    Die Dissertation entwickelt unterschiedliche Verhandlungsmodelle und untersucht die GrΓΌnde fΓΌr den Zusammenbruch von Verhandlungen in drei bestimmten Verhandlungssituationen. Neben Verhandlungen zwischen autonomen Staaten in einem internationalen Rahmen, wird auch die Verhandlung zwischen KΓ€ufer und VerkΓ€ufer analysiert. Als GrΓΌnde fΓΌr den Zusammenbruch von Verhandlungen werden zum einen asymmetrische Information und zum anderen Commitment Probleme identifiziert.The dissertation develops different bargaining models and analyzes the reasons for the breakdown of negotiations in three specific bargaining situations.     Epidemiologic analysis of families with isolated anorectal malformations suggests high prevalence of autosomal dominant inheritance

    Background: Anorectal malformations (ARM) are rare abnormalities that occur in approximately 1 in 3000 live births with around 40% of patients presenting with isolated forms. Multiple familial cases reported, suggest underlying genetic factors that remain largely unknown. The recurrence in relatives is considered rare, however transmission rates of ARM by affected parents have never been determined before. The inheritance pattern of ARM was investigated in our database of patients with isolated ARM. Results: Within our cohort of 327 patients with isolated ARM we identified eight adult patients from eight families who had in total 16 children with their healthy spouse. Of these ten had ARM, resulting in a recurrence risk of approximately one in two live births (10 of 16; 62%). From 226 families with 459 siblings we found two affected siblings in five families. Hence, the recurrence risk of ARM among siblings is approximately one in 92 live births (5 of 459; 1.0%). Conclusions: Comparing the observed recurrence risk in our cohort with the prevalence in the general population, we see a 1500-fold increase in recurrence risk for offspring and a 32-fold increase if a sibling is affected. The recurrence risk of approximately 62% indicates an autosomal dominant mode of inheritance. Reliable figures on recurrence of ARM are becoming increasingly important since improved surgical techniques are able to maintain sexual function resulting in more offspring of patients with ARM. These data allow more precise counseling of families with ARM and support the need for genetic studies

    A CENP-S/X complex assembles at the centromere in S and G2 phases of the human cell cycle

    The functional identity of centromeres arises from a set of specific nucleoprotein particle subunits of the centromeric chromatin fibre. These include CENP-A and histone H3 nucleosomes and a novel nucleosome-like complex of CENPs -T,-W,-S and -X. Fluorescence cross-correlation spectroscopy and Forster resonance energy transfer (FRET) revealed that human CENP-S and -X exist principally in complex in soluble form and retain proximity when assembled at centromeres. Conditional labelling experiments show that they both assemble de novo during S phase and G2, increasing approximately three-to fourfold in abundance at centromeres. Fluorescence recovery after photobleaching (FRAP) measurements documented steady-state exchange between soluble and assembled pools, with CENP-X exchanging approximately 10 times faster than CENP-S (t(1/2) similar to 10 min versus 120 min). CENP-S binding to sites of DNA damage was quite distinct, with a FRAP half-time of approximately 160 s. Fluorescent two-hybrid analysis identified CENP-T as a uniquely strong CENP-S binding protein and this association was confirmed by FRET, revealing a centromere-bound complex containing CENP-S, CENP-X and CENP-T in proximity to histone H3 but not CENP-A. We propose that deposition of the CENP-T/W/S/X particle reveals a kinetochore-specific chromatin assembly pathway that functions to switch centromeric chromatin to a mitosis-competent state after DNA replication.     Light-sheet imaging of cleared and expanded samples creates terabyte-sized datasets that consist of many unaligned three-dimensional image tiles, which must be reconstructed before analysis. We developed the BigStitcher software to address this challenge. BigStitcher enables interactive visualization, fast and precise alignment, spatially resolved quality estimation, real-time fusion and deconvolution of dual-illumination, multitile, multiview datasets. The software also compensates for optical effects, thereby improving accuracy and enabling subsequent biological analysis

    Touch actions in digital learning: Increasing the performance of visual memory through active stimulus processing

    Perception and storage of new impressions and experiences from the environment is a necessary basis for a large part of the thinking and learning processes. Accordingly the humans are good at absorbing stimuli from their environment and integrating them into the long-term memory transfer. Previous experiments have shown that this retention performance can be optimized under certain conditions. For example, stimuli in the form of everyday objects can be well memorized if they are not only viewed passively, but actively processed, for example by touching them. The present study transfers these findings to learning with digital devices with touch surfaces. For this purpose, 42 persons either only viewed objects on the screen or actively moved them by dragging them. The results of this experiment prove that the additional haptic stimulus significantly increases retention in the visual memory and that the use of the touch function can thus have an effect on the learning process in addition to improved usability
