    Analysis of auto-purification response of the Apies River, Gauteng, South Africa, to treated wastewater effluent

    The assimilative capacity of water bodies is an important factor in the integrated management of surface water resources. The current study examined the auto-recovery processes of the Apies River from wastewater discharged into it from a municipal wastewater treatment facility, using a series of equations, including the modified Streeter-Phelps equation. Field data obtained include dissolved oxygen (DO), temperature, stream velocity, depth, and width. Water samples were also obtained at 10 sampling stations for the determination of biochemical oxygen demand (BOD) using standard methods. It was observed that the DO and BOD level (5.59 mg/L and 8.5 mg/L respectively) of the effluent from the wastewater treatment facility indicated better water quality than the Apies River background DO level (5.42 mg/L) and BOD level (13 mg/L). Also, at 270 m downstream of the effluent discharge point, another effluent stream (Skinnerspruit) adversely impacted on the Apies River with DO and BOD levels of 6.5 mg/L and 9.0 mg/L, respectively, compared to the Apies River background values of 6.81 mg/L and 8.0 mg/L, respectively. The stream, however, recovered well from both the background and imposed pollution sources as it had a computed positive auto-recovery factor of 1.74. Furthermore, the measured DO deficit was plotted against predicted DO deficit. The plot revealed a close match between the measured and predicted DO deficit, indicating that the model could be used for predicting DO deficit along other segments of the river. To further improve on the natural auto-recovery processes of the Apies River, it was recommended that flow along the Skinnerspruit should be enhanced by clearing the observed aquatic plants growing within the channel. Also, suspected pollution activities taking place further upstream on the Apies River should be investigated and appropriately addressed

    A Model Selection Procedure for Stream Re-Aeration Coefficient Modelling

    Model selection is finding wide applications in a lot of modelling and environmental problems. However, applications of model selection to re-aeration coefficient studies are still limited. The current study explores the use of model selection in re-aeration coefficient studies by combining several suggestions from numerous authors on the interpretation of data regarding re-aeration coefficient modelling. The model selection procedure applied in this research made use of Akaike information criteria, measures of agreement such as percent bias (PBIAS), Nash-Sutcliffe Efficiency (NSE) and root mean square error (RMSE) observation Standard deviation Ratio (RSR) and gragh analysis in selecting the best performing model. An algorithm prescribing a generic model selection procedure was also provided. Out of ten candidates models used in this study, the O’Connor and Dobbins (1958) model emerged as the top performing model in its application to data collected from River Atuwara in Nigeria. The suggested process could save software and model developers lots of time and resources, which would otherwise be spent in investigating and developing new models. The procedure is also ideal in selecting a model in situations where there is no overwhelming support for any particular model by observed data

    Using GIS for spatial exploratory analysis of borehole data: a firsthand approach towards groundwater development

    Groundwater is an inimitable resource that provides water to communities especially in arid and semi-arid regions. However, the spatial variability of the resource as well as the heterogeneity and complex nature of aquifer systems that store groundwater presents difficulties for groundwater development. Thus, understanding the spatial structure of aquifer characteristics could be used as a resourceful tool and as a first point of call towards groundwater development. The study investigated aquifer characteristics particularly on transmissivity and specific capacity to determine how aquifer characteristics could be used in making predictions for new sites that need to be identified for potential groundwater development. Prediction maps were developed for transmissivity and specific capacity using ordinary kriging J-Bessel and exponential semi variograms techniques. The study revealed that 28% of the study area had high potential for groundwater development with transmissivity ranges of 34.40 – 46.52 m2/d. Equally, viable areas of 16% and 20% were identified with transmissivities of 27.99 – 34.39 m2/d and 13.07 – 19.47 m2/d respectively. Based on specific capacity, 21% of the study area was identified to have groundwater development potential with corresponding specific capacity of 13.54 – 20.7 m2/d. Similarly, 24% and 34% of the study area were also identified to be good for the exploitation of groundwater with corresponding specific capacities of 8.74 – 13.55 m2/d and 5.52 – 8.74 m2/d respectively. In terms of spatial structure, transmissivity was found to be variable compared to specific capacity. The application of Geographic Information Systems (GIS) is thus considered to be an immeasurable tool that could be used in mapping aquifer characteristics that can serve as firsthand information for groundwater development.Keywords: Spatial, Groundwater, Transmissivity, Specific capacity, Kriging, Semivariogram        

