    STABILITY INDICATING METHOD DEVELOPMENT AND VALIDATION OF METFORMIN AND ERTUGLIFLOZIN BY HIGH-PERFORMANCE LIQUID CHROMATOGRAPHY WITH PDA DETECTION AND ITS APPLICATION TO TABLET DOSAGE FORM

    Objective: A sensitive, rapid, accurate, and precise stability indicating high-performance liquid chromatography method was developed and validated for the simultaneous estimation of metformin (MET) and ertugliflozin (ERT). Methods: The planned chromatographic method was developed using Kromasil C18 column using 0.1 M sodium dihydrogen phosphate methanol (50:50, by volume, pH 4.0) as mobile phase system followed by peak area response measurement at 238 nm. The developed method was validated by means of ICH guidelines about system suitability, linearity, sensitivity, selectivity, accuracy, precision, robustness, and specificity. Results: Calibration curves of MET and ERT are linear from 250 to 750 μg/ml and 3.75 to 11.25 μg/ml, respectively. Relative standard deviation is <2.0% and recovery is ̴ 100%. Successfully, the developed method was applied for the simultaneous determination of MET and ERT in tablets and to study degradation of MET and ERT in acidic, alkaline, oxidation, thermal, and photolytic degradation conditions. No obstructions from additional common excipients of tablets and degradants were observed. Conclusion: The results recommended method suitability for the analysis of MET and ERT in quality control laboratories

    A Survey on Secret Key Extraction Using Received Signal Strength in Wireless Networks

    Secure wireless communications typically rely on secret keys, which are difficult to establish in an ad hoc network without a key management infrastructure. The channel reciprocity and spatial decorrelation properties can be used to extract secret key, especially in a Rayleigh fading channel. But the intervention of intermediate objects between the communication nodes reduces the strength of the secret key generated through such methods. Furthermore, the impact of small fluctuations also reduces the bit matching rate of such key agreement methods. This paper is based on the survey conducted on secret key generation from Received Signal Strength (RSS). By consider ing uniqueness property of RSS as base, various authors have proposed different methods for secret key extraction. Due to use of RSS for key extraction the existing systems suffer from predictable filter response at random period. The existing system also faces signal fading and drop in RSS because of intermediate object. By this survey we specify that even after generating high entropy bits for key extraction, there are considerable drawbacks in extracted key du e to intervention of intermediate objects and remarkable fading and drop in RSS

    Murraya Paniculata Mediated Synthesis Of CdS Nanoparticles For Potent Biomedical Applications

    The leaf extract of Murraya paniculata (MPL) is used to create cadmium sulphide nanoparticles, or CdS NPs and the reducing and stabilizing agent is plant extract. A crucial capping agent in nano production is played by phytochemicals. Analytical methods such as XRD and FTIR are used to characterize CdS NPs. The biomedical applications of prepared CdS NPs were examined, including their anticancer, antifungal, and antibacterial properties. The antibacterial performance was carried out with S.flexneri, C.perfringens, S.typhimurium, and E.faecalis which were all susceptible to the antibacterial action of CdS NPs. At 150µg/ml, S.flexneri and E.faecalis showed a maximum zone of inhibition is 20mm. In addition, A.niger and C.albicans were used to test the antifungal activities, results shows the concentration of 400 µg/ml CdS NPs inhibited the growth of A.flavus (16mm). By using the NRU assay, it was found that the biosynthesized CdS NPs exhibited cytotoxic action against the MCF-7 cell line. Analysis using the NRU assay revealed that treating cell lines with increasing concentrations of NPs had lethal effects. The 24-hour treatment's IC50 was found to be 153.2 µg/ml

    Background/Objective: Soft tick in the ear is a very common acute painful and distressing condition in the flowering months of October to March. It’s a common condition in the rubber growing belt of Sullia.The mouthparts of the tick grips firmly the skin of the external auditory canal or the tympanic membrane and sucks blood and swells up. Otoscopy and removal of the tick from the ear can be done in outpatients in adults and difficult in a frightened irritable child.Materials and methods: 312 cases of intra-aural ticks presenting to the opd and casualty which included 131 males and 181 females were included in the study.Results: Out of the 312 cases of intra-aural ticks, 170 cases were treated in the outpatient and 140 cases under short general anesthesia with oto-microscopy.Conclusion: Intra-aural tick infestation is an acute painful condition which needs prompt management by an experienced otolaryngologist. Proper visualization and instrumentation is necessary to avoid complications.

