    Does Logic Help Us Beat Monty Hall?

    The classical Monty Hall problem entails that a hypothetical game show contestant be presented three doors and told that behind one door is a car and behind the other two are far less appealing prizes, like goats. The contestant then picks a door, and the host (Monty) is to open a different door which contains one of the bad prizes. At this point in the game, the contestant is given the option of keeping the door she chose or changing her selection to the remaining door (since one has already been opened by Monty), after which Monty opens the chosen door and the contestant wins the prize which lies behind it. Inspired by the work of Morrow, Oman and Salminen (2016, “Game Show Shenanigans: Monty Hall Meets Mathematical Logic,” Elemente der Mathematik 71(4), pp. 145-155), we consider several logic-themed variants of this problem. Among these are versions where d doors and p prizes reside behind some p of these doors, and the contestant is permitted to present Monty with q random true/false questions concerning the location of the prizes, to which Monty must respond truthfully. Our results extend those of the original paper, and involve a combination of probabilistic techniques and exhaustive computation using a computer program

    Antioxidant Potential of Juglans Nigra, Black Walnut, Husks Extracted Using Supercritical Carbon Dioxide with an Ethanol Modifier

    The black walnut, Junglas nigra, is indigenous to eastern North America, and abscission of its fruit occurs around October. The fruit consists of a husk, a hard shell, and kernel. The husk is commonly discarded in processing, though it contains phenolic compounds that exhibit antioxidant and antimicrobial properties. For this study, black walnut husks were extracted using supercritical carbon dioxide with an ethanol modifier. The effects of temperature, ethanol concentration, and drying of walnut husks prior to extraction upon antioxidant potential were evaluated using a factorial design of experiments. The solvent density was held constant at 0.75 g/mL. The optimal extraction conditions were found to be 68°C and 20 wt-% ethanol in supercritical carbon dioxide. At these conditions, the antioxidant potential as measured by the ferric reducing ability of plasma (FRAP) assay was 0.027 mmol trolox equivalent/g (mmol TE/g) for dried walnut husk and 0.054 mmol TE/g for walnut husks that were not dried. Antioxidant potential was also evaluated using the total phenolic content (TPC) and 1,1-diphenyl-2-picryl-hydrazyl (DPPH) assays and the FRAP assay was found to linearly correlate to the TPC assa

    The Rice Miniature Inverted Repeat Transposable Element mPing Is an Effective Insertional Mutagen in Soybean

    Insertional mutagenesis of legume genomes such as soybean (Glycine max) should aid in identifying genes responsible for key traits such as nitrogen fixation and seed quality. The relatively low throughput of soybean transformation necessitates the use of a transposon-tagging strategy where a single transformation event will produce many mutations over a number of generations. However, existing transposon-tagging tools being used in legumes are of limited utility because of restricted transposition (Ac/Ds: soybean) or the requirement for tissue culture activation (Tnt1: Medicago truncatula). A recently discovered transposable element from rice (Oryza sativa), mPing, and the genes required for its mobilization, were transferred to soybean to determine if it will be an improvement over the other available transposon-tagging tools. Stable transformation events in soybean were tested for mPing transposition. Analysis of mPing excision at early and late embryo developmental stages revealed increased excision during late development in most transgenic lines, suggesting that transposition is developmentally regulated. Transgenic lines that produced heritable mPing insertions were identified, with the plants from the highest activity line producing at least one new insertion per generation. Analysis of the mPing insertion sites in the soybean genome revealed that features displayed in rice were retained including transposition to unlinked sites and a preference for insertion within 2.5 kb of a gene. Taken together these findings indicate that mPing has the characteristics necessary for an effective transposon-tagging resource

    Arid3b Is Critical for B Lymphocyte Development

    Arid3a and Arid3b belong to a subfamily of ARID (AT-rich interaction domain) transcription factors. The Arid family is involved in regulating chromatin accessibility, proliferation, and differentiation. Arid3a and Arid3b are closely related and share a unique REKLES domain that mediates their homo- and hetero-multimerization. Arid3a was originally isolated as a B cell transcription factor binding to the AT rich matrix attachment regions (MARS) of the immunoglobulin heavy chain intronic enhancer. Deletion of Arid3a results in a highly penetrant embryonic lethality with severe defects in erythropoiesis and hematopoietic stem cells (HSCs). The few surviving Arid3a-/- (<1%) animals have decreased HSCs and early progenitors in the bone marrow, but all mature lineages are normally represented in the bone marrow and periphery except for B cells. Arid3b-/- animals die around E7.5 precluding examination of hematopoietic development. So it is unclear whether the phenotype of Arid3a loss on hematopoiesis is dependent or independent of Arid3b. In this study we circumvented this limitation by also examining hematopoiesis in mice with a conditional allele of Arid3b. Bone marrow lacking Arid3b shows decreased common lymphoid progenitors (CLPs) and downstream B cell populations while the T cell and myeloid lineages are unchanged, reminiscent of the adult hematopoietic defect in Arid3a mice. Unlike Arid3a-/- mice, HSC populations are unperturbed in Arid3b-/- mice. This study demonstrates that HSC development is independent of Arid3b, whereas B cell development requires both Arid3a and Arid3b transcription factors

    Real-time individualized training vectors for experiential learning.

    Military training utilizing serious games or virtual worlds potentially generate data that can be mined to better understand how trainees learn in experiential exercises. Few data mining approaches for deployed military training games exist. Opportunities exist to collect and analyze these data, as well as to construct a full-history learner model. Outcomes discussed in the present document include results from a quasi-experimental research study on military game-based experiential learning, the deployment of an online game for training evidence collection, and results from a proof-of-concept pilot study on the development of individualized training vectors. This Lab Directed Research & Development (LDRD) project leveraged products within projects, such as Titan (Network Grand Challenge), Real-Time Feedback and Evaluation System, (America's Army Adaptive Thinking and Leadership, DARWARS Ambush! NK), and Dynamic Bayesian Networks to investigate whether machine learning capabilities could perform real-time, in-game similarity vectors of learner performance, toward adaptation of content delivery, and quantitative measurement of experiential learning

    Recycling of Dilute Deacetylation Black Liquor to Enable Efficient Recovery and Reuse of Spent Chemicals and Biomass Pretreatment Waste

    Deacetylation/dilute alkaline pretreatment followed by mechanical refining (DMR) has been proven as an effective process for biomass sugar liberation without severe chemical modification to lignin. Previous research has been focused on optimizing deacetylation conditions, reducing energy consumptions in mechanical refining, and improving sugar yields and titers in enzymatic hydrolysis. To successfully commercialize this process, another critical challenge is to develop a robust process to balance water usage, recover spent chemicals, and utilize waste carbons from the dilute deacetylation waste liquor. In this work, a new process modification and strategy is pioneered to recycle and reuse the weak black liquor (WBL) in order to reduce water, chemical, and energy usage while increasing both inorganic and organic contents in the WBLto facilitate downstream processing. Results suggest that the accumulation did not lower acetyl and lignin removal in alkaline pretreatment, resulting in comparable sugar yields in enzymatic hydrolysis. Sodium and potassium were found to be the two most important inorganic compounds in the recycled WBL. Moreover, the accumulated sodium and phenolic compounds did not inhibit the downstream ethanol fermentation processes. Finally, techno-economic analysis (TEA) showed a decrease in the minimum ethanol selling price (MESP) by ~5 to 15 cents per gallon of ethanol resulting from the inclusion of the recycling of weak black liquor when compared to a conventional non-recycling process
