A numerical study of tillage tool wear during plowing of sandy soil
Wear of tillage tools during tilling operations leads to increase in power requirements, frequent downtimes for tool replacement, loss of production and an increase in the equipment and operational costs. A large number of variables---such as the moisture content in the soil, soil particle sizes, plowing speed, plowing depth, tool material properties, etc.---influence the tool wear rates. In turn, the tool wear has an effect on the power requirements for the tilling operations since a worn out tool requires larger amounts of power. The purpose of the present work is to (a). develop finite element models for predicting tool wear and power requirements, (b). validate the finite element models with experiments, and (c). study the effect of the above-mentioned variables on tool wear and power requirements. The ultimate objective is to understand the primary mechanisms behind tool wear and devise new designs for tillage tools to mitigate tool wear rates.
The tool wear process is treated as a two-body abrasion where a rigid sand particle scratches against the surface of the tillage tool at a prescribed speed. A finite element model is developed to model the scratching process. The deformations predicted by the finite element models are used in conjunction with the classical ploughing theory and the material removal factor to identify the mechanisms underlying the wear of the tillage tool. The predicted material removal factors are compared with experimental results and a parametric study is carried out to study the effect of various parameters on tool wear.
A two-dimensional finite element model simulating the soil-tool interaction during tilling is also developed. The purpose of this model is to study the feasibility of using the finite element methods to predict power requirements during a tilling process. The tillage tool is modeled as a rigid body moving through soil at a prescribed speed. The constitutive behavior of the soil is assumed to be given by the Drucker-Prager model. The forces acting on the tillage tool as the tool moves through soil are calculated and the power required for the tilling operation is calculated using these forces.
The computed values for tool material loss and power consumed during plowing operations are found to be in reasonable agreement with the experimental results. In addition, the parametric studies show that the primary wear mechanism is a combination of ploughing and cutting with cutting being dominant. More sophisticated models and analyses are needed to further understand the tillage tool wear process. The present work provides the foundation on which these advances can be made
Mouse Sphingosine Kinase 1a Is Negatively Regulated through Conventional PKC-Dependent Phosphorylation at S373 Residue
Sphingosine kinase is a lipid kinase that converts sphingosine into sphingosine-1-phosphate, an important signaling molecule with intracellular and extracellular functions. Although diverse extracellular stimuli influence cellular sphingosine kinase activity, the molecular mechanisms underlying its regulation remain to be clarified. In this study, we investigated the phosphorylation-dependent regulation of mouse sphingosine kinase (mSK) isoforms 1 and 2. mSK1a was robustly phosphorylated in response to extracellular stimuli such as phorbol ester, whereas mSK2 exhibited a high basal level of phosphorylation in quiescent cells regardless of agonist stimulation. Interestingly, phorbol ester-induced phosphorylation of mSK1a correlated with suppression of its activity. Chemical inhibition of conventional PKCs (cPKCs) abolished mSK1a phosphorylation, while overexpression of PKC alpha, a cPKC isoform, potentiated the phosphorylation, in response to phorbol ester. Furthermore, an in vitro kinase assay showed that PKC alpha directly phosphorylated mSK1a. In addition, phosphopeptide mapping analysis determined that the S373 residue of mSK1a was the only site phosphorylated by cPKC. Interestingly, alanine substitution of S373 made mSK1a refractory to the inhibitory effect of phorbol esters, whereas glutamate substitution of the same residue resulted in a significant reduction in mSK1a activity, suggesting the significant role of this phosphorylation event. Taken together, we propose that mSK1a is negatively regulated through cPKC-dependent phosphorylation at S373 residueopen
Withanolides and related steroids
Since the isolation of the first withanolides in the mid-1960s, over 600 new members of this group of compounds have been described, with most from genera of the plant family Solanaceae. The basic structure of withaferin A, a C28 ergostane with a modified side chain forming a δ-lactone between carbons 22 and 26, was considered for many years the basic template for the withanolides. Nowadays, a considerable number of related structures are also considered part of the withanolide class; among them are those containing γ-lactones in the side chain that have come to be at least as common as the δ-lactones. The reduced versions (γ and δ-lactols) are also known. Further structural variations include modified skeletons (including C27 compounds), aromatic rings and additional rings, which may coexist in a single plant species. Seasonal and geographical variations have also been described in the concentration levels and types of withanolides that may occur, especially in the Jaborosa and Salpichroa genera, and biogenetic relationships among those withanolides may be inferred from the structural variations detected. Withania is the parent genus of the withanolides and a special section is devoted to the new structures isolated from species in this genus. Following this, all other new structures are grouped by structural types.
