    Functional analysis of RF2a, a rice transcription factor

    RF2a is a bZIP transcription factor that regulates expression of the promoter of rice tungro bacilliform bad-navirus. RF2a is predicted to include three domains that contribute to its function. The results of transient assays with mutants of RF2a from which one or more domains were removed demonstrated that the acidic domain was essential for the activation of gene expression, although the proline-rich and glutamine-rich domains each played a role in this function. Studies using fusion proteins of different functional domains of RF2a with the 2C7 synthetic zinc finger DNA-binding domain showed that the acidic region is a relatively strong activation domain, the function of which is dependent on the context in which the domain is placed. Data from transgenic plants further supported the conclusion that the acidic domain was important for maintaining the biological function of RF2a. RF2a and TBP (TATA-binding protein) synergistically activate transcription in vitro (Zhu, Q., Ordiz, M. I., Dabi, T., Beachy, R. N., and Lamb, C. (2002) Plant Cell 14, 795-803). In vitro and in vivo assays showed that RF2a interacts with TBP through the glutamine-rich domain but not the acidic domain. Functional analysis of such interactions indicates that the acidic domain activates transcription through mechanisms other than via the direct recruitment of TBP.Fil: Dai, Shunhong.     Transcription factor RF2a alters expression of the rice tungro bacilliform virus promoter in transgenic tobacco plants

    The promoter from rice tungro bacilliform badnavirus (RTBV) is expressed only in phloem tissues in transgenic rice plants. RF2a, a b-Zip protein from rice, is known to bind to the Box II cis element near the TATA box of the promoter. Here, we report that the full-length RTBV promoter and a truncated fragment E of the promoter, comprising nucleotides -164 to +45, result in phloem-specific expression of β-glucuronidase (GUS) reporter genes in transgenic tobacco plants. When a fusion gene comprising the cauliflower mosaic virus 35S promoter and RF2a cDNA was coexpressed with the GUS reporter genes, GUS activity was increased by 2-20-fold. The increase in GUS activity was positively correlated with the amount of RF2a, and the expression pattern of the RTBV promoter was altered from phloem-specific to constitutive. Constitutive expression of RF2a did not induce morphological changes in the transgenic plants. In contrast, constitutive overexpression of the b-ZIP domain of RF2a had a strong effect on the development of transgenic plants.     Qualitative flood risk assessment for road and railway infrastructures: the experience of the MOVIDA project

    The Po River District Authority promoted the MOVIDA project with the aim to define appropriate methodologies for flood risk assessment and being compliant with the European Floods Directive (Directive 2007/60/EC). A dedicated Open Source Geographic Information System (i.e. QGIS geoprocessing modules) has been developed for mapping the expected damages in all areas at significant risk in the Po District (Northern Italy), considering five categories of exposed elements (population, infrastructures, economic activities, environmental and cultural heritage, and na-tech sites). Focusing on road and railway infrastructures, the methodology proposed within the project adopts information coming from different data sources (Regional Geoportals, Open Street Map, etc.) and allows to qualitatively estimate the potential risk associated with a flood event. Different risk classes (High, Medium, Low and Null) are assigned in relation to roads category (i.e., Highways, Main, Secondary, Service, Other) or railways type (High-Speed train or not), thus considering both the relevance of the infrastructure itself (as well as its topographical characteristics: e.g. tunnel, bridge, etc.) and the magnitude of the expected event (i.e., hazard). The definition of the risk matrix led to the estimation of the lengths of the sections exposed to different risk levels, which is useful to support the definition of potential mitigation measures and support the competent bodies in the organization of the rescue.</p

    Quantifying the effects of nature-based solutions in reducing risks from hydrometeorological hazards: Examples from Europe

    The combination of climate change and social and ecological factors will increase risks societies face from hydrometeorological hazards (HMH). Reducing these risks is typically achieved through the deployment of engineered (or grey) infrastructure but increasingly, nature-based so-lutions (NBS) are being considered. Most risk assessment frameworks do not allow capturing well the role NBS can play in addressing all components of risk, i.e., the hazard characteristics and the exposure and vulnerability of social-ecological systems. Recently, the Vulnerability and Risk as-sessment framework developed to allow the assessment of risks in the context of NBS implemen-tation (VR-NBS framework) was proposed. Here, we carry out the first implementation of this framework using five case study areas in Europe which are exposed to various HMH. Our results show that we can demonstrate the effect NBS have in terms of risk reduction and that this can be achieved by using a flexible library of indicators that allows to capture the specificities of each case study hazard, social and ecological circumstances. The approach appears to be more effec-tive for larger case study areas, but further testing is required in a broader variety of contexts

