    Se ha estudiado la farmacocinética de la absorción gastrointestinal del busulfán en dos grupos de ratas, tras su administración por vía oral. La dosis influye en el valor de la constante de absorción (Ka) y en la concentración máxima normalizada (Cmax), presentando ambos parámetros una disminución al aumentar la dosis de 0,5 a 5 mg. El empleo de un modelo aparente monocompartimental con absorción de primer orden permite una adecuada descripción del perfil de niveles plasmáticos del antineoplásico. El dimetilsulfóxido, utilizado como disolvente del busulfán, no interfiere en los parámetros farmacocinéticos orales del mismo.We have studied the pharmacokinetics of gastrointestinal absorption of busulfan in two groups of rats, after oral administration. The dose effect on the value of the absorption constant (Ka) and the normalized maximum concentration (Cmax), presenting both parameters decreased with increasing dose from 0.5 to 5 mg. Using a model with monocompartimental apparent first order absorption allows an adequate description of the profile of plasma levels of antineoplastic. The dimethylsulfoxide used as busulfan solvent, does not interfere with oral pharmacokinetic parameters of it

    Affine convex body semigroups

    In this paper we present a new kind of semigroups called convex body semigroups which are generated by convex bodies of R^k. They generalize to arbitrary dimension the concept of proportionally modular numerical semigroup of [7]. Several properties of these semigroups are proven. Affine convex body semigroups obtained from circles and polygons of R^2 are characterized. The algorithms for computing minimal system of generators of these semigroups are given. We provide the implementation of some of them

    The pseudogenes of Mycobacterium leprae reveal the functional relevance of gene order within operons

    Almost 50 years following the discovery of the prokaryotic operon, the functional relevance of gene order within operons remains unclear. In this work, we take advantage of the eroded genome of Mycobacterium leprae to add evidence supporting the notion that functionally less important genes have a tendency to be located at the end of its operons. M. leprae’s genome includes 1133 pseudogenes and 1614 protein-coding genes and can be compared with the close genome of M. tuberculosis. Assuming M. leprae’s pseudogenes to represent dispensable genes, we have studied the position of these pseudogenes in the operons of M. leprae and of their orthologs in M. tuberculosis. We observed that both tend to be located in the 3′ (downstream) half of the operon (P-values of 0.03 and 0.18, respectively). Analysis of pseudogenes in all available prokaryotic genomes confirms this trend (P-value of 7.1 × 10−7). In a complementary analysis, we found a significant tendency for essential genes to be located at the 5′ (upstream) half of the operon (P-value of 0.006). Our work provides an indication that, in prokarya, functionally less important genes have a tendency to be located at the end of operons, while more relevant genes tend to be located toward operon starts

    Use of nanomaterials to conserve the stone of the Roman Theatre of Cartagena

    [EN] The Roman Theatre of Cartagena is an important example of the monumental architecture of Hispania. Various types of local stone were used in its construction, including a type of sandstone known as Tabaire. This paper describes the physical characteristics of Tabaire. We also identify its deterioration patterns and study its consolidation using lime nanoparticles and silicon dioxide (ethyl silicate). Initial analyses using the imaging techniques Optical Microscopy (OM), Scanning Electron Microscopy (SEM) and X-Ray Tomography (CT) show that it is an extremely porous, soft stone. The effectiveness of the consolidating treatments was verified using non-destructive methods based on identifying dispersion and penetrability in the substrate (OM and SEM) and detecting colour variation (CIE-L*a*b*) or minimally destructive methods, such as the peeling test and Shore-D hardness, to determine their effect on the characteristics of the material.[ES] El Teatro de Cartagena es un importante exponente de la arquitectura monumental de la Hispania romana. Para su construcción fueron utilizados diversos pétreos locales, entre ellos una arenisca conocida como Tabaire. Este artículo tiene como objetivo la caracterización física del Tabaire, así como la identificación de sus formas de deterioro y el estudio de su consolidación mediante nanopartículas de cal y de dióxido de silicio (silicato de etilo). Los análisis iniciales mediante técnicas de imagen como la Microscopía Óptica (OM), Microscopía Electrónica de Barrido (SEM) y Tomografía de Rayos X (CT) muestran que es una roca de elevada porosidad y escasa dureza.     Forest plantations in Manabí (Ecuador): assessment of fragmentation and connectivity to support dry tropical forests conservation