    Multi-locus sequence typing of African swine fever viruses from endemic regions of Kenya and Eastern Uganda (2011–2013) reveals rapid B602L central variable region evolution

    The central variable region (CVR) within the B602L gene of the African swine fever virus (ASFV) is highly polymorphic within the 23 ASFV genotypes defined by sequencing of the C-terminal end of the p72 locus. Sequencing the p54 gene further discriminates ASFV genotypes that are conserved at the p72 locus. Variation in the thymidine kinase locus is a novel additional tool for ASFV genotyping whose application for this purpose is described for the first time herein. We evaluated genetic variation at these four polymorphic loci in 39 ASFV isolates obtained from outbreaks in Kenya and a region of Eastern Uganda between 2011 and 2013. Analysis of the p72 and p54 loci revealed high genetic conservation among these isolates; all clustered within p72 genotype IX and were similar to isolates associated with earlier outbreaks in East Africa. The thymidine kinase gene of the Kenyan isolates in this study were distinct relative to Southern African isolates and synonymous substitutions were observed among viruses from central Kenya. Analysis of the CVR within the B602L gene revealed two previously unknown polymorphisms that were restricted to Western Kenya and Eastern Uganda. A novel variant was revealed within CVR subgroup XXIV and a novel CVR subgroup XXIVa that contains tetrameric repeat F which has previously only been associated with p72 genotype I, was also identified for the first time in East Africa. Phylogeographic analysis of isolates based on CVR polymorphisms revealed rapid evolution and dissemination of variants present within ASFV genotype IX in East Africa

    Pregnancy rates among female participants in phase 1 and phase 2A AIDS vaccine clinical trials in Kenya

    Background: Female participants in AIDS candidate vaccine clinical trials must agree to use effective contraception to be enrolled into the studies, and for a specified period after vaccination, since the candidate vaccines’ effects on the embryo or foetus are unknown.Objectives: To review data on female participants’ pregnancy rates from phase I and IIA AIDS vaccine clinical trials conducted at the Kenya AIDS Vaccine Initiative (KAVI) and to discuss the challenges of contraception among female participants.Design: Descriptive observational retrospective study.Setting: KAVI clinical trial site, Kenyatta National Hospital and University of Nairobi, Kenya.Subjects: Thirty nine female participants were enrolled into these trials. They received family planning counselling and were offered a choice of different contraceptive methods, as per the protocols. All contraception methods chosen by the participants were offered at the study site at no cost to the participant.Results: Four women conceived during the study period when pregnancies were to be avoided. All four had opted for sexual abstinence as a contraceptive method, but reported having been coerced by their partners to have unprotected sexual intercourse.Conclusion: Abstinence is clearly not a reliable contraceptive option for women in developing-country settings. Effective female-controlled contraceptives, administered at the clinical trial site, may empower female participants to better control their fertility, leading to more complete clinical trial data

    Inherited tolerance in cattle to the apicomplexan protozoan Theileria parva is associated with decreased proliferation of parasite-infected lymphocytes

    Theileria parva is the causative agent of East Coast fever and Corridor disease, which are fatal, economically important diseases of cattle in eastern, central and southern Africa. Improved methods of control of the diseases are urgently required. The parasite transforms host lymphocytes, resulting in a rapid, clonal expansion of infected cells. Resistance to the disease has long been reported in cattle from T. parva-endemic areas. We reveal here that first- and second-generation descendants of a single Bos indicus bull survived severe challenge with T. parva, (overall survival rate 57.3% compared to 8.7% for unrelated animals) in a series of five field studies. Tolerant cattle displayed a delayed and less severe parasitosis and febrile response than unrelated animals. The in vitro proliferation of cells from surviving cattle was much reduced compared to those from animals that succumbed to infection. Additionally, some pro-inflammatory cytokines such as IL1ÎČ, IL6, TNFα or TGFÎČ which are usually strongly expressed in susceptible animals and are known to regulate cell growth or motility, remain low in tolerant animals. This correlates with the reduced proliferation and less severe clinical reactions observed in tolerant cattle. The results show for the first time that the inherited tolerance to T. parva is associated with decreased proliferation of infected lymphocytes. The results are discussed in terms of whether the reduced proliferation is the result of a perturbation of the transformation mechanism induced in infected cells or is due to an innate immune response present in the tolerant cattle
