    USE OF A HOMEOPATHIC PRODUCT IN DOGS REDUCES THE NEGATIVE EFFECTS OF CANINE ATOPIC DERMATITIS

    This study aimed to determine whether a homeopathic product formulated based on Hepar sulfur, Rhus toxicodendron, Graphites, and Urtica urens would prevent atopic dermatitis in dogs. Ten male beagle dogs were used, divided into two groups with five repetitions per group. The control (CO) animals received a diet with 0.5 mL/day of placebo for 30 consecutive days in a preventive manner, and we later added ethoxyquin at 0.4 mg/day. The treated group (TRA) received a ration with 0.5 mL/day of homeopathic for 30 consecutive days as a preventive, and we later added ethoxyquin at 0.4 mg/day. Blood samples were collected on days 1, 30, and 40 to analyze antioxidant and oxidant status. In an ethogram, the presence of a crusted lesion, alopecic lesion, or hair loss were evaluated at three timepoints: pre-challenge, 5, and 10 days post-challenge. No lesions were observed in the pre-challenge period. Dogs fed the homeopathic on day 5 after challenge presented 40% less crusted lesion and 60% less hair loss than the control. Nitric oxide (NOx) and reactive oxygen species (ROS) levels increased over time in both groups (P < 0.001). On day 40 of the experiment, lower levels of NOx (P < 0.001) and ROS (P < 0.025) were observed in the TRA group than in the CO group. In contrast, glutathione S-transferase activity increased over time in both groups (P < 0.001); however, this increase on day 40 was more significant in the dogs in the treated group than in the control (P < 0.010). We conclude that the preventive addition of homeopathic reduced the incidence of skin lesions and prevented oxidative stress caused by canine atopic dermatitis

    La adición de diseleniuro de difenilo en las dietas de codorniz mejora la calidad de la carne

    Objetivo. El diseleniuro de difenilo (Ph2Se2) es un compuesto de selenio orgánico que es conocido por sus características antioxidantes. Por lo tanto, el objetivo de este estudio fue evaluar si Ph2Se2 en las dietas de codorniz (Coturnix japonica) influye en el estado oxidativo/antioxidante y la calidad de la carne. Materiales y métodos. Se proporcionaron cuatro dietas (0; 0.3; 0.6; 0.9 ppm Ph2Se2) a 56 codornices machos (Coturnix japonica) distribuidas en un diseño completamente aleatorizado con 14 repeticiones para verificar si Ph2Se2 cambiaría su sangre y tejido oxidativo/antioxidante, estado, lo que llevaría a una mejora en la calidad de la carne. Resultados. La adición de Ph2Se2 en las dietas provocó un aumento de la actividad antioxidante de enzimas como la catalasa, la superóxido dismutasa y la glutatión peroxidasa, lo que redujo los niveles de oxidación en la sangre y los tejidos. Además de eso, observamos una mejora en la calidad de la carne de codorniz; en otras palabras, observamos una mayor capacidad para retener agua, una reducción en la pérdida de agua debido a la cocción y una menor intensidad del color amarillo en las mamas de las aves que fueron alimentadas con Ph2Se2. Conclusiones. Por lo tanto, concluimos que la mejora de la defensa antioxidante en los tejidos proporcionada por Ph2Se2 tiene un efecto beneficioso sobre la calidad de la carne