    In many tropical regions, national forests plantation programs have been promoted. Those plantations frequently contribute to habitat changes. However, the associated effects of forest plantations on habitat fragmentation and landscape connectivity are unclear. From 2008 to 2018, we examined land use changes, plantations, and deforestation of the Manabí province (Ecuador) provided by the Ecuadorian Ministry of the Environment. Four scenarios were created: (i) land uses in 2008, (ii) land uses in 2018, (iii) land uses in 2018 without deforestation, and iv) land uses in 2018 including reforestation. Fragmentation and connectivity metrics were analyzed using ArcGisPro and Graphad 2.6 software, respectively. Puma yagouaroundi was selected as the reference species. At regional scale, forest plantations had a significant effect on land uses changes and fragmentation during the study period. Forests decreased from 33.7% to 32.4% between 2008 and 2018, although other natural land uses, mostly those involving shrubs, increased by almost double (from 2.4% to 4.6%). Most of the deforestation affected native forests during this period, and most reforested areas in 2018 covered former agricultural land. Fragmentation decreased in the number of patches and increased in the average patch size. When considering reforestation, deforestation was higher than the reforested area (58 km2 of difference), increasing the number of patches but with smaller size. Reforestation increased connectivity with a higher number of links and distance, particularly in central and extreme northeast areas of Manabí province. The scenario without deforestation also increased connectivity for Puma yagouaroundi in the west part of the Manabí province. Our findings suggest that forest plantations contribute to forest conservation by increasing the connectivity between fragmented patches

    Evaluación del comportamiento mecánico de mezclas asfálticas templadas con 100 % de material reciclado

    The use of Half Warm Mixes with high Reclaimed Asphalt content (HWMRA) has the potential to generate significant environmental advantages such as the reduction in consumption of natural resources and the emission of gases into the atmosphere. This paper therefore focuses on demonstrating the viability of using these types of mixes in wearing courses. For this purpose, an HWMRA with 70 % and 100 % Reclaimed Asphalt Pavement (RAP) and emulsion were designed in the laboratory. The performance of the mixes was then assessed and compared with that of conventional Hot Mix Asphalt. In a second stage, the mixes were manufactured in-plant, and laid and compacted in an Accelerated Pavement Test track. The cores were then extracted and tested for stiffness modulus and resistance to fatigue. The results from the tests conducted with both the laboratory specimens and the cores showed that the performance of HWMRA is comparable to that of HMA. These findings encourage greater confidence in promoting the use of these types of sustainable asphalt mixes.La utilización de mezclas asfálticas templadas con alto contenido de asfalto reciclado (HWMRA) conlleva ventajas medioambientales como la reducción del consumo de recursos naturales y la emisión de gases a la atmósfera. Este artículo se centra en mostrar la viabilidad de este tipo de mezclas para capas de rodadura. Para ello, se diseñaron mezclas HWMRA con 70 % y 100 % de asfalto reciclado en el laboratorio y se evaluó y comparó su comportamiento con una mezcla caliente convencional. En una segunda etapa, las mezclas fueron fabricadas en planta, extendidas y compactadas en una pista de ensayo acelerado de pavimentos. A continuación, se extrajeron testigos y se ensayaron para conocer su módulo de rigidez y resistencia a fatiga. Tanto los resultados de laboratorio como tras la fabricación en planta y puesta en obra mostraron que el comportamiento de mezclas HWMRA es comparable al de mezclas calientes convencionales. Dicha conclusión puede aportar confianza a este tipo de mezclas sostenibles promoviendo su mayor utilización

    Tryptophan depletion affects compulsive behaviour in rats:strain dependent effects and associated neuromechanisms

    RATIONALE: Compulsive behaviour, present in different psychiatric disorders, such as obsessive-compulsive disorder, schizophrenia and drug abuse, is associated with altered levels of monoamines, particularly serotonin (5-hydroxytryptamine) and its receptor system. OBJECTIVES: The present study investigated whether 5-HT manipulation, through a tryptophan (TRP) depletion by diet in Wistar and Lister Hooded rats, modulates compulsive drinking in schedule-induced polydipsia (SIP) and locomotor activity in the open-field test. The levels of dopamine, noradrenaline, serotonin and its metabolite were evaluated, as well as the 5-HT(2A) and 5-HT(1A) receptor binding, in different brain regions. METHODS: Wistar rats were selected as high (HD) or low (LD) drinkers according to their SIP behaviour, while Lister hooded rats did not show SIP acquisition. Both strains were fed for 14 days with either a TRP-free diet (T−) or a TRP-supplemented diet (T+) RESULTS: The TRP depletion diet effectively reduced 5-HT levels in the frontal cortex, amygdala and hippocampus in both strains of rats. The TRP-depleted HD Wistar rats were more sensitive to 5-HT manipulation, exhibiting more licks on SIP than did the non-depleted HD Wistar rats, while the LD Wistar and the Lister Hooded rats did not exhibit differences in SIP. In contrast, the TRP-depleted Lister Hooded rats increased locomotor activity compared to the non-depleted rats, while no differences were found in the Wistar rats. Serotonin 2A receptor binding in the striatum was significantly reduced in the TRP-depleted HD Wistar rats. CONCLUSIONS: These results suggest that alterations of the serotonergic system could be involved in compulsive behaviour in vulnerable populations

    Effect of temperature on RNA silencing of a negative-stranded RNA plant virus: Citrus psorosis virus

