    Medium-term performance and maintenance of SUDS:a case-study of Hopwood Park Motorway Service Area, UK

    One of the main barriers to implementing SUDS is concern about performance and maintenance costs since there are few well-documented case-studies. This paper summarizes studies conducted between 2000 and 2008 of the performance and maintenance of four SUDS management trains constructed in 1999 at the Hopwood Park Motorway Service Area, central England. Assessments were made of the wildlife value and sedimentation in the SUDS ponds, the hydraulic performance of the coach park management train, water quality in all management trains, and soil/sediment composition in the grass filter strip, interceptor and ponds. Maintenance procedures and costs were also reviewed. Results demonstrate the benefits of a management train approach over individual SUDS units for flow attenuation, water treatment, spillage containment and maintenance. Peak flows, pond sediment depth and contaminant concentrations in sediment and water decreased through the coach park management train. Of the 2007 annual landscape budget of £15,000 for the whole site, the maintenance costs for SUDS only accounted for £2,500 compared to £4,000 for conventional drainage structures. Furthermore, since sediment has been attenuated in the management trains, the cost of sediment removal after the recommended period of three years was only £554 and, if the design is not compromised, less frequent removal will be required in future

    Febrile Convulsions as a Problem in Waiting Times

    A simple hypothesis on the occurrence of febrile convulsions is posed. This is (1) that certain children are genetically predisposed, and (2) that for such children, the probability of the first attack occurring in year 1, 2, etc., is essentially constant up to the end of the period during which the attacks can take place. These assumptions lead to a probability model which agrees well with data on age at first attack in the Town of Tecumseh, Mich. It appears that susceptibility does not gradually diminish, but rather disappears abruptly. The calculation yields an estimate of the proportion of children, among those susceptible, who will have at least one attack before they exceed the age of suceptibility. Penetrance of the condition, measured in this way, is nearly complete: 93.8% will have manifested the condition by age 7. The population frequency of the susceptible type of child is found to be 3.90%. If one assumes the simple dominant mode of inheritance of Frantzen et al. to be correct, this estimate of population frequency leads to an estimate of the gene frequency. This is approximately p = 0.02. The homozygote would therefore have a frequency of 4 per 10,000. Some speculations on verifying this theory of inheritance by identifying the homozygote are given.     The opioid-related receptor, ORL1, is activated by the neuropeptide nociceptin/orphanin FQ (N/OFQ) and inhibits high-voltage-activated (HVA) calcium channel currents (I(Ca)) via a G-protein-coupled mechanism. Endocytosis of ORL1 receptor during prolonged N/OFQ exposure was proposed to cause N-type voltage-gated calcium channel (VGCC) internalization via physical interaction between ORL1 and the N-type channel. However, there is no direct electrophysiological evidence for this mechanism in dorsal root ganglion (DRG) neurons or their central nerve terminals. The present study tested this using whole-cell patch-clamp recordings of HVA I(Ca) in rat DRG neurons and primary afferent excitatory synaptic currents (eEPSCs) in spinal cord slices. DRG neurons were classified on the basis of diameter, isolectin-B4 (IB4) binding and responses to capsaicin, N/OFQ and a μ-opioid agonist, DAMGO. IB4-negative neurons less than 20 μm diameter were selectively responsive to N/OFQ as well as DAMGO. In these neurons, ORL1 desensitization by a supramaximal concentration of N/OFQ was not followed by a decrease in HVA I(Ca) current density or proportion of whole-cell HVA I(Ca) contributed by N-type VGCC as determined using the N-type channel selective blocker, ω-conotoxin CVID. There was also no decrease in the proportion of N-type I(Ca) when neurons were incubated at 37°C with N/OFQ for 30 min prior to recording. In spinal cord slices, N/OFQ consistently inhibited eEPSCs onto dorsal horn neurons. As observed in DRG neurons, preincubation of slices in N/OFQ for 30 min produced no decrease in the proportion of eEPSCs inhibited by CVID. In conclusion, no internalization of the N-type VGCC occurs in either the soma or central nerve terminals of DRG neurons following prolonged exposure to high, desensitizing concentrations of N/OFQ.NHMRC Grant: 056992

    The social construction of 101 non-emergency video relay services for deaf signers