    Objective. The aim of this study was to evaluate AChE activity in total blood and the FRAP levels in samples from dogs with mammary tumors before and after surgery, as well as the relationship between these variables with immunohistochemical markers of tumor (E-caderina, ki-67, COX-2). Materials and methods. In this study, 13 dogs with mammary tumors were divided into two groups (A and B). The group A was formed by dogs with tumors smaller than 3 cm of diameter, and the group B was formed by dogs with tumor of 3 cm of diameter or larger. The AChE activity and FRAP levels were evaluated before and after surgery and the immunohistochemistry were performed at the tumors. Results. The AChE activity was significantly increased (p&lt;0.05) in dogs with mammary cancer compared to control animals, and neither surgery or tumor size affected the AChE activity (p&gt;0.05). FRAP levels before surgery were significantly lower (p&lt;0.05) compared to control animals. Also, FRAP levels increased significantly after surgery in animals of the group A compared to data before surgery, a fact not observed in dogs from the group B. E-cadherin showed low significant positive correlation with FRAP levels (r=0.37, P-value=0.05); COX-2 showed a moderate significant positive correlation to FRAP (r=0.55, P-value&lt;0.05); and COX-2 showed a low significant positive correlation to AChE (r=0.32, P-value=0.01). Conclusions. AChE and antioxidant levels are modified in dogs with mammary cancer. These variables are involved in various physiological functions, and thus, they might be related to disease pathogenesis.Objetivo. El objetivo de este estudio fue evaluar la actividad de la AChE en sangre total y los niveles de FRAP en muestras de perros con tumores mamarios antes y después de la cirugía, así como la relación entre estas variables con marcadores inmunohistoquímicos de tumores (E-caderina, ki-67 , COX - 2). Materiales y métodos. En este estudio, 13 perros con tumores mamarios se dividieron en dos grupos (A y B). El grupo A estaba formado por perros con tumores menores de 3 cm de diámetro y el grupo B estaba formado por perros con tumor de 3 cm de diámetro o más. La actividad de AChE y los niveles de FRAP se evaluaron antes y después de la cirugía y la inmunohistoquímica se realizó en los tumores. Resultados. La actividad de la AChE aumentó significativamente (p &lt;0,05) en perros con cáncer mamario en comparación con los animales control, y ni la cirugía ni el tamaño tumoral afectaron la actividad de la AChE (p&gt; 0,05). Los niveles de FRAP antes de la cirugía fueron significativamente más bajos (p &lt;0,05) en comparación con los animales control. Además, los niveles de FRAP aumentaron significativamente después de la cirugía en animales del grupo A en comparación con los datos antes de la cirugía, hecho que no se observó en perros del grupo B.     Dietary addition of curcumin favors weight gain and has antioxidant, antiinflammatory and anticoccidial action in dairy calves

