Structure of Staphylococcus aureus 1,4-dihydroxy-2-naphthoyl-CoA synthase (MenB) in complex with acetoacetyl-CoA
Vitamin K-2, or menaquinone, is an essential cofactor for many organisms and the enzymes involved in its biosynthesis are potential antimicrobial drug targets. One of these enzymes, 1,4-dihydroxy-2-naphthoyl-CoA synthase (MenB) from the pathogen Staphylococcus aureus, has been obtained in recombinant form and its quaternary structure has been analyzed in solution. Cubic crystals of the enzyme allowed a low-resolution structure (2.9 angstrom) to be determined. The asymmetric unit consists of two subunits and a crystallographic threefold axis of symmetry generates a hexamer consistent with size-exclusion chromatography. Analytical ultracentrifugation indicates the presence of six states in solution, monomeric through to hexameric, with the dimer noted as being particularly stable. MenB displays the crotonase-family fold with distinct N- and C-terminal domains and a flexible segment of structure around the active site. The smaller C-terminal domain plays an important role in oligomerization and also in substrate binding. The presence of acetoacetyl-CoA in one of the two active sites present in the asymmetric unit indicates how part of the substrate binds and facilitates comparisons with the structure of Mycobacterium tuberculosis MenB.</p

High surface area bio-waste based carbon as a superior electrode for vanadium redox flow battery
Activated carbon (AC) with high surface area (1901 m2 g�1) is synthesized from low cost bio-waste orange
(Citrus sinensis) peel for vanadium redox flow battery (VRB). The composition, structure and
electrochemical properties of orange peel derived AC (OP-AC) are characterized by elemental analyzer,
field emission-scanning electron microscopy, X-ray diffraction, Raman spectroscopy, X-ray photoelectron
spectroscopy, cyclic voltammetry (CV), and electrochemical impedance spectroscopy. CV results show
that OP-AC coated bipolar plate demonstrates improved electro-catalytic activity in both positive and
negative side redox couples than the pristine bipolar plate electrode and this is ascribed to the high
surface area of OP-AC which provides effective electrode area and better contact between the porous
electrode and bipolar plate. Consequently, the performance of VRB in a static cell shows higher energy
efficiency for OP-AC electrode than the pristine electrode at all current densities tested. The results
suggest the OP-AC to be a promising electrode for VRB applications and can be incorporated into making
conducting plastics electrode to lower the VRB cell stack weight and cost

Synthesis and Structural Characterization of Novel Dimers of Dipyridothiazine as Promising Antiproliferative Agents
Many new isomeric dipyridothiazine dimers have been presented as molecules with anticancer potential. These compounds were obtained in efficient syntheses of 1,6-, 1,8-, 2,7- and 3,6-diazaphenothiazines with selected alkylaromatic linkers. The structures of these compounds has been proven with two-dimensional spectroscopic techniques (COSY, NOESY, HSQC and HMBC) and high-resolution mass spectrometry (HRMS). In silico analyses of probable molecular targets were performed using the Way2Drug server. All new dimers were tested for anticancer activity against breast cancer line MCF7 and colon cancer line SW480. Cytotoxicity was assessed on normal L6 muscle cells. The tested dimers had high anticancer potential expressed as IC50 and the selectivity index SI. The most active derivative, 4c, showed an IC50 activity of less than 1 µM and an SI selectivity index higher than 100. Moreover, the compounds were characterized by low toxicity towards normal cells, simultaneously indicating a high cytostatic potential