    How the police prepare for and engage with a citizen who is deaf and uses British Sign Language (BSL) is a national problem. From the perspective of deaf sign language users, the police remain largely inaccessible and unprepared in how to accommodate their linguistic needs. Four regional forces have responded to this issue by introducing a local solution, a bespoke 101 non-emergency video relay service (101VRS). Independent VRS companies function as the auxiliary service, mediating video calls to a 101 helpline. This service was identified as a simple solution that relied on minimal resourcing and input from the police. In using Pinch and Bijker’s social construction of technology (SCOT) framework, we look at competing interpretations of the 101VRS concept and how this has led to a range of intended and unintended solutions and problems (Pinch TJ and Bijker WE (1984) The social construction of facts and artefacts: or how the sociology of science and the sociology of technology might benefit each other. To maintain the investment in improving access to the police, we recommend harmonization of 101VRS nationally, and ongoing consultation with how front-line services can become better prepared at assisting deaf citizens

    Using field evaluation and systematic iteration to rationalize the accumulation of omega-3 long-chain polyunsaturated fatty acids in transgenic Camelina sativa

    The Brassicaceae Camelina sativa (gold of pleasure) is now an established niche crop and being used as a transgenic host for a range of novel seed traits. Most notable of these is the accumulation of omega-3 long-chain polyunsaturates such as eicosapentaenoic acid (EPA) and docosahexaenoic acid (DHA), fatty acids normally only found in marine organisms. As part of continued efforts to optimize the accumulation of these non-native fatty acids via seed-specific expression of algal genes, a new series of iterative constructs was built and introduced into Camelina. Seed fatty acid composition was determined, and the presence of EPA and DHA was confirmed. To provide an additional level of evaluation, full environmental release was carried out on selected events, providing a real-world gauntlet against which to assess the performance of these novel lines. Composition of the seed oil triacylglycerol was determined by mass spectrometry, allowing for conclusions as to the contribution of different activities to the final accumulation of EPA and DHA. Since these data were derived from field-grown material, they also represent a robust demonstration of the stability of the omega-3 LC-PUFA trait in Camelina.     Long-term follow-up of residual masses after chemotherapy in patients with non-seminomatous germ cell tumours

    This retrospective study was undertaken to determine the outcome of patients with non-seminomatous germ cell tumour who achieved a serological complete response but who had residual radiologic abnormalities upon completion of primary platinum-based chemotherapy. This was an analysis of 76 consecutive patients treated at Mount Vernon Hospital between 1983 and 1997. The patients were placed into two groups based upon whether they had surgical resection (surgery group, 48 patients) or observation (observation group, 28 patients) of residual radiologic masses on completion of initial chemotherapy (to enter the surgery group, complete surgical resection must have been achieved). The primary end-points were progression-free and overall survival. The percentage of patients alive with median follow-up 66 months was 90% for the surgery group and 80% for the observation group (P= 0.53, not significant). The percentage of patients continuously disease-free was 70% in the surgery group and 80% in the observation group (P= 0.31, not significant). In the small sub-group of patients with differentiated teratoma (TD) in the primary lesion who were observed, there was no excess risk of relapse or death. Patients who achieve a serological complete response after primary chemotherapy, but are left with ≤ 2 cm radiological masses that are not cystic and have responded, can be safely observed with diligent follow-up. © 2000 Cancer ResearchCampaig

    Field trial evaluation of the accumulation of omega-3 long chain polyunsaturated fatty acids in transgenic Camelina sativa: making fish oil substitutes in plants

Here we describe the first field-based evaluation of a terrestrial source of these essential nutrients, synthesised in the seeds of transgenic Camelina sativa plants via the heterologous reconstitution of the omega-3 long chain polyunsaturated fatty acid biosynthetic pathway. Our data demonstrate the robust nature of this novel trait, and the feasibility of making fish oils in genetically modified crops.     Transgenic plants as a sustainable, terrestrial source of fish oils

    An alternative, sustainable source of omega-3 long chain polyunsaturated fatty acids is widely recognized as desirable, helping to reduce pressure on current sources (wild capture fisheries) and providing a de novo source of these health beneficial fatty acids. This review will consider the efforts and progress to develop transgenic plants as terrestrial sources of omega-3 fish oils, focusing on recent developments and the possible explanations for advances in the field. We also consider the utility of such a source for use in aquaculture, since this industry is the major consumer of oceanic supplies of omega-3 fish oils. Given the importance of the aquaculture industry in meeting global requirements for healthy foodstuffs, an alternative source of omega-3 fish oils represents a potentially significant breakthrough for this production system.
