    Identification of an isoform with an extremely large Cys-rich region of PC6, a Kex2-like processing endoprotease

    AbstractIn the previous study [1993, J. Biochem. (Tokyo) 113, 132-135] we identified PC6, a member of the Kex2 family of processing endoproteases. In this study, we identified another cDNA encoding an isoform of PC6, and designated it as PC6B and redesignated the originally identified PC6 as PC6A. PC6B had a very large Cys-rich region consisting of 22-times repeats of a Cys-rich motif, and a putative transmembrane domain which is not present in PC6A. A PC6B transcript was found mainly in the intestine, while PC6A transcripts were in various tissues. These results suggest distinct roles of PC6A and PC6B in endoproteolytic processing of precursor proteins

    Purpose The objective of this article is to evaluate functional and radiological outcomes of vascularized bone grafts for stage 2 and 3 Kienböck disease. The outcomes of three different donor sites via dorsal approach of the wrist were compared. Pearls and pitfalls in surgical technique were discussed. Methods There were 28 patients who underwent vascularized bone grafts, including the extensor fourth and fifth compartmental artery graft of distal radius in 8 patients, the first and second supraretinacular intercompartmental artery graft of distal radius in 12 patients, and the second dorsal metacarpal neck graft in 8 patients. Average age was 32 years, and radiological grading according to Lichtman classification was stage 2 in 8 patients, stage 3A in 10 patients, and stage 3B in 10 patients. Temporary pinning fixing the midcarpal joint was conducted for 10 weeks postoperatively. Results Follow-up periods averaged 70 months. Pain reduced in 27 patients, and visual analog scale for pain of pre- and postoperative level averaged 59 and 18. Range of wrist flexion and extension motion improved from 87 to 117 degrees, and average grip strength improved from 21 kg preoperatively to 33 kg postoperatively. Carpal height ratio had almost no change from 0.52 to 0.53. Fragmentation of necrotic bone healed in 7 of the 14 cases. Comparative analyses of functional and radiological outcomes between three donor sites found no significant difference. Conclusion Three different vascularized bone grafts from the dorsal wrist and hand area demonstrated favorable and comparable functional outcomes.     Brain-derived Neurotrophic Factor Regulates Energy Expenditure Through the Central Nervous System in Obese Diabetic Mice

    It has been previously demonstrated that brain-derived neurotrophic factor (BDNF) regulates glucose metabolism and energy expenditure in rodent diabetic models such as C57BL/KsJ-leprdb/leprdb (db/db) mice. Central administration of BDNF has been found to reduce blood glucose in db/db mice, suggesting that BDNF acts through the central nervous system. In the present study we have expanded these investigations to explore the effect of central administration of BDNF on energy metabolism. Intracerebroventricular administration of BDNF lowered blood glucose and increased pancreatic insulin content of db/db mice compared with vehicle-treated pellet pair-fed db/db mice. While body temperatures of the pellet pair-fed db/db mice given vehicle were reduced because of restricted food supply in this pair-feeding condition, BDNF treatment remarkably alleviated the reduction of body temperature suggesting the enhancement of thermogenesis. BDNF enhanced norepinephrine turnover and increased uncoupling protein-1 mRNA expression in the interscapular brown adipose tissue. Our evidence indicates that BDNF activates the sympathetic nervous system via the central nervous system and regulates energy expenditure in obese diabetic animals

    Superconductivity protected by spin-valley locking in ion-gated MoS2

    Symmetry-breaking has been known to play a key role in noncentrosymmetric superconductors with strong spin-orbit-interaction (SOI). The studies, however, have been so far mainly focused on a particular type of SOI, known as Rashba SOI, whereby the electron spin is locked to its momentum at a right-angle, thereby leading to an in-planar helical spin texture. Here we discuss electric-field-induced superconductivity in molybdenum disulphide (MoS2), which exhibits a fundamentally different type of intrinsic SOI manifested by an out-of-plane Zeeman-type spin polarization of energy valleys. We find an upper critical field of approximately 52 T at 1.5 K, which indicates an enhancement of the Pauli limit by a factor of four as compared to that in centrosymmetric conventional superconductors. Using realistic tight-binding calculations, we reveal that this unusual behaviour is due to an inter-valley pairing that is symmetrically protected by Zeeman-type spin-valley locking against external magnetic fields. Our study sheds a new light on the interplay of inversion asymmetry with SOI in confined geometries, and its unprecedented role in superconductivity.Comment: 37 pages, 11 figures, http://meetings.aps.org/Meeting/MAR15/Session/G11.1

    Self-medication is the mostly effort to coped health complaint, therefore to drug consume decrease unsuitable, it needed safety socialization to self-medication. The study was take to analyze the effect of drug counseling to self-medication increase in Banyumas regency. In addition to analyze the method effectiveness used in drug counseling to self-medication act. The instrument research used questionnaires in this study and purposive sampling used to took the sample. The respondents were 192 residents whom live in Banyumas regency. The type of research quasi-experimental with non-equivalent control group pretest and posttest research design. The data then analyzed by dependent-sampel t-test to find the effect of counseling to self-medication behavior in α = 0.05. While to find the effectiveness of between discussion group and lecture method by leaflet used independen-sampel t-test in α = 0.05. The results showed there significant different between before and after drug counseling by discussion goup and lecture method used leaflet media, able to self-medication attitude improved such as knowledge and attitude about self-medication (P= 0.000), also it can to know both of methods as effective as to self-medication attitude which showed P=0.05.     Interstitial lung disease in gefitinib-treated Japanese patients with non-small cell lung cancer – a retrospective analysis: JMTO LC03-02

    <p>Abstract</p> <p>Background</p> <p>In Japan, high incidences of interstitial lung disease (ILD) and ILD-related deaths have been reported among gefitinib-treated patients with non-small cell lung cancer (NSCLC). We investigated the efficacy of gefitinib, the incidence of ILD and risk factors for ILD in these patients.</p> <p>Findings</p> <p>We obtained patient data retrospectively using questionnaires sent to 22 institutions. We asked for demographic and clinical data on NSCLC patients for whom gefitinib treatment had begun between July 2002 and February 2003. Data from a total of 526 patients were analyzed. The patient characteristics were as follows: 64% male, 69% with adenocarcinoma, 61% with a performance score of 0–1, and 5% with concurrent interstitial pneumonitis. The objective response proportion was 80/439 (18.2%; 95% CI: 14.7–22.0). ILD developed in 17 patients (3.2%; 95% CI 1.9–5.1%), of whom 7 died. According to multivariate analysis, female sex, history of prior chemotherapy, low absolute neutrophil count before gefitinib treatment, and adenocarcinoma histology were associated with response to gefitinib treatment. None of the factors we evaluated were associated with the development of ILD.</p> <p>Conclusion</p> <p>The results of this study are consistent with previously published values for treatment response proportions and incidence of ILD during gefitinib treatment in Japanese patients. Future studies should be aimed at identifying factors indicating that a patient has a high probability of receiving benefit from gefitinib and a low risk of developing ILD.</p