    Background: Curcumin has been used as an additive in the diet of animals in recent years due to the potent medicinal properties of this molecule. Objective: To evaluate whether the addition of curcumin to the diet of calves at different phases (pre- and post-weaning) has a positive effect on metabolic profile, performance, and anti-coccidian action. Methods: Thirtythree Holstein calves were selected at various phases of development: Experiment 1 (E1: n=10) 18±7 (pre-weaning), Experiment 2 (E2: n=11) 64±4 (pre-weaning) and Experiment 3 (E3: n=12) 95±8 (post-weaning) days of life. The calves were separated in three groups according to their phase of development. In each experiment, animals were divided into two sub-groups: control and curcumin. The curcumin groups received 200 mg of additive per animal/day either in milk (pre-weaning) or concentrate (post-weaning). Fecal collections were performed on days 0, 10 and 15 of the experiment to count Eimeria oocysts per gram of feces and to perform fecal score analysis. Complete blood counts, oxidant and antioxidant profiles, protein metabolism markers, lipid levels, glucose levels, and animal weights were measured. Analyses of digestibility and composition of the diet used in Experiment 3 (post-weaning) were also performed. Results: Independent of phase, animals that received curcumin had greater weight gain on days 0 to 15 (E1, E2 and E3 p=0.04, 0.001 and 0.001, respectively), probably due to the increased digestibility of hay and concentrate at 72h (p=0.03 and 0.02, respectively). The supplemented calves had lower level of oxidants (thiobarbituric acid reactive substances –TBARS- and reactive oxygen species –ROS-), indicating that free radical levels in serum and lipid peroxidation were lower. This was probably due to increased enzymatic antioxidants gluthatione S-transferase (E1, E2 and E3 p=0.001, 0.001 and 0.02, respectively), catalase (E1 p=0.001) and superoxide dismutase (E3 p=0.001) in treated animals at day 15. Furthermore, calves receiving curcumin had lower numeric number of Eimeria infection during the experimental period, and the difference was significant in day 15 (E1 and E2 p=0.02, and 0.001, respectively). Conclusion: Curcumin supplementation to dairy calves has coccidiostatic potential, favoring weight gain.Antecedentes: Curcumina tem sido usado como aditivo na dieta de animais nos últimos anos, devido as propriedades medicinais potente dessa molécula. Objetivo: Avaliar se a adição de curcumina na alimentação de bezerras em diferentes fases (pré e pós-desmame) apresenta efeito positivo sobre perfil metabólico, desempenho e ação anti-coccidéo. Métodos: Para isso, 33 bezerros holandeses foram selecionados em vários estágios de desenvolvimento: Experimento 1 (E1: n=10) 18±7 (prédesmame), Experimento 2 (E2: n=11) 64±4 (pré-desmame) e Experimento 3 (E3: n=12) 95±8 (pós-desmame) dias de vida. Para todos os experimentos o período experimental foi de 15 dias, assim como foram delineados com dois grupos: controle e tratados com curcumina. Os grupos de curcumina receberam 200 mg do aditivo por animal/dia no leite (pré-desmame) ou em concentrado (pós-desmame). Coletas de fezes e sangue foram realizadas nos dias 0, 10 e 15 de experimento para contagem de oocistos de Eimeria por grama de fezes e análise de escore fecal. Do sangue colhido foram realizados: hemograma, perfil oxidante e antioxidante, metabolismo proteico, lipídico, glicose, além da pesagem dos animais. Também foi realizado análise de digestibilidade da dieta total ofertada aos animais do Experimento 3 (pós-desmame). Resultados: Independentemente da fase, os animais que receberam curcumina tiveram maior ganho de peso do dia 0 a 15 (E1, E2 e E3, p=0,04, 0,001 e 0,001, respectivamente), provavelmente devido ao aumento da digestibilidade ao feno e concentrado após 72 h (p=0,03 e 0,02, respectivamente). Nos bezerros suplementados, observou-se menor nível de oxidantes (substâncias reativas ao ácido tiobarbitúrico e espécies reativas de oxigênio), ou seja, os níveis séricos de radicais livres e a peroxidação lipídica foram menores. Isto foi provavelmente devido a antioxidantes enzimáticos glutationa S-transferase (E1, E2 e E3, p=0,001, 0,001 e 0,02, respectivamente), catalase (E1, p=0,001) e superóxido dismutase (E3, p=0,001) aumentando nestes animais tratados no dia 15. Além disso, bezerros recebendo curcumina tiveram menores níveis de infecção por Eimeria durante o período experimental e significativos no dia 15 (E1 e E2, p=0,02 e 0,001, respectivamente). Conclusão: A suplementação de curcumina aumenta o potencial coccidiostático e favorece o ganho de peso.Antecedentes: Curcumina ha sido utilizada como aditivo en la dieta de animales en los últimos años, debido a las potentes propiedades medicinales de esa molécula. Métodos: Se seleccionaron 33 terneros Holstein en varias etapas de desarrollo: Experimento 1 (E1: n=10) 18±7 (pre-destete), Experimento 2 (E2: n=11) 64±4 (pre-destete) y Experimento 3 (E3: n=12) 95±8 (post-destete) días de vida. Para todos los experimentos, el período experimental fue de 15 días. Los animales se dividieron en dos grupos: control y tratados con curcumina. Los grupos con curcumina recibieron una dosis de 200 mg del aditivo por animal/día en la leche (pre-destete) o en el concentrado (post-destete). Las colectas de heces y sangre fueron realizadas en los días 0, 10 y 15 para conteo de ooquistes de Eimeria por gramo de heces y análisis de puntaje fecal. Se realizó hemograma, perfil oxidante y antioxidante, metabolismo proteico, lipídico, glucosa sanguínea y pesaje de los animales. También se realizó análisis de digestibilidad de la dieta total para los animales del Experimento 3 (post-destete). Resultados: Encontramos que, independientemente de la fase, los animales que recibieron curcumina tuvieron una mayor ganancia de peso en los días 0 a 15 (E1, E2 y E3, p=0,04, 0,001 y 0,001, respectivamente), probablemente debido al aumento de la digestibilidad del heno y concentrado a las 72 h (p=0,03 y 0,02, respectivamente). En los terneros suplementados observamos un nivel más bajo de oxidantes (oxidantes (sustancias reactivas al ácido tiobarbitúrico y especies reactivas de oxígeno); es decir, los niveles séricos de radicales libres y la peroxidación lipídica fueron más bajos. Esto se debió, probablemente, a los antioxidantes enzimáticos glutatión S-transferasa (E1, E2 y E3 p=0,001, 0,001 y 0,02 respectivamente), catalasa (E1, p=0,001) y superóxido dismutasa (E3, p=0,001) que aumentaron en los animales tratados al día 15. Además, los terneros que recibieron curcumina tuvieron niveles más bajos de infección por Eimeria durante el período experimental y fueron significativos en el día 15 (E1 y E2, p=0,02 y 0,001, respectivamente).     Spray-dried porcine plasma added to diets contaminated with aflatoxins and fumonisins shows beneficial effects to piglet health

