Chloroplast microsatellites: measures of genetic diversity and the effect of homoplasy
Chloroplast microsatellites have been widely used in population genetic
studies of conifers in recent years. However, their haplotype configurations
suggest that they could have high levels of homoplasy, thus limiting the power
of these molecular markers. A coalescent-based computer simulation was used to
explore the influence of homoplasy on measures of genetic diversity based on
chloroplast microsatellites. The conditions of the simulation were defined to
fit isolated populations originating from the colonization of one single
haplotype into an area left available after a glacial retreat. Simulated data
were compared with empirical data available from the literature for a species
of Pinus that has expanded north after the Last Glacial Maximum. In the
evaluation of genetic diversity, homoplasy was found to have little influence
on Nei's unbiased haplotype diversity (H(E)) while Goldstein's genetic distance
estimates (D2sh) were much more affected. The effect of the number of
chloroplast microsatellite loci for evaluation of genetic diversity is also
discussed

Egocentric Social Network Structure, Health, and Pro-Social Behaviors in a National Panel Study of Americans
Using a population-based, panel survey, we study how egocentric social networks change over time, and the relationship between egocentric network properties and health and pro-social behaviors. We find that the number of prosocial activities is strongly positively associated with having more friends, or an increase in degree, with approximately 0.04 more prosocial behaviors expected for every friend added. Moreover, having more friends is associated with an improvement in health, while being healthy and prosocial is associated with closer relationships. Specifically, a unit increase in health is associated with an expected 0.45 percentage-point increase in average closeness, while adding a prosocial activity is associated with a 0.46 percentage-point increase in the closeness of one’s relationships. Furthermore, a tradeoff between degree and closeness of social contacts was observed. As the number of close social contacts increases by one, the estimated average closeness of each individual contact decreases by approximately three percentage-points. The increased awareness of the importance of spillover effects in health and health care makes the ascertainment of egocentric social networks a valuable complement to investigations of the relationship between socioeconomic factors and health
Testing for Network and Spatial Autocorrelation
Testing for dependence has been a well-established component of spatial
statistical analyses for decades. In particular, several popular test
statistics have desirable properties for testing for the presence of spatial
autocorrelation in continuous variables. In this paper we propose two
contributions to the literature on tests for autocorrelation. First, we propose
a new test for autocorrelation in categorical variables. While some methods
currently exist for assessing spatial autocorrelation in categorical variables,
the most popular method is unwieldy, somewhat ad hoc, and fails to provide
grounds for a single omnibus test. Second, we discuss the importance of testing
for autocorrelation in data sampled from the nodes of a network, motivated by
social network applications. We demonstrate that our proposed statistic for
categorical variables can both be used in the spatial and network setting
NLRP3 inflammasome activation by mycobacterial ESAT-6 and dsRNA in intraocular tuberculosis
The molecular basis of intraocular tuberculosis (TB) is not well understood. In this study, we investigated the role of two constituents of viable Mycobacterium tuberculosis - Early Secreted Antigenic Target-6 (ESAT-6), and mycobacterial RNA- in inflammasome activation in the retinal pigment epithelium (RPE) - a key site of inflammation in intraocular TB. We found that ESAT-6 induced caspase-1 activation and inflammasome priming in mouse RPE cells, significantly more in wild-type than in Tlr2/3/4/7/9-/-, Myd88-/- and Nlrp3-/- RPE cells. Sub-retinal ESAT-6 injection resulted in greater RPE degeneration in wild-type than Nlrp3-/- mice. In human ocular TB tissue sections, NLRP3 staining was noted in retina as well as RPE. Mycobacterial RNA, specifically its double stranded component, also induced caspase-1 activation, and the double stranded RNA was immunolocalized to human ocular TB sections. Our observations suggest that inflammasome activation in RPE by viable M. tuberculosis could potentially contribute to human intraocular TB
Born to lead? A twin design and genetic association study of leadership role occupancy
