    New observations on the integrity, structure and physiology of flesh cells from fully ripened grape berry

    The physiological/structural status of the soft ripened berry is still a matter of debate. In this paper isolated mesocarp cells from ripened berries of both wine and table varieties were studied by bright-field, fluorescence and confocal microscopy and flow cytometry to highlight the organization of berry flesh cell, function and viability. Flow cytometry analysis confirmed that protoplasting from grape berry mesocarp tissue yields a single heterogenous population of intact and viable cells. Also, the integrity of the plasma membrane and the architecture and complexity of the intracellular membranous system were shown by FM1-43 staining coupled to confocal microscopy imaging. The observed incorporation of the fluorescent glucose analogue 2-NBDG suggests that endocytosis is involved in the transport and intracellular compartmentation of apoplastic sugars. Neutral Red staining confirmed the intricate organization, size, diversity and integrity of the vacuolar apparatus that is probably related to the multifaceted roles of the vacuoles in the developing fruit.This work was supported in part by the Fundação para a Ciência e a Tecnologia (research projects PTDC/AGR-ALI/100636/2008 and PTDC/BIA-BCM/69448/2006 and grant no.     Artroscopia do Tornozelo Anterior vs Posterior. Comparação de Resultados e Complicações

    Introdução: Recentemente têm surgido debates na literatura internacional acerca da segurança e necessidade da abordagem artroscópica posterior para tratamento da patologia intra e extra articular do tornozelo. A artroscopia por via posterior realiza-se utilizando portais postero-interno e postero-externo, e com o doente em decúbito ventral. Permite acesso à região posterior do tornozelo, articulação subtalar, osso trígono, tendões peroneias e tendão do Longo flexor do Hallux, e ainda à porção posterior do ligamento deltoideu. Permite ainda uma melhor visualização, menor morbilidade e recuperação mais rápida que na abordagem a céu aberto. Neste estudo comparamos os resultados clínicos e funcionais e dos doentes submetidos a apenas a artroscopia anterior e dos doentes submetidos a artroscopia anterior e posterior do tornozelo, bem como as complicações independentemente da patologia inicial. Material e Métodos: Estudo retrospectivo, tendo sido analisados os processos clínicos de todos os doentes submetidos a tratamento artroscópico do tornozelo em duas instituições, pelo mesmo cirurgião. De um universo de 299 procedimentos artroscópicos do tornozelo, excluímos todos os casos em que foi utilizada concomitantemente uma via aberta para tratamento de outras lesões. Obtivémos resultados clínicos e funcionais de 185 doentes, 97 homens, 88 mulheres, tendo sido aplicado o score Aofas para o retropé e tornozelo. Foi realizada artroscopia posterior em 87 doentes e anterior em 98 doentes. A média de idades foi de 36 anos (17‐59). O follow-up mínimo foi de 6 meses (6‐60). Cento e trinta e dois doentes apresentavam patologia de origem traumática, e destes, 105 estavam relacionados com acidentes de trabalho. Resultados: Artroscopia anterior: a média do score Aofas foi de 87(43‐ 100). Oitenta e sete doentes retomaram a sua atividade profissional ou desportiva previa, e 16 doentes ainda não retomaram atividade na altura da avaliação. O tempo médio de retorno à atividade previa foi de 4 meses (2‐10). Registamos complicações em 13 doentes. Estas incluem infecção superficial das postas de entrada (3 casos), síndrome de dor regional complexa (3 casos), artrofibrose (2 casos), recidiva da lesão inicial (5 casos). Foram reoperados 6 doentes (2 casos de artrofibrose e 4 por recidiva da patologia inicial)Artroscopia anterior e posterior: a média do score Aofas foi de 83 (38‐100). Setenta e cinco doentes já retomaram a sua atividade prévia, sendo que 10 ainda se encontram em tratamento. O tempo médio de retorno à atividade foi de 4,3 meses (2‐ 12). Registámos complicações em 10 doentes (11,4%). Estas incluem 4 casos de lesão do nervo peroneal superficial, dos quais 3 recuperaram totalmente, 3 casos de artrofibrose, 2 casos de síndrome de dor regional complexa e 1 caso de infecção superficial das portas de entrada. Foram reoperados 4 doentes, dos quais 2 por artrofibrose, 2 casos por recidiva da patologia inicial. Discussão: Constatamos que a média do score Aofas e a taxa de complicações é sobreponível entre as duas diferentes abordagens. Os nossos resultados, em termos de complicações, são ligeiramente inferiores aos publicados na literatura internacional, quer para a artroscopia anterior, quer para a anterior e posterior, o 7 que poderá estar relacionado com o elevado número de doentes com lesões relacionadas com acidentes de trabalho. Conclusão: A abordagem posterior não tem mais complicações que a abordagem anterior isolada. Os resultados em doentes vítimas de acidente de trabalho são inferiores em ambas as abordagens. Os resultados e complicações de ambas as abordagens são sobreponíveis aos da literatura internacional

