Modulation of Toll-like receptors in psoriatic patients during therapy with adalimumab
Toll-like receptors (TLRs) are a key part of the innate immune system that detect pathogen-associated molecular patterns (PAMPs) of microorganisms and their stimulation results in the activation of signaling pathways leading to the modulation of inflammatory and immune responses. Since psoriasis is a complex, inflammatory and immune skin disease, characterized by an abnormal immune response and increased proliferation of keratinocytes, with an increased production of proinflammatory cytokines, TLRs could play an important role in the pathogenesis of the disease. We propose to assess the modulation of TLR expression on psoriatic skin of patients treated with Adalimumab and systemic conventional therapies. We therefore recruited fifteen patients: ten were treated with adalimumab and five with systemic conventional therapies; their clinical conditions were analyzed by PASI index and skin biopsies were evaluated for TLR1 and TLR2 expression by immunohistochemistry assays. Our data suggest adalimumab is not only able to improve the clinical condition of psoriatic patients, but also to modulate TLR1 and TLR2 expression involved in psoriasis, as in healthy skin. Adalimumab is a most promising biological drug able to orchestrate immune and inflammatory responses in psoriatic lesions, recovering TLR expression on basal keratinocytes and improving clinical conditions of psoriatic patients, with no evident side effects
Pathology of myelinated axons in the PLP-deficient mouse model of spastic paraplegia type 2 revealed by volume imaging using focused ion beam-scanning electron microscopy
Advances in electron microscopy including improved imaging techniques and state-of-the-art detectors facilitate imaging of larger tissue volumes with electron microscopic resolution. In combination with genetic tools for the generation of mouse mutants this allows assessing the three-dimensional (3D) characteristics of pathological features in disease models. Here we revisited the axonal pathology in the central nervous system of a mouse model of spastic paraplegia type 2, the Plp−/Y mouse. Although PLP is a bona fide myelin protein, the major hallmark of the disease in both SPG2 patients and mouse models are axonal swellings comprising accumulations of numerous organelles including mitochondria, gradually leading to irreversible axonal loss. To assess the number and morphology of axonal mitochondria and the overall myelin preservation we evaluated two sample preparation techniques, chemical fixation or high-pressure freezing and freeze substitution, with respect to the objective of 3D visualization. Both methods allowed visualizing distribution and morphological details of axonal mitochondria. In Plp−/Y mice the number of mitochondria is 2-fold increased along the entire axonal length. Mitochondria are also found in the excessive organelle accumulations within axonal swellings. In addition, organelle accumulations were detected within the myelin sheath and the inner tongue. We find that 3D electron microscopy is required for a comprehensive understanding of the size, content and frequency of axonal swel- lings, the hallmarks of axonal pathology
Polyethylene Glycol Epirubicin-Loaded Transcatheter Arterial Chemoembolization Procedures Utilizing a Combined Approach with 100 and 200 μm Microspheres: A Promising Alternative to Current Standards
PURPOSE:To report clinical effectiveness, toxicity profile, and prognostic factors of combined 100 μm ± 25 and 200 μm ± 50 epirubicin-loaded polyethylene glycol (PEG) microsphere drug-eluting embolic transcatheter arterial chemoembolization protocol in patients with hepatocellular carcinoma.
MATERIALS AND METHODS:
In this prospective, single-center, single-arm study with 18 months of follow-up, 36 consecutive patients (mean age 69.9 y ± 10.8; 26 men, 10 women; 54 naïve lesions) were treated. Embolization was initiated with 100 μm ± 25 microspheres, and if stasis (10 heart beats) was not achieved, 200 μm ± 50 microspheres were administered. Each syringe (2 mL) of PEG microsphere was loaded with 50 mg of epirubicin. Results were evaluated using Modified Response Evaluation Criteria In Solid Tumors with multidetector computed tomography/magnetic resonance imaging at 1, 3-6, 9-12, and 15-18 months. Toxicity profile was assessed by laboratory testing before and after the procedure. Complications were recorded. Postembolization syndrome (PES) was defined as onset of fever/nausea/pain after the procedure. Patient/lesion characteristics and treatment results were correlated with predicted outcome using regression analysis. Child-Pugh score was A in 86.1% of patients (31/36) and B in 13.9% (5/36).
