhingeā€ region between the two domains, lead to regulated conformational changes that initially promote NHEJ and eventually disengage NHEJ