    SELECTIVE CECAL BACTERIAL CHANGES MEDIATE THE ADVERSE EFFECTS ASSOCIATED WITH HIGH PALMOLEIN OR HIGH STARCH DIETS: PROPHYLACTIC ROLE OF FLAX OIL

    Objective: Studies on the dynamics of gut bacteria in relation to metabolic adverse effects induced by high palmolein or high starch diets and in relation to health benefits of uncommon foods are lacking. Our aim was to assess under controlled conditions, the impact of vegetable based palmitic acid rich, high fat diet or a high starch diet on various metabolic parameters in relation to selective gut bacterial alterations in rats and also to see the effect of flaxseed oil supplementation on these parameters.Methods: Wistar Rats were fed for 4 mo either a control diet(CT) or a 30% high fat diet (HF) or HF diet with flax oil supplemented at two different doses (HFF1 and HFF2) or a 78% high starch diet (HC) after which they were sacrificed and analyzed for selective cecal bacteria, hematology, immune function and body composition.Results: High palmolein diet fed rats showed a decrease in colony forming units of lactobacillus, enterococci, streptococci bacteria and an increase in enterobacteriaceae in the cecum unlike HC fed rats. While high palmolein diet was found to impair immunity and increase inflammation, high starch diet affected body composition and lipid profile. Supplementing the flax seed oil ameliorated most of the adverse effects of high palmolein diet.Conclusions: Independent of energy intakes both high palmolein and high starch intakes have differential adverse effects. It can be envisaged that the adverse effects of feeding palmolein are mediated through immune impairment and inflammatory response, which in turn are associated with altered gut bacteria profile; and flax oil was found to have a prophylactic role in controlling these adverse effects. This study emphasizes the need to evaluate immunological as well as bacterial profile while assessing the safety of dietary fats in addition to traditional methods.Â

    CD23 expression in mantle cell lymphoma is associated with CD200 expression, leukemic non-nodal form, and a better prognosis

    Mantle cell lymphoma (MCL) is usually CD23 negative, a feature helpful in distinguishing MCL from chronic lymphocytic leukemia/small lymphocytic lymphoma. However, a subset of MCL cases can be CD23+. Limited data are available regarding the clinicopathological features and prognosis of patients with CD23+ MCL. In this study, we reviewed 798 cases of MCL and identified 103 (13%) that were CD23+ by flow cytometry, all of which were positive for cyclin D1 and/or associated with CCND1/IGH. In all cases of CD23+ MCL, CD23 expression was dim partial or dim, unlike moderate to bright CD23 expression observed in chronic lymphocytic leukemia/small lymphocytic lymphoma. The clinicopathological features and outcome of patients with CD23+ MCL were compared with 240 patients with typical MCL negative for CD23. Patients with CD23+ MCL more often had an elevated leukocyte count (33% versus 18%, P = .009), bone marrow involvement (89% versus 78%, P = .02), stage 4 disease (87% versus 77%, P = .03), and a leukemic presentation (42% versus 11%, P = .0001). CD23+ MCL was also more often positive for CD200 (17% versus. 4.6%, P = .0005) and less commonly positive for SOX11 (55% versus. 74%, P = .027). All other clinicopathological features were similar. With similar treatment regimens and observation times, patients with CD23+ MCL had a significant better overall survival (P = .02) and progression-free survival (P = .029). In conclusion, CD23 expression was observed in 13% of MCL cases and is associated with a better prognosis in patients with MCL. CD23 is associated with leukocytosis, a leukemic presentation, bone marrow involvement, CD200 expression, and a lower frequency of SOX11 positivity

    Mutations in PYCR2, Encoding Pyrroline-5-Carboxylate Reductase 2, Cause Microcephaly and Hypomyelination

    Despite recent advances in understanding the genetic bases of microcephaly, a large number of cases of microcephaly remain unexplained, suggesting that many microcephaly syndromes and associated genes have yet to be identified. Here, we report mutations in PYCR2, which encodes an enzyme in the proline biosynthesis pathway, as the cause of a unique syndrome characterized by postnatal microcephaly, hypomyelination, and reduced cerebral white-matter volume. Linkage mapping and whole-exome sequencing identified homozygous mutations (c.355C>T [p.Arg119Cys] and c.751C>T [p.Arg251Cys]) in PYCR2 in the affected individuals of two consanguineous families. A lymphoblastoid cell line from one affected individual showed a strong reduction in the amount of PYCR2. When mutant cDNAs were transfected into HEK293FT cells, both variant proteins retained normal mitochondrial localization but had lower amounts than the wild-type protein, suggesting that the variant proteins were less stable. A PYCR2-deficient HEK293FT cell line generated by genome editing with the clustered regularly interspaced short palindromic repeat (CRISPR)-Cas9 system showed that PYCR2 loss of function led to decreased mitochondrial membrane potential and increased susceptibility to apoptosis under oxidative stress. Morpholino-based knockdown of a zebrafish PYCR2 ortholog, pycr1b, recapitulated the human microcephaly phenotype, which was rescued by wild-type human PYCR2 mRNA, but not by mutant mRNAs, further supporting the pathogenicity of the identified variants. Hypomyelination and the absence of lax, wrinkly skin distinguishes this condition from that caused by previously reported mutations in the gene encoding PYCR2’s isozyme, PYCR1, suggesting a unique and indispensable role for PYCR2 in the human CNS during development
