    This research aims to develop a new program for first year education with a view to cultivating students\u27 abilities to work in groups, to conduct case studies, and to analyze and present their findings in a creative way. In this program, each group was assigned a case study of an NPO in Himeji area. Five or six students work in a group and conduct research on the target organization. They interviewed NPO workers and participate in social activities to deepen their understanding of the organization. In the end, each group shared the result of the study through powerpoint presentations and short films. Survey was conducted to analyze the responses of both students and local NPO workers to this educational experiment. The survey shows that this program contributed developing students\u27 interests in social activities. NPO workers also evaluated this attempt positively as an approach to co-develop project-based learning that could offer merits both for students and NPOs

    Solitary Peutz-Jeghers type hamartomatous polyps in the duodenum are not always associated with a low risk of cancer: two case reports

    INTRODUCTION: A hamartomatous polyp without associated mucocutaneous pigmentation or a family history of Peutz-Jeghers Syndrome is diagnosed as a solitary Peutz-Jeghers type hamartomatous polyp. As compared with Peutz-Jeghers Syndrome, Peutz-Jeghers type hamartomatous polyps are diagnosed with a lower risk of cancer and are regarded as a different disorder. CASE PRESENTATION: In case one, we describe an 84-year-old Japanese man with a 14 mm duodenal polyp. Endoscopic mucosal resection was performed and histological examination showed findings suggestive of a hamartomatous polyp with a focus of well-differentiated adenocarcinoma. In case two, we describe a 76-year-old Japanese man who had been treated for prostate, rectal and lung cancer. Upper gastrointestinal endoscopy revealed a duodenal polyp measuring 15 mm in diameter. Endoscopic mucosal resection was performed, and histological examination showed findings suggestive of a hamartomatous polyp. Liver and thyroid cancers were found after the endoscopic treatment. CONCLUSION: Although duodenal solitary hamartomatous polyps are associated with a lower risk of cancer, four patients, including our cases, have been diagnosed with cancerous polyps. Patients with duodenal solitary hamartomatous polyps should be treated by endoscopic or surgical resection and need whole-body screening

    Early effects of oral administration of lafutidine with mosapride compared with lafutidine alone on intragastric pH values

    <p>Abstract</p> <p>Background</p> <p>The ideal medication for treatment of acid related diseases should have a rapid onset of action to promote hemostasis and resolution of symptoms. The aim of our study was to investigate the inhibitory effects on gastric acid secretion after a single oral administrations of lafutidine, is a newly synthesized H2-receptor antagonist, with mosapride 5 mg or lafutidine alone.</p> <p>Methods</p> <p>Ten <it>Helicobacter pylori </it>negative male subjects participated in this randomized, two-way crossover study. Intragastric pH was monitored continuously for 4 hours after a single oral administration of lafutidine 10 mg or lafutidine 10 mg with mosapride 5 mg (the lafutidine being administrated one hour after the mosapride). Each administration was separated by a 7-day washout period.</p> <p>Results</p> <p>The average pH during the 4-hour period after administration of lafutidine 10 mg with mosapride 5 mg was higher than after lafutidine 10 mg alone (median: 5.25 versus 4.58, respectively; <it>p </it>= 0.0318). During the 3–4 hour study period, lafutidine 10 mg with mosapride 5 mg provided a higher pH, compared to lafutidine 10 mg alone (median: 7.28 versus 6.42; <it>p </it>= 0.0208).</p> <p>Conclusion</p> <p>In <it>H. pylori </it>negative healthy male subjects, an oral dose of lafutidine 10 mg with mosapride 5 mg more rapidly increased intragastric pH than lafutidine 10 mg alone.</p

    The impacts of convective parameterization and moisture triggering on AGCM-simulated convectively coupled equatorial waves

    This study examines the impacts of convective parameterization and moisture convective trigger on convectively coupled equatorial waves simulated by the Seoul National University (SNU) atmospheric general circulation model (AGCM). Three different convection schemes are used, including the simplified Arakawa-Schubert (SAS) scheme, the Kuo (1974) scheme, and the moist convective adjustment (MCA) scheme, and a moisture convective trigger with variable strength is added to each scheme. The authors also conduct a "no convection" experiment with deep convection schemes turned off. Space-time spectral analysis is used to obtain the variance and phase speed of dominant convectively coupled equatorial waves, including the Madden-Julian oscillation (MJO), Kelvin, equatorial Rossby (ER), mixed Rossby-gravity (MRG), and eastward inertio-gravity (EIG) and westward inertio-gravity (WIG) waves. The results show that both convective parameterization and the moisture convective trigger have significant impacts on AGCM-simulated, convectively coupled equatorial waves. The MCA scheme generally produces larger variances of convectively coupled equatorial waves including the MJO, more coherent eastward propagation of the MJO, and a more prominent MJO spectral peak than the Kuo and SAS schemes. Increasing the strength of the moisture trigger significantly enhances the variances and slows down the phase speeds of all wave modes except the MJO, and usually improves the eastward propagation of the MJO for the Kuo and SAS schemes, but the effect for the MCA scheme is small. The no convection experiment always produces one of the best signals of convectively coupled equatorial waves and the MJO.open585

