    Implications of Chinese and American mothers’ goals for children’s emotional distress: Investigation of a cultural transmission model

    This research examined a cultural transmission model in which differences in Chinese and American parents’ socialization goals foster differences in children’s emotional distress via parents’ responses to children’s performance. 397 Chinese and American mothers and their children (mean age = 13.19 years) participated in a 2-wave study spanning a year. Mothers reported on their self-improvement (i.e., children striving to improve) and self-worth (i.e., children feeling worthy) goals, as well as their responses to children’s performance. Children reported on their emotional distress (e.g., anxiety and depression). Chinese (vs. American) mothers’ greater endorsement of self-improvement goals predicted their more frequent use of failure-focused responses (e.g., highlighting children’s mistakes), which accounted for Chinese (vs. American) children’s heightened emotional distress over early adolescence

    Do Parents' Socialization Goals Influence How They Respond To Children's Performance?

    Much research has shown that Chinese and American parents endorsed different goals for their children (e.g., Chao, K., Ruth 1996; Ng, F. F., Pomerantz, E. M., & Lam, S. 2007) and substantial evidence reveals differences in Chinese and American parents’ practices (e.g. Bornstein, H. Marc, 2012; Miller, P., Wiley, A., Fung, H., & Liang, C. 1997). However, whether parents’ goals directly guide their practices has received little if any empirical attention. The goal of the current research is to investigate whether the differences in American and Hong Kong parents’ self-worth and self-improvement goals drive differences in how they respond to their children’s performance. 177 American and 196 Chinese mothers and their 4th-5th grade children participated. Half of the parents in both countries read a brochure that highlighted the importance of children’s feelings of worth in their reasoning abilities; the other half read an otherwise identical brochure highlighting the importance of children’s focus on improvement. Children completed a reasoning task in a separate room. After being informed of how children did, parents spent 5 minutes with their children. Parents also reported on their responses to children’s hypothetical success and failure after the reunion. The results indicate no significant differences in positive and negative verbal comments made during the parent-child reunion between goal manipulation conditions. However, above and beyond country differences, parents in the self-worth (vs. self-improvement) condition reported that they would focus more on their child’s success and less on their child’s failure.Ope

    Using Tetracysteine-Tagged TDP-43 with a Biarsenical Dye To Monitor Real-Time Trafficking in a Cell Model of Amyotrophic Lateral Sclerosis.

    TAR DNA-binding protein 43 (TDP-43) has been identified as the major constituent of the proteinaceous inclusions that are characteristic of most forms of amyotrophic lateral sclerosis (ALS) and ubiquitin positive frontotemporal lobar degeneration (FTLD). Wild type TDP-43 inclusions are a pathological hallmark of >95% of patients with sporadic ALS and of the majority of familial ALS cases, and they are also found in a significant proportion of FTLD cases. ALS is the most common form of motor neuron disease, characterized by progressive weakness and muscular wasting, and typically leads to death within a few years of diagnosis. To determine how the translocation and misfolding of TDP-43 contribute to ALS pathogenicity, it is crucial to define the dynamic behavior of this protein within the cellular environment. It is therefore necessary to develop cell models that allow the location of the protein to be defined. We report the use of TDP-43 with a tetracysteine tag for visualization using fluorogenic biarsenical compounds and show that this model displays features of ALS observed in other cell models. We also demonstrate that this labeling procedure enables live-cell imaging of the translocation of the protein from the nucleus into the cytosol

    Efficacy and tolerability of trastuzumab emtansine in advanced human epidermal growth factor receptor 2–positive breast cancer