    Early adversity predicts adoptees’ enduring emotional and behavioral problems in childhood

    Children adopted from the public care system are likely to experience a cluster of inter-related risk factors that place them on a trajectory of mental health problems that persist across the life course. However, the specific effects of putative risk factors on children’s mental health post-placement are not well understood. We conducted a prospective, longitudinal study of children placed for adoption between 2014 and 2015 (N = 96). Adoptive parents completed questionnaires at approximately 5-, 21-, 36-, and 48 months post-placement. We used time series analysis to examine the impact of pre-adoptive risk factors (adverse childhood experiences [ACEs], number of moves, days with birth parents and in care) on children’s internalizing and externalizing problems, and prosocial behaviour over four years post-placement. Adoptees’ internalizing and externalizing problems remained consistently high over the four-year study period but more ACEs predicted increases in internalizing and externalizing problems. Contrary to expectations, more pre-placement moves and time in care predicted fewer problems over time, but exploratory analyses of interactive effects revealed this was only the case in rare circumstances. We identify pre- and post-removal factors that may incur benefits or have a deleterious impact on adoptees’ outcomes in post-adoptive family life. Our findings provide knowledge for front-line professionals in the support of adoptive families and underscore the vital need for effective early intervention

    Quality of life in liver transplant recipients during the Corona virus disease 19 pandemic: A multicentre study

    Background: Liver transplant recipients require specific clinical and psychosocial attention given their frailty. Main aim of the study was to assess the quality of life after liver transplant during the current pandemic. Methods: This multicentre study was conducted in clinically stable, liver transplanted patients. Enrollment opened in June and finished in September 2021. Patients completed a survey including lifestyle data, quality of life (Short Form health survey), sport, employment, diet. To examine the correlations, we calculated Pearson coefficients while to compare subgroups, independent samples t-tests and ANOVAs. To detect the predictors of impaired quality of life, we used multivariable logistic regression analysis. Results: We analysed data from 511 patients observing significant associations between quality of life’s physical score and both age and adherence to Mediterranean diet (p &lt;.01). A significant negative correlation was observed between mental score and the sedentary activity (p &lt;.05). Female patients scored significantly lower than males in physical and mental score. At multivariate analysis, females were 1.65 times more likely to report impaired physical score than males. Occupation and physical activity presented significant positive relation with quality of life. Adherence to Mediterranean diet was another relevant predictor. Regarding mental score, female patients were 1.78 times more likely to show impaired mental score in comparison with males. Sedentary activity and adherence to Mediterranean diet were further noteworthy predictors. Conclusions: Females and subjects with sedentary lifestyle or work inactive seem to show the worst quality of life and both physical activity and Mediterranean diet might be helpful to improve it

    Long Term Running Biphasically Improves Methylglyoxal-Related Metabolism, Redox Homeostasis and Neurotrophic Support within Adult Mouse Brain Cortex

    Oxidative stress and neurotrophic support decline seem to be crucially involved in brain aging. Emerging evidences indicate the pro-oxidant methylglyoxal (MG) as a key player in the age-related dicarbonyl stress and molecular damage within the central nervous system. Although exercise promotes the overproduction of reactive oxygen species, habitual exercise may retard cellular aging and reduce the age-dependent cognitive decline through hormetic adaptations, yet molecular mechanisms underlying beneficial effects of exercise are still largely unclear. In particular, whereas adaptive responses induced by exercise initiated in youth have been broadly investigated, the effects of chronic and moderate exercise begun in adult age on biochemical hallmarks of very early senescence in mammal brains have not been extensively studied. This research investigated whether a long-term, forced and moderate running initiated in adult age may affect the interplay between the redox-related profile and the oxidative-/MG-dependent molecular damage patterns in CD1 female mice cortices; as well, we investigated possible exercise-induced effects on the activity of the brain derived neurotrophic factor (BDNF)-dependent pathway. Our findings suggested that after a transient imbalance in almost all parameters investigated, the lately-initiated exercise regimen strongly reduced molecular damage profiles in brains of adult mice, by enhancing activities of the main ROS- and MG-targeting scavenging systems, as well as by preserving the BDNF-dependent signaling through the transition from adult to middle age