We address leadership emergence and the possibility that there is a partially innate predisposition to occupy a leadership role. Employing twin design methods on data from the National Longitudinal Study of Adolescent Health, we estimate the heritability of leadership role occupancy at 24%. Twin studies do not point to specific genes or neurological processes that might be involved. We therefore also conduct association analysis on the available genetic markers. The results show that leadership role occupancy is associated with rs4950, a single nucleotide polymorphism (SNP) residing on a neuronal acetylcholine receptor gene (CHRNB3). We replicate this family-based genetic association result on an independent sample in the Framingham Heart Study. This is the first study to identify a specific genotype associated with the tendency to occupy a leadership position. The results suggest that what determines whether an individual occupies a leadership position is the complex product of genetic and environmental influences, with a particular role for rs4950
A new parametric equation of state and quark stars
It is still a matter of debate to understand the equation of state of cold
supra-nuclear matter in compact stars because of unknown on-perturbative strong
interaction between quarks. Nevertheless, it is speculated from an
astrophysical view point that quark clusters could form in cold quark matter
due to strong coupling at realistic baryon densities. Although it is hard to
calculate this conjectured matter from first principles, one can expect the
inter-cluster interaction to share some general features to nucleon-nucleon
interaction. We adopt a two-Gaussian component soft-core potential with these
general features and show that quark clusters can form stable simple cubic
crystal structure if we assume Gaussian form wave function. With this
parameterizing, Tolman-Oppenheimer-Volkoff equation is solved with reasonable
constrained parameter space to give mass-radius relation of crystalline solid
quark star. With baryon densities truncated at 2 times nuclear density at
surface and range of interaction fixed at 2fm we can reproduce similar
mass-radius relation to that obtained with bag model equations of state. The
maximum mass ranges from about 0.5 to 3 solar mass. Observed maximum pulsar
mass (about 2 solar mass) is then used to constrain parameters of this simple

Infectious Disease Modeling of Social Contagion in Networks
Many behavioral phenomena have been found to spread interpersonally through social networks, in a manner similar to infectious diseases. An important difference between social contagion and traditional infectious diseases, however, is that behavioral phenomena can be acquired by non-social mechanisms as well as through social transmission. We introduce a novel theoretical framework for studying these phenomena (the SISa model) by adapting a classic disease model to include the possibility for ‘automatic’ (or ‘spontaneous’) non-social infection. We provide an example of the use of this framework by examining the spread of obesity in the Framingham Heart Study Network. The interaction assumptions of the model are validated using longitudinal network transmission data. We find that the current rate of becoming obese is 2 per year and increases by 0.5 percentage points for each obese social contact. The rate of recovering from obesity is 4 per year, and does not depend on the number of non-obese contacts. The model predicts a long-term obesity prevalence of approximately 42, and can be used to evaluate the effect of different interventions on steady-state obesity. Model predictions quantitatively reproduce the actual historical time course for the prevalence of obesity. We find that since the 1970s, the rate of recovery from obesity has remained relatively constant, while the rates of both spontaneous infection and transmission have steadily increased over time. This suggests that the obesity epidemic may be driven by increasing rates of becoming obese, both spontaneously and transmissively, rather than by decreasing rates of losing weight. A key feature of the SISa model is its ability to characterize the relative importance of social transmission by quantitatively comparing rates of spontaneous versus contagious infection. Monoubiquitination of syntaxin 3 leads to retrieval from the basolateral plasma membrane and facilitates cargo recruitment to exosomes
Syntaxin 3 (Stx3), a SNARE protein located and functioning at the apical plasma membrane of epithelial cells, is required for epithelial polarity. A fraction of Stx3 is localized to late endosomes/lysosomes, although how it traffics there and its function in these organelles is unknown. Here we report that Stx3 undergoes monoubiquitination in a conserved polybasic domain. Stx3 present at the basolateral—but not the apical—plasma membrane is rapidly endocytosed, targeted to endosomes, internalized into intraluminal vesicles (ILVs), and excreted in exosomes. A nonubiquitinatable mutant of Stx3 (Stx3-5R) fails to enter this pathway and leads to the inability of the apical exosomal cargo protein GPRC5B to enter the ILV/exosomal pathway. This suggests that ubiquitination of Stx3 leads to removal from the basolateral membrane to achieve apical polarity, that Stx3 plays a role in the recruitment of cargo to exosomes, and that the Stx3-5R mutant acts as a dominant-negative inhibitor. Human cytomegalovirus (HCMV) acquires its membrane in an intracellular compartment and we show that Stx3-5R strongly reduces the number of excreted infectious viral particles. Altogether these results suggest that Stx3 functions in the transport of specific proteins to apical exosomes and that HCMV exploits this pathway for virion excretion