Introduction: The management of human epidermal growth factor receptor 2 (HER2)–positive breast cancer has changed dramatically with the introduction and widespread use of HER2-targeted therapies. There is, however, relatively limited real-world information about the effectiveness and safety of trastuzumab emtansine (T-DM1) in Hong Kong Chinese patients. We assessed the efficacy and toxicity profiles among local patients with HER2-positive advanced breast cancer who had received T-DM1 therapy in the second-line setting and beyond. Methods: This retrospective study involved five local centres that provide service for over 80% of the breast cancer population in Hong Kong. The study period was from December 2013 to December 2015. Patients were included if they had recurrent or metastatic histologically confirmed HER2+ breast cancer who had progressed after at least one line of anti-HER2 therapy including trastuzumab. Patients were excluded if they received T-DM1 as first-line treatment for recurrent or metastatic HER2+ breast cancer. Patient charts including biochemical and haematological profiles were reviewed for background information, T-DM1 response, and toxicity data. Adverse events were documented during chemotherapy and 28 days after the last dose of medication. Results: Among 37 patients being included in this study, 28 (75.7%) had two or more lines of anti-HER2 agents and 26 (70.3%) had received two or more lines of palliative chemotherapy. Response assessment revealed that three (8.1%) patients had a complete response, eight (21.6%) a partial response, 11 (29.7%) a stable disease, and 12 (32.4%) a progressive disease; three patients could not be assessed. The median duration of response was 17.3 (95% confidence interval, 8.4-24.8) months. The clinical benefit rate (complete response + partial response + stable disease, ≄12 weeks) was 37.8% (95% confidence interval, 22.2%-53.5%). The median progression-free survival was 6.0 (95% confidence interval, 3.3-9.8) months and the median overall survival had not been reached by the data cut-off date. Grade 3 or 4 toxicities included thrombocytopaenia (13.5%), raised alanine transaminase (8.1%), anaemia (5.4%), and hypokalaemia (2.7%). No patient died as a result of toxicities. Conclusions: In patients with HER2-positive advanced breast cancer who have been heavily pretreated with anti-HER2 agents and cytotoxic chemotherapy, T-DM1 is well tolerated and provided a meaningful progression-free survival of 6 months and an overall survival that has not been reached.     Association of antipsychotic use with breast cancer:a systematic review and meta-analysis of observational studies with over 2 million individuals-CORRIGENDUM

    AIMS: Despite reports of an elevated risk of breast cancer associated with antipsychotic use in women, existing evidence remains inconclusive. We aimed to examine existing observational data in the literature and determine this hypothesised association. METHODS: We searched Embase, PubMed and Web of Scienceℱ databases on 27 January 2022 for articles reporting relevant cohort or case-control studies published since inception, supplemented with hand searches of the reference lists of the included articles. Quality of studies was assessed using the Newcastle-Ottawa Scale. We generated the pooled odds ratio (OR) and pooled hazard ratio (HR) using a random-effects model to quantify the association. This study was registered with PROSPERO (CRD42022307913). RESULTS: Nine observational studies, including five cohort and four case-control studies, were eventually included for review (N = 2 031 380) and seven for meta-analysis (N = 1 557 013). All included studies were rated as high-quality (seven to nine stars). Six studies reported a significant association of antipsychotic use with breast cancer, and a stronger association was reported when a greater extent of antipsychotic use, e.g. longer duration, was operationalised as the exposure. Pooled estimates of HRs extracted from cohort studies and ORs from case-control studies were 1.39 [95% confidence interval (CI) 1.11–1.73] and 1.37 (95% CI 0.90–2.09), suggesting a moderate association of antipsychotic use with breast cancer. CONCLUSIONS: Antipsychotic use is moderately associated with breast cancer, possibly mediated by prolactin-elevating properties of certain medications. This risk should be weighed against the potential treatment effects for a balanced prescription decision

    Pain Controlling and Cytokine-regulating Effects of Lyprinol, a Lipid Extract of Perna Canaliculus, in a Rat Adjuvant-induced Arthritis Model

    Using an adjuvant-induced arthritis rat model, we investigated the effects of a lipid extract of Perna canaliculus (LyprinolÂź) on pain. Radiological examinations, as well as levels of pro- and anti-inflammatory (AI) cytokines, were measured aiming to provide independent objective data to the pain controlling investigation. We confirmed the ability of LyprinolÂź to control pain at the initial phase of its administration; with similar efficacy to that observed with Naproxen. The pain scores slowly increased again in the group of rats treated with LyprinolÂź after day 9–14. The Naproxen-treated rats remained pain-free while treated. Both Naproxen and LyprinolÂź decreased the levels of the pro-inflammatory cytokines TNF-α and IFN-Îł, and increased that of IL-10. Extra-virgin olive oil was ineffective on cytokine secretion. Rats treated with LyprinolÂź were apparently cured after 1 year. This study confirms the AI efficacy of this lipid extract of P. canaliculus, its initial analgesic effect, its perfect tolerance and its long-term healing properties