    Citrus psorosis virus (CPsV), genus Ophiovirus, causes a bark scaling disease of citrus. CPsV virions are kinked filaments with three negative-stranded RNA molecules (vRNA) and a 48 kDa coat protein. The effect of temperature on symptom expression, virus accumulation and RNA silencing was examined in sweet orange seedlings (Citrus sinensis) graft-inoculated with three different CPsV isolates and grown in a glasshouse at 26/18°C or 32/26°C (day/night). Most plants kept in the cooler glasshouse showed a shock reaction in the first flush with shoot necrosis, and then moderate to intense chlorotic flecking and spotting in young leaves, whereas plants incubated at 32/26°C did not exhibit shoot necrosis, and young leaf symptoms were milder. Virus titre estimated by ELISA and by northern and dot blot hybridization paralleled symptom intensity, with significantly higher virus accumulation in plants incubated at 26/18°C. The amount of CPsV-derived small RNAs (CPsV-sRNAs) slightly increased at 32/26°C, with the ratio of CPsV-sRNA/vRNA being higher at 32/26°C than at 26/18°C. These results suggest that (i) CPsV infection induces RNA silencing in citrus plants, (ii) symptom intensity is associated with virus accumulation, and (iii) temperature increase enhances the RNA silencing response of citrus plants and decreases virus accumulation.Facultad de Ciencias Exacta

    Understanding the influence of filler type and asphalt binder content on the moisture and fatigue resistance of asphalt mortars

    An adequate moisture resistance is a key element to guarantee the durability of asphalt materials. This paper identifies the influence of filler typology and bitumen content on the mechanical response of asphalt mortars before and after water action. Two fillers were evaluated: Portland cement and Calcium carbonate, along with different contents of a penetration bitumen (B35/50). Stiffness, ductility, and fatigue were evaluated through a new protocol for asphalt mortar samples using a 3-point-bending test on DMA (Dynamic Mechanical Analyzer). The use of Portland cement presents higher stiffness, lower ductility, and improved fatigue and water resistance compared to Calcium carbonate. It is also possible to optimize bitumen content based on fatigue results. Content beyond the optimal reduce variations after water action but compromise fatigue resistance. Lower content leads to a poorer performance in both terms. This methodology enables asphalt mortar characterisation as a tool to optimise the design of asphalt materials.La influencia del tipo de polvo mineral y contenido de ligante bituminoso en la resistencia a la humedad y fatiga de morteros bituminosos. La resistencia al agua es clave para garantizar la durabilidad de los materiales asfálticos. Este estudio identifica la influencia del polvo mineral y contenido de betún en la respuesta mecánica de morteros bituminosos antes y después de la humedad. Se evaluaron dos tipos de filler: cemento Portland y carbonato cálcico (filler calizo), junto con distintos contenidos de betún (B35/50). Rigidez, ductilidad y fatiga fueron evaluados mediante un nuevo ensayo de fatiga a tres puntos para morteros empleando DMA (Dynamic Mechanical Analyzer). El cemento presenta más rigidez, menos ductilidad y mayor resistencia al agua y fatiga que el filler calizo. Los resultados de fatiga permiten además optimizar el contenido de betún. Contenidos por encima del óptimo reducen variaciones tras la humedad pero comprometen la resistencia a fatiga. Contenidos menores conllevan un peor comportamiento en ambos términos. Esta metodología permite usar la caracterización de morteros bituminosos para optimizar el diseño de materiales asfálticos

    Comparación del efecto de la concentración de polvo de neumático reciclado y polímeros en el comportamiento de ligantes para mezclas bituminosas

    Crumb rubber modified binders are environmental-friendly alternatives to polymer modified bitumens in asphalt mixtures. This paper compares the performance of both types of binders with different modifier contents. Six binders were characterised by conventional tests and analysed using the UCL method. This method evaluates different properties of binders regarding their role in asphalt mixtures (cohesion, water and thermal sensitivity and resistance to ageing). Results showed that i) crumb rubber concentration has to be higher than that of SBS-polymers in order to obtain a similar performance to that of SBS-polymer modified bitumen; ii) crumb rubber modified binders are more stable than SBS-polymer modified binders in terms of modifier concentration; iii) crumb rubber modified binders exhibited less water sensitivity and similar thermal and ageing susceptibility to SBS-polymer modified binders; iv) linear relationships have been found between modifier concentration and the properties studied for both kind of binders.Los betunes modificados con polvo de neumático (PN) son alternativas ambientalmente sostenibles a los betunes modificados con polímeros. Este artículo compara el comportamiento de ambos tipos de betunes con varios contenidos de modificador. Para ello, se caracterizaron seis betunes mediante ensayos convencionales y método UCL. Este método evalúa propiedades de ligantes para su uso en mezclas bituminosas (cohesión, sensibilidad al agua, térmica y al envejecimiento). Los resultados muestran que i) la concentración de PN ha de ser más elevada que la de polímeros para obtener comportamiento semejante; ii) los betunes con PN son más estables ante cambios en la concentración de modificador que los betunes con polímeros; iii) los betunes con PN presentan menor sensibilidad al agua y equivalente susceptibilidad térmica y al envejecimiento que los betunes con polímeros; iv) se encontraron buenos ajustes lineales entre la concentración de modificadores y la evolución de las propiedades estudiadas