    Abstract This study was aimed to analyze the effects of spray-dried porcine plasma (SDPP) on the health of post weaning piglets challenged with diets contaminated with aflatoxins and fumonisins. Fifty-six male piglets (7.15 ± 0.61 kg) were allocated in four groups: CTL group received a regular diet; SDPP group received a regular diet and 6% SDPP; MYC group received a diet containing 300 µg/kg aflatoxins and 8,000 µg/kg fumonisins; group MYC+SDPP received 300 µg/kg aflatoxins, 8,000 µg/kg fumonisins and 6% SDPP. The animals that received the experimental diet containing mycotoxins (MYC group) had lower weight gain at the end of the experiment compared to the other treatments. Animals receiving SDPP showed decreased urea levels throughout the experiment (P<0.05). Animals from MYC group presented an increased on reactive oxygen species (ROS) and thiobarbituric acid reactive substances (TBARS) levels and decreased catalase activity (P<0.05). In contrast, SDPP prevented the increase of ROS and TBARS and stimulated superoxide dismutase activity (P<0.05). In conclusion, diet contaminated with mycotoxins (group MYC) caused subclinical intoxication in the piglets, as observed by the increase on free radical’s production and lipid peroxidation. Conversely, SDPP presented a protective effect, minimizing the effects of oxidative stress caused by aflatoxins and fumonisins ingestion

    Influence of experimental canine ehrlichiosis on the E-ADA activity and purine levels in serum and possible functional correlations with pathogenesis

    The aim of this study was to evaluate adenosine deaminase activity and purines levels in serum of dogs experimentally infected by Ehrlichia canis. Banked serum samples of dogs divided into two groups with five animals each: healthy animals and animals infected by E. canis. The concentration of purines (adenosine triphosphate (ATP), adenosine diphosphate (ADP), adenosine monophosphate (AMP), adenosine, inosine, hypoxanthine, xanthine and uric acid), and adenosine deaminase (E-ADA) activity in sera were evaluated. Samples were collected on days 12 and 30 post-infection (PI). The E-ADA activity showed a significant reduction on day 12 PI, and increased on day 30 PI in dogs infected with E. canis. On day 12, an increase in seric concentration of ATP, ADP and adenosine was verified, and different levels of hypoxanthine, xanthine and uric acid had a drastic reduction in infected compared healthy dogs (P< 0.05). However, on day 30 PI, the levels of seric ADP and AMP decreased, unlike the concentration of xanthine and uric acid that increased significantly in infected dogs (P< 0.05). Therefore, the activity of E-ADA and purine levels are altered in experimental canine ehrlichiosis, probably with the purpose of modulating the pathogenesis of the disease related to immune response, oxidative stress and coagulation disorders in acute phase. © 2013 Elsevier B.V

    Novel markers of inflammatory response and hepatic dysfunction in canine leishmaniasis

    Dogs are the main host of Leishmania infantum, and the clinical presentation may range from asymptomatic to systemic manifestations. The immune mechanisms in infected, but clinically healthy dogs, prevails Th1 response mediated by cytokines. In this sense, adenosine deaminase (ADA) and butyrylcholinesterase (BChE) are considered as key enzymes in several physiological processes, including the modulation of inflammatory process. Considering the variable immune response against Leishmania and the known participation of ADA and BChE, the aim of this study was to assess the relation between these two enzymes with the inflammatory response as well as hepatic function in dogs naturally infected with L. infantum. For this purpose, the activity of ADA and BChE was assessed in sera of 24 dogs naturally infected with L. infantum, plus 17 healthy dogs. The naturally infected dogs had clinical signs compatible with leishmaniasis and sera activities of ADA (P<0.01) and BChE (P<0.05) decreased, when compared to the healthy group. The reduction of ADA activity probably represented an effect on inflammatory response, especially due to the decreased hydrolysis of extracellular adenosine, might in order to protect against tissue damage and, also, setting a down-regulation on pro-inflammatory cytokines. BChE enzyme had no effect on modulating the immune response in leishmaniasis, but it decreased, a fact may related to deficiency of synthesis in the liver. Therefore, ADA and BChE activities reduced probably in order to protect against extra tissue damage and due failure in synthesis, respectively
