    Synthesis of Macrocyclic Hexaoxazole (6OTD) Dimers, Containing Guanidine and Amine Functionalized Side Chains, and an Evaluation of Their Telomeric G4 Stabilizing Properties

    Structure-activity relationship studies were carried out on macrocyclic hexaoxazole (6OTD) dimers, whose core structure stabilizes telomeric G-quadruplexes (G4). Two new 6OTD dimers having side chain amine and guanidine functional groups were synthesized and evaluated for their stabilizing ability against a telomeric G4 DNA sequence. The results show that the 6OTD dimers interact with the DNA to form 1:1 complexes and stabilize the antiparallel G4 structure of DNA in the presence of potassium cation. The guanidine functionalized dimer displays a potent stabilizing ability of the G4 structure, as determined by using a FRET melting assay (ΔTm = 14°C)

    Relationship between Weanling and Yearling Fleece Characters in the Corriedale Sheep

    This study has been conducted to test if the yearling fleece characters may be predicted from the weanling fleece characters, by investigating the relationship between the weanling and the yearling fleece characters with special reference to the grease yield, clean yield, clean yield percentage, stretched length, density and the fineness of the Corriedale sheep wool. The animals used were 11 Corriedale ewes born in the spring, 1955 at the Chugoku Stock Breeding Farm of the Department of Agriculture and Forestry. The wool samples were taken from left and right side and thigh regions of each sheep at weanling and yearling ages and each fleece character at both ages was measured and its relationship was analyzed. The results obtained were as follows: (1) Considerably high and significant regressions and correlations were found between weanling and yearling characters, such as the grease wool yield, stretched wool length at side region and the wool fineness at thigh region. (2) While the clean wool yields, clean wool yields percentages, wool densities and the wool fineness at side regions between both ages had no significant regressions and correlations. (3) It seems from the results of the present study that of all fleece characters measured, grease wool yield, stretched wool length at side region and the wool fineness at thigh region have considerably high values for predicting the yearling fleece characters from the weanling characters of the Corriedale sheep

    Detection of CpG Methylation in G-Quadruplex Forming Sequences Using G-Quadruplex Ligands

    Genomic DNA methylation is involved in many diseases and is expected to be a specific biomarker for even the pre-symptomatic diagnosis of many diseases. Thus, a rapid and inexpensive detection method is required for disease diagnosis. We have previously reported that cytosine methylation in G-quadruplex (G4)-forming oligonucleotides develops different G4 topologies. In this study, we developed a method for detecting CpG methylation in G4-forming oligonucleotides based on the structural differences between methylated and unmethylated G4 DNAs. The differences in G4 topologies due to CpG methylation can be discriminated by G4 ligands. We performed a binding assay between methylated or unmethylated G4 DNAs and G4 ligands. The binding abilities of fluorescent G4 ligands to BCL-2, HRAS1, HRAS2, VEGF G4-forming sequences were examined by fluorescence-based microtiter plate assay. The differences in fluorescence intensities between methylated and unmethylated G4 DNAs were statistically significant. In addition to fluorescence detection, the binding of G4 ligand to DNA was detected by chemiluminescence. A significant difference was also detected in chemiluminescence intensity between methylated and unmethylated DNA. This is the first study on the detection of CpG methylation in G4 structures, focusing on structural changes using G4 ligands

    FAN1 interaction with ubiquitylated PCNA alleviates replication stress and preserves genomic integrity independently of BRCA2

    Interstrand cross-link (ICL) hypersensitivity is a characteristic trait of Fanconi anemia (FA). Although FANCD2-associated nuclease 1 (FAN1) contributes to ICL repair, FAN1 mutations predispose to karyomegalic interstitial nephritis (KIN) and cancer rather than to FA. Thus, the biological role of FAN1 remains unclear. Because fork stalling in FAN1-deficient cells causes chromosomal instability, we reasoned that the key function of FAN1 might lie in the processing of halted replication forks. Here, we show that FAN1 contains a previously-uncharacterized PCNA interacting peptide (PIP) motif that, together with its ubiquitin-binding zinc finger (UBZ) domain, helps recruit FAN1 to ubiquitylated PCNA accumulated at stalled forks. This prevents replication fork collapse and controls their progression. Furthermore, we show that FAN1 preserves replication fork integrity by a mechanism that is distinct from BRCA2-dependent homologous recombination.     Ultra-High-Resolution Computed Tomography of the Lung: Image Quality of a Prototype Scanner

    Purpose: The image noise and image quality of a prototype ultra-high-resolution computed tomography (U-HRCT) scanner was evaluated and compared with those of conventional high-resolution CT (C-HRCT) scanners. Materials and Methods: This study was approved by the institutional review board. A U-HRCT scanner prototype with 0.25 mm × 4 rows and operating at 120 mAs was used. The C-HRCT images were obtained using a 0.5 mm × 16 or 0.5 mm × 64 detector-row CT scanner operating at 150 mAs. Images from both scanners were reconstructed at 0.1-mm intervals; the slice thickness was 0.25 mm for the U-HRCT scanner and 0.5 mm for the C-HRCT scanners. For both scanners, the display field of view was 80 mm. The image noise of each scanner was evaluated using a phantom. U-HRCT and C-HRCT images of 53 images selected from 37 lung nodules were then observed and graded using a 5-point score by 10 board-certified thoracic radiologists. The images were presented to the observers randomly and in a blinded manner. Results: The image noise for U-HRCT (100.87 ± 0.51 Hounsfield units [HU]) was greater than that for C-HRCT (40.41 ± 0.52 HU; P <.0001). The image quality of U-HRCT was graded as superior to that of C-HRCT (P <.0001) for all of the following parameters that were examined: margins of subsolid and solid nodules, edges of solid components and pulmonary ves sels in subsolid nodules, air bronchograms, pleural indentations, margins of pulmonary vessels, edges of bronchi, and interlobar fissures. Conclusion: Despite a larger image noise, the prototype U-HRCT scanner had a significantly better image quality than the C-HRCT scanners

    A new skull of an early diverging rorqual (Balaenopteridae, Mysticeti, Cetacea) from the late Miocene to early Pliocene of Yamagata, northeastern Japan

    The family of rorquals and humpback whales, Balaenopteridae includes the larg-est living animal on Earth, the blue whale Balaenoptera musculus. Many new taxa have been named, but not many from the western Pacific, except Miobalaenoptera numataensis from Japan. Here we describe an early balaenopterid, cf. M. numataensis from a late Miocene to early Pliocene sediment in Yamagata Prefecture, northeastern Japan. The species has a straight and sharp lateral ridge of the fovea epitubaria at the ventral surface of the periotic, and a dorsoventrally thin pars cochlearis. The new spec-imen provides knowledge of supposed ontogenetic variation and periotic morphology in poorly known fossil balaenopterids