RESULTS:
In 10 of 21 lesions, < 2 cm in diameter (47.5%) stasis was achieved with 100 μm ± 25 microspheres only, whereas all other lesions required adjunctive treatment with 200 μm ± 50 microspheres. Reported adverse events were grade 1 acute liver bile duct injury (3/39 cases, 7.7%) and PES (grade 2; 3/39 cases, 7.7%). Complete response (CR) at 1, 3-6, 9-12, and 15-18 months was 61.1%, 65.5%, 63.63%, and 62.5%. Objective response (CR + partial response) at 1, 3-6, 9-12, and 15-18 months was 83.3%, 65.85%, 63.63%, and 62.5%. No single factor (laboratory testing, etiology, patient status, hepatic status, tumor characteristics, administration protocol) predicted outcomes except for albumin level at baseline for CR (P < .05, odds ratio = 1.09).
CONCLUSIONS:
The combined microsphere sizing strategy was technically feasible and yielded promising results in terms of effectiveness and toxicity
Basin-scale interaction between post-LGM faulting and morpho-sedimentary processes in the S. Eufemia Gulf (Southern Tyrrhenian Sea)
The integrated interpretation of high-resolution multibeam bathymetry, seismic profiles and backscatter data in the S. Eufemia Gulf (SEG; Calabro-Tyrrhenian continental margin, south-eastern Tyrrhenian Sea) documents the relationship between postglacial fault activity and morpho-sedimentary processes. Three systems of active normal faults that affect the seafloor or the shallow subsurface, have been identified: 1) the S. Eufemia fault system located on the continental shelf with fault planes mainly oriented N26E-N40E; 2) the offshore fault system that lies on the continental slope off Capo Suvero with fault planes mainly oriented N28E-N60E; 3) the Angitola Canyon fault system located on the seafloor adjacent to the canyon having fault planes oriented N60EN85E. The faults produce a belt of linear escarpments with vertical displacement varying from a few decimeters to about 12 m. One of the most prominent active structures is the fault F1 with the highest fault length (about 9.5 km). Two main segments of this fault are identified: a segment characterised by seafloor deformation with metric slip affecting Holocene deposits; a segment characterised by folding of the seafloor. A combined tectonostratigraphic model of an extensional fault propagation fold is proposed here to explain such different deformation.In addition to the seabed escarpments produced by fault deformation, in the SEG, a strong control of fault activity on recent sedimentary processes is clearly observed. For example, canyons and channels frequently change their course in response to their interaction with main tectonic structures. Moreover, the upper branch of the Angitola Canyon shows straight flanks determined by fault scarps. Tectonics also determined different sediment accumulation rates and types of sedimentation (e.g., the accumulation of hanging wall turbidite deposits and the development of contourite deposits around the Maida Ridge). Furthermore, the distribution of landslides is often connected to main fault scarps and fluids are locally confined in the hanging wall side of faults and can escape at the seabed, generating pockmarks aligned along their footwall
Investigation of VOCs associated with different characteristics of breast cancer cells
The efficacy of breath volatile organic compounds (VOCs) analysis for the screening of patients bearing breast cancer lesions has been demonstrated by using gas chromatography and artificial olfactory systems. On the other hand, in-vitro studies suggest that VOCs detection could also give important indications regarding molecular and tumorigenic characteristics of tumor cells. Aim of this study was to analyze VOCs in the headspace of breast cancer cell lines in order to ascertain the potentiality of VOCs signatures in giving information about these cells and set-up a new sensor system able to detect breast tumor-associated VOCs. We identified by Gas Chromatography-Mass Spectrometry analysis a VOCs signature that discriminates breast cancer cells for: i) transformed condition; ii) cell doubling time (CDT); iii) Estrogen and Progesterone Receptors (ER, PgR) expression, and HER2 overexpression. Moreover, the signals obtained from a temperature modulated metal oxide semiconductor gas sensor can be classified in order to recognize VOCs signatures associated with breast cancer cells, CDT and ER expression. Our results demonstrate that VOCs analysis could give clinically relevant information about proliferative and molecular features of breast cancer cells and pose the basis for the optimization of a low-cost diagnostic device to be used for tumors characterization
Excitons and charged excitons in semiconductor quantum wells
A variational calculation of the ground-state energy of neutral excitons and
of positively and negatively charged excitons (trions) confined in a
single-quantum well is presented. We study the dependence of the correlation
energy and of the binding energy on the well width and on the hole mass. The
conditional probability distribution for positively and negatively charged