Many withanolides have shown a variety of interesting biological activities ranging from antitumor, cytotoxic and potential cancer chemopreventive effects, to feeding deterrence for several insects as well as selective phytotoxicity towards monocotyledoneous and dicotyledoneous species. Trypanocidal, leishmanicidal, antibacterial, and antifungal activities have also been reported. A comprehensive description of the different activities and their significance has been included in this chapter. The final section is devoted to chemotaxonomic implications of withanolide distribution within the Solanaceae.

Governance in Family Business Groups: Resolving Multiple Contingencies to Sustain Entrepreneurial Capability
Family business groups (FBGs) are ubiquitous throughout the world. FBGs also pose unique governance issues that merit attention in order to explain FBGs’ evolution and longevity, as well as to understand FBGs’ performance on a variety of dimensions. In this chapter, we develop a framework to highlight the governance issues in FBGs, starting from understanding governance as managing goal congruence among different stakeholders and recognizing the owning family as an entrepreneurial entity. We suggest that an optimal FBG governance system involves a dynamically contingent system, based on managing complexity at both macro and micro levels, and that optimal governance in an FBG protects and enhances the capability of the owners to be entrepreneurial and construct opportunities from its environment. Based on our exposition, we provide several directions in which to advance research in this area.Post-print / Final draf

Tumor Volume Estimation and Quasi- Continuous Administration for Most Effective Bevacizumab Therapy
Bevacizumab is an exogenous inhibitor which inhibits the biological activity of human VEGF. Several studies have investigated the effectiveness of bevacizumab therapy according to different cancer types but these days there is an intense debate on its utility. We have investigated different methods to find the best tumor volume estimation since it creates the possibility for precise and effective drug administration with a much lower dose than in the protocol.We have examined C38 mouse colon adenocarcinoma and HT-29 human colorectal adenocarcinoma. In both cases, three groups were compared in the experiments. The first group did not receive therapy, the second group received one 200 μg bevacizumab dose for a treatment period (protocol-based therapy), and the third group received 1.1 μg bevacizumab every day (quasi-continuous therapy). Tumor volume measurement was performed by digital caliper and small animal MRI. The mathematical relationship between MRI-measured tumor volume and mass was investigated to estimate accurate tumor volume using caliper-measured data. A two-dimensional mathematical model was applied for tumor volume evaluation, and tumor- and therapy-specific constants were calculated for the three different groups. The effectiveness of bevacizumab administration was examined by statistical analysis.In the case of C38 adenocarcinoma, protocol-based treatment did not result in significantly smaller tumor volume compared to the no treatment group; however, there was a significant difference between untreated mice and mice who received quasi-continuous therapy (p = 0.002). In the case of HT-29 adenocarcinoma, the daily treatment with one-twelfth total dose resulted in significantly smaller tumors than the protocol-based treatment (p = 0.038). When the tumor has a symmetrical, solid closed shape (typically without treatment), volume can be evaluated accurately from caliper-measured data with the applied two-dimensional mathematical model.Our results provide a theoretical background for a much more effective bevacizumab treatment using optimized administration
Protocol for a randomised controlled trial of a decision aid for the management of pain in labour and childbirth [ISRCTN52287533]
BACKGROUND: Women report fear of pain in childbirth and often lack complete information on analgesic options prior to labour. Preferences for pain relief should be discussed before labour begins. A woman's antepartum decision to use pain relief is likely influenced by her cultural background, friends, family, the media, literature and her antenatal caregivers. Pregnant women report that information about analgesia was most commonly derived from hearsay and least commonly from health professionals. Decision aids are emerging as a promising tool to assist practitioners and their patients in evidence-based decision making. Decision aids are designed to assist patients and their doctors in making informed decisions using information that is unbiased and based on high quality research evidence. Decision aids are non-directive in the sense that they do not aim to steer the user towards any one option, but rather to support decision making which is informed and consistent with personal values. METHODS/DESIGN: We aim to evaluate the effectiveness of a Pain Relief for Labour decision aid, with and without an audio-component, compared to a pamphlet in a three-arm randomised controlled trial. Approximately 600 women expecting their first baby and planning a vaginal birth will be recruited for the trial. The primary outcomes of the study are decisional conflict (uncertainty about a course of action), knowledge, anxiety and satisfaction with decision-making and will be assessed using self-administered questionnaires. The decision aid is not intended to influence the type of analgesia used during labour, however we will monitor health service utilisation rates and maternal and perinatal outcomes. This study is funded by a competitive peer-reviewed grant from the Australian National Health and Medical Research Council (No. 253635). DISCUSSION: The Pain Relief for Labour decision aid was developed using the Ottawa Decision Support Framework and systematic reviews of the evidence about the benefits and risks of the non-pharmacological and pharmacological methods of pain relief for labour. It comprises a workbook and worksheet and has been developed in two forms – with and without an audio-component (compact disc). The format allows women to take the decision aid home and discuss it with their partner
