    Effect of Vibrio cholerae on Chemokine Gene Expression in HT29 cells and its Modulation by Lactobacillus GG

    Epithelial cells participate in the innate immune response to pathogenic bacteria by elaborating chemokines. This study examined the effect of Vibrio cholerae and Lactobacillus rhamnosus GG on inflammatory chemokine gene expression in the HT29 human intestinal epithelial cell line. HT29 cells were exposed to V. cholerae 0139, Lactobacillus or both for 2 h and cultured further thereafter for 4 h. RNA was extracted from the cells and expression of genes for chemokines and related molecules was quantitated by real time PCR using a pathway-focused PCR array. TLR4 was silenced using shRNA and output of interleukin-8 (IL-8) into the media quantitated with and without V. cholerae exposure. NFκB and p38 MAP kinase activation were determined by immunoblotting for IκBα and phosphorylated p38. Vibrio cholerae significantly upregulated gene expression for the neutrophil chemoattractant CXCL chemokines, IL-8, CXCL and CXCL in HT29 cells, while downregulating the expression of macrophage-attracting C-C chemokines. TLR4 silencing did not reduce IL-8 output from HT29 cells in response to V. cholerae. IκBα degradation was noted in the HT29 cells soon after exposure to V. cholerae and this recovered over time after removal of bacteria. p38 MAP kinase activation was not noted. Vibrio cholerae upregulated the expression of neutrophil attractant chemokines, most prominently IL-8, in HT29 cells, but downregulated macrophage-attracting chemokines. Probiotic lactobacilli modulated the IL-8, but not the other chemokine gene changes, in response to V. cholerae

    Fish biomass estimation by calibrating the echointegrator deflection against catch data

    Acoustic survey for fish resources was conducted using echosounder (EK-400) with echointegrator (QD). The echointegrator coupled with echosounder sums-up the echo signal received. The sum of the echo signal received per nautical mile covered is an index of the quantum of fish recorded and therefore a measure of the relative density of fish in surveyed area. It is converted into absolute biomass using the calibrarion constant obtained by correlating the trawl catch data against the echointegrator reading corresponding to the opening of the net. The calibration constant thus arrived at was 1327 kg/n.mile corresponding to 1 mm integrator deflection per nautical mile covered

    Nature of fireside deposits in a bagasse and groundnut shell fired 20 MW thermal boiler

    The nature of deposit formation on the fireside surfaces of the boiler tubes in the various parts (water walls, platen superheater, final superheater, economizer, electrostatic precipitator etc.) of a commercial 20 MW stoker-fired boiler being fired with a mixture of 80% bagasse and 20% groundnut shell has been analyzed. The deposits in the various portions of the boiler were characterized by particle size analysis, chemical analysis, X-ray diffraction and scanning electron microscopy. The deposits were found to be mainly quartz, alkali and alkaline earth silicates and sulfates. From the phase constitution and other microscopic characteristics of the deposit, it can be inferred that the silicates in the deposit formed through inertial impaction and the sulfates formed by vapor phase deposition

    Combustion and deposit formation behavior on the fireside surfaces of a pulverized fuel boiler fired with a blend of coal and petroleum coke

    The thermochemistry of the combustion of a blend of coal and 5% petroleum coke was analyzed. Thermodynamic modeling and microscopic techniques were used to study the behavior of the inorganic constituents upon combustion of the blend of coal and petroleum coke. The chemical composition and phase constitution of the combustion products, as well as the deposits at several temperatures corresponding to those at the various parts of the boiler, were deduced by free-energy minimization. These results were compared with actual results obtained from a commercial pulverized fuel boiler fired with coal and petroleum coke blend. The deposits on the fireside surfaces of the boiler tubes in the various parts (water walls, platen superheater, final superheater, economizer, and electrostatic precipitator) of the commercial pulverized fuel boiler fired with coal and 5% petroleum coke were characterized by particle size analysis, chemical analysis, x-ray diffraction, optical microscopy, and scanning electron microscopy. The combustion gas composition was measured using a portable on-line gas analyzer for N 2 , O 2 , CO 2 , H 2 O, CO, NO, and SO 2 . The thermodynamically predicted compositions and phase constitutions for the gas phase as well as the condensed phases are in good agreement with the experimental results

    Temperature varied from 17.6 to 28.5°C, salinity values from 32.12 to 35.21 x 10"" and dissolved oxygen from 0.8 to 4.41 ml/1. No identifiable relationship could be established between these three parameters and the total fish abundance, probably because the catch was made up of many species having different requirements. Higher abundance of trawl catches was from January to May when bottom water temperature was relatively low. The highest level of abundance of 2764 kg/hr in February 1989 was recorded when the parameters were 26°C, 33.6 x 10" and 2.71 ml/1; and the lowest level of 43.2 kg/hr in July 1988 was when the parameters were 25.3°C, 34.48 x 10'^ and 1.6 ml/1

    We report on test results of a novel ring imaging Cherenkov (RICH) detection system consisting of a 3 meter long gaseous C4F8O radiator, a focusing mirror, and a photon detector array based on Hamamatsu multi-anode photomultiplier tubes.     Nonlinear Behaviour of Perforated Plate with Lining

    Perforated plate with lining has a construction of plate with perforation and a lining plate welded together to form a single plate. This type of plate is used as an acoustic sonar dome. Perforated plate with lining (PPL) is prone to stress concentration and subsequently such structural system falls into the large strain category. Experimental investigation on PPL is carried out in the present study to determine the static deflection of the plate. Numerical method is also followed for geometric nonlinear analysis using finite element method as an iterative interactive procedure. The deflection obtained from the numerical method is 8 per cent less than that obtained from experimental method.