excitons is obtained, providing information on the correlation and the charge
distribution in the system. A comparison is made with available experimental
data on trion binding energies in GaAs-, ZnSe-, and CdTe-based quantum well
structures, which indicates that trions become localized with decreasing

Macromolecular and electrical coupling between inner hair cells in the rodent cochlea
Inner hair cells (IHCs) are the primary receptors for hearing. They are housed in the cochlea and convey sound information to the brain via synapses with the auditory nerve. IHCs have been thought to be electrically and metabolically independent from each other. We report that, upon developmental maturation, in mice 30% of the IHCs are electrochemically coupled in ‘mini-syncytia’. This coupling permits transfer of fluorescently-labeled metabolites and macromolecular tracers. The membrane capacitance, Ca2+-current, and resting current increase with the number of dye-coupled IHCs. Dual voltage-clamp experiments substantiate low resistance electrical coupling. Pharmacology and tracer permeability rule out coupling by gap junctions and purinoceptors. 3D electron microscopy indicates instead that IHCs are coupled by membrane fusion sites. Consequently, depolarization of one IHC triggers presynaptic Ca2+-influx at active zones in the entire mini-syncytium. Based on our findings and modeling, we propose that IHC-mini-syncytia enhance sensitivity and reliability of cochlear sound encoding
Surgical resection is superior to TACE in the treatment of HCC in a well selected cohort of BCLC-B elderly patients—A retrospective observational study
Simple Summary Hepatocellular carcinoma (HCC) is the most common primary liver malignancy. Liver transplantation (LT) and surgical resection (SR) are currently the primary treatments with curative intent. Nevertheless, more than two-thirds of patients are elderly and, therefore, excluded from LT; while, according to the Barcelona Clinic Liver Cancer (BCLC) system, SR should only be offered to a small group of patients with early stage HCC. The identification in stage B of an intermediate subgroup of patients that fulfill the criteria for surgery may play an important role in the implementation of potentially curative treatments. Hepatocellular carcinoma (HCC) usually develops in cirrhotic liver, with high recurrence rates. However, considering its increasing detection in non-cirrhotic liver, the choice of treatment assumes particular relevance. This study aimed to investigate outcomes of patients among BCLC stages and enrolled for surgical resection (SR) according to a more complex evaluation, to establish its safety and efficacy. A total of 186 selected HCC patients (median age 73.2 yrs), submitted to SR between January 2005 and January 2021, were retrospectively analyzed. Of which, 166 were staged 0, A, B according to the BCLC system, while 20 with a single large tumor (>5 cm) were classified as stage AB. No perioperative mortality was recorded; complications occurred in 48 (25.80%) patients, and all but two were Clavien-Dindo grade I-II. Median follow-up was 9.2 years. Subsequently, 162 recurrent patients (87,1%) were selected for new treatments. Comparable overall survival rates (OS) were observed at 1, 3, 5, and 10 years in 0, A, B and AB stages (p = 0.2). Eventually, the BCLC-B group was matched to 40 BCLC-B patients treated (2015-2021) with TACE. Significant differences in baseline characteristics (p <0.0001) and in OS were observed at 1 and 3 years (p <0.0001); a significant difference was also observed in oncological outcomes, in terms of the absence, residual, or relapse of disease (p <0.05). Surgery might be a valid treatment in HCC for patients affected by chronic liver disease in a condition of compensation, up to BCLC-B stage. Surgical indication for liver resection in case of HCC should be extensively revised
Exigências nutricionais de mudas de bananeira tipo prata submetidas à deficiência de nutrientes.
O objetivo deste trabalho foi avaliar a produção de matéria seca de mudas de banana tipo prata submetidas a omissão de macronutrientes em solução nutritiva. Um experimento foi conduzido em casa de vegetação no Campus JK da UFVJM, no delineamento experimental inteiramente casualizado, com três repetições com uma planta por vaso em solução nutritiva. Os tratamentos foram: Completo (macro e micronutrientes) e a omissão de um nutriente por vez (-N, -P, -K, -Ca, -Mg , -S, -B, -Cu, -Fe, -Mn e -Zn) em solução nutritiva e dois genótipos (Prata-Ana e seu híbrido PA42-44). A adubação no tratamento completo consistiu de: 210,1 mg de N, 31 mg de P, 234,6 mg de K, 200,4 mg de Ca, 48,6 mg de Mg, 64,2 mg de S, 500 ?g de B, 20 ?g de Cu, 648 ?g de Cl, 5.022 ?g de Fe, 502 ?g de Mn, 11 ?g de Mo e 50 ?g de Zn por litro. O período experimental foi de 100 dias e foram avaliadas as seguintes características: área foliar, peso de massa seca da parte aérea e de raízes. O genótipo Prata-Anã é mais exigente em N e Ca e é menos exigente em K e Mg e micronutrientes do que o genótipo PA42-44, tendo a mesma exigência em P, S e B