In particular, studies of gene expression dynamics and regulation landscape in COVID-19 infected individuals are limited. Here, we report on a thorough analysis of whole blood RNA-seq data from 465 genotyped samples from the Japan COVID-19 Task Force, including 359 severe and 106 non-severe COVID-19 cases. We discover 1169 putative causal expression quantitative trait loci (eQTLs) including 34 possible colocalizations with biobank fine-mapping results of hematopoietic traits in a Japanese population, 1549 putative causal splice QTLs (sQTLs; e.g. two independent sQTLs at TOR1AIP1), as well as biologically interpretable trans-eQTL examples (e.g., REST and STING1), all fine-mapped at single variant resolution. We perform differential gene expression analysis to elucidate 198 genes with increased expression in severe COVID-19 cases and enriched for innate immune-related functions. Finally, we evaluate the limited but non-zero effect of COVID-19 phenotype on eQTL discovery, and highlight the presence of COVID-19 severity-interaction eQTLs (ieQTLs; e.g., CLEC4C and MYBL2). Our study provides a comprehensive catalog of whole blood regulatory variants in Japanese, as well as a reference for transcriptional landscapes in response to COVID-19 infection

    DOCK2 is involved in the host genetics and biology of severe COVID-19

Here we conducted a genome-wide association study (GWAS) involving 2, 393 cases of COVID-19 in a cohort of Japanese individuals collected during the initial waves of the pandemic, with 3, 289 unaffected controls. We identified a variant on chromosome 5 at 5q35 (rs60200309-A), close to the dedicator of cytokinesis 2 gene (DOCK2), which was associated with severe COVID-19 in patients less than 65 years of age. This risk allele was prevalent in East Asian individuals but rare in Europeans, highlighting the value of genome-wide association studies in non-European populations. RNA-sequencing analysis of 473 bulk peripheral blood samples identified decreased expression of DOCK2 associated with the risk allele in these younger patients. DOCK2 expression was suppressed in patients with severe cases of COVID-19. Single-cell RNA-sequencing analysis (n = 61 individuals) identified cell-type-specific downregulation of DOCK2 and a COVID-19-specific decreasing effect of the risk allele on DOCK2 expression in non-classical monocytes. Immunohistochemistry of lung specimens from patients with severe COVID-19 pneumonia showed suppressed DOCK2 expression. Moreover, inhibition of DOCK2 function with CPYPP increased the severity of pneumonia in a Syrian hamster model of SARS-CoV-2 infection, characterized by weight loss, lung oedema, enhanced viral loads, impaired macrophage recruitment and dysregulated type I interferon responses. We conclude that DOCK2 has an important role in the host immune response to SARS-CoV-2 infection and the development of severe COVID-19, and could be further explored as a potential biomarker and/or therapeutic target

    Global distribution of material stocks: iron, copper and nickel

    Economic growth has caused a rapid rise in the use of metals and minerals. In addition, the world’s population is projected to rapidly increase, with increases concentrated in Asia and Africa. Asia has become the largest global user of natural resources, and established systems of production and consumption are tailored to the current high levels of natural resource use and the associated emissions. Closing the material cycle loop is a principal strategy to reduce natural resource consumption and associated emissions. Material flow analysis (MFA), widely used to assess the flow and stocks of materials within a system defined in time and space, is regarded as a core tool to assess the sustainability of material use. The objective of this study was to estimate material stocks of iron, copper, and nickel, for which the global demand has risen rapidly in recent years. Specifically, the goal was to examine transitions of the material stock of each substance from 1995 to 2010. The worldwide material stocks of iron, copper, and nickel drastically increased during this period, which coincided with an increase in demand for materials in Asia. The material stock of each substance in Asia increased almost threefold from 1995 to 2010. The material stock of iron, copper, and nickel in Asia accounted for 30%, 26%, and 20% of the world total in 1995, respectively, but these percentages increased to 42%, 37%, and 32% in 2010, respectively. There was also a drastic increase in the per-capita material stock of iron, copper, and nickel in Asia during the same period, but the per-capita material stock of each of these substances in Asia was not yet reached that of North America and Western Europe. The material stocks in Africa increased in line with the population growth in Africa from 1995 to 2010, so the per-capita material stock of each substance in Africa stayed about the same. The per-capita material stocks in Africa are also small compared with developed countries and regions. This situation shows the potential for a greatly increased demand as a result of expansions in production through accelerated industrialization as well as the increased demand for resources accompanying population and economic growth in Asia and Africa