    A luxação palmar isolada da quinta articulação carpo-metacárpica é rara. Esta lesão pode passar despercebida, sendo importante a suspeita clínica, levando a uma avaliação radiológica no perfil e incidências obliquas. O tratamento é controverso, estando descritas a redução fechada com fixação percutânea com fios de Kirshner percutâneos e a redução aberta com fixação interna. Apresentamos um caso clínico de luxação palmar do tipo cubital isolada e o seu tratamento. O resultado funcional aos 6 meses, de acordo com o score DASH, foi de 0.8. Grounded on some of the most prominent adoption models for technological innovations, we developed a conceptual model especially suited for BAI. Based on this we propose an instrument in which relevant hypotheses will be derived and tested by means of statistical analysis. We hope that the findings derived from our analysis may offer important insights for practitioners and researchers regarding the drivers that lead to BAI adoption in firms.     Reduction of volatile acidity of wines by selected yeast strains

    Herein we isolate and characterize wine yeasts with ability to reduce volatile acidity of wines using a refermentation process, which consists in mixing the acidic wine with freshly crushed grapes or musts or, alternatively, in the incubation with the residual marc. From a set of 135 yeast isolates, four strains revealed ability to use glucose and acetic acid simultaneously. Three of them were identified as Saccharomyces cerevisiae and one as Lachancea thermotolerans. Among nine commercial S. cerevisiae strains, strains S26, S29 and S30 display similar glucose and acetic acid initial simultaneous consumption pattern and were assessed in refermentation assays. In a medium containing an acidic wine with high glucose/low ethanol concentrations, under low oxygen availability, strain S29 is the most efficient one, whereas L. thermotolerans 44C is able to decrease significantly acetic acid similar to the control strain Zygosaccharomyces bailii ISA 1307, but only under aerobic conditions. Conversely, for low glucose/high ethanol concentrations, under aerobic conditions, S26 is the most efficient acid degrading strain, while under limited-aerobic conditions, all the S. cerevisiae strains studied display acetic acid degradation efficiencies identical to Z. bailii. Moreover, S26 strain also reveals capacity to decrease volatile acidity of wines.     Empirical Investigation of Data Analytics Capability and Organizational Flexibility as Complements to Supply Chain Resilience

    Supply chain resilience and data analytics capability have generated increased interest in academia and among practitioners. However, existing studies often treat these two streams of literature independently. Our study model reconciles two different streams of literature: data analytics capability as a means to improve information-processing capacity and supply chain resilience as a means to reduce a ripple effect in supply chain or quickly recover after disruptions in the supply chain. We have grounded our theoretical model in the organisational information processing theory (OIPT). Four research hypotheses are tested using responses from 213 Indian manufacturing organisations collected via a pre-tested survey-based instrument. We further test our model using variance-based structural equation modelling, popularly known as PLS-SEM. All of the hypotheses were supported. The findings of our study offer a unique contribution to information systems (IS) and operations management (OM) literature. The findings further provide numerous directions to the supply chain managers. Finally, we note our study limitations and provide further research directions

    The fate of acetic acid during glucose co-metabolism by the spoilage yeast Zygosaccharomyces bailii

    Zygosaccharomyces bailii is one of the most widely represented spoilage yeast species, being able to metabolise acetic acid in the presence of glucose. To clarify whether simultaneous utilisation of the two substrates affects growth efficiency, we examined growth in single- and mixed-substrate cultures with glucose and acetic acid. Our findings indicate that the biomass yield in the first phase of growth is the result of the weighted sum of the respective biomass yields on single-substrate medium, supporting the conclusion that biomass yield on each substrate is not affected by the presence of the other at pH 3.0 and 5.0, at least for the substrate concentrations examined. In vivo(13)C-NMR spectroscopy studies showed that the gluconeogenic pathway is not operational and that [2-(13)C]acetate is metabolised via the Krebs cycle leading to the production of glutamate labelled on C(2), C(3) and C(4). The incorporation of [U-(14)C]acetate in the cellular constituents resulted mainly in the labelling of the protein and lipid pools 51.5% and 31.5%, respectively. Overall, our data establish that glucose is metabolised primarily through the glycolytic pathway, and acetic acid is used as an additional source of acetyl-CoA both for lipid synthesis and the Krebs cycle. This study provides useful clues for the design of new strategies aimed at overcoming yeast spoilage in acidic, sugar-containing food environments.     Irf4 is a positional and functional candidate gene for the control of serum IgM levels in the mouse

    Natural IgM are involved in numerous immunological functions but the genetic factors that control the homeostasis of its secretion and upholding remain unknown. Prompted by the finding that C57BL/6 mice had significantly lower serum levels of IgM when compared with BALB/c mice, we performed a genome-wide screen and found that the level of serum IgM was controlled by a QTL on chromosome 13 reaching the highest level of association at marker D13Mit266 (LOD score¼3.54). This locus was named IgMSC1 and covered a region encompassing the interferon-regulatory factor 4 gene (Irf4). The number of splenic mature B cells in C57BL/6 did not differ from BALB/c mice but we found that low serum levels of IgM in C57BL/6 mice correlated with lower frequency of IgM-secreting cells in the spleen and in the peritoneal cavity. These results suggested that C57BL/6 mice have lower efficiency in late B-cell maturation, a process that is highly impaired in Irf4 knockout mice. In fact, we also found reduced Irf4 gene expression in B cells of C57BL/6 mice. Thus, we propose Irf4 as a candidate for the IgMSC1 locus, which controls IgM homeostatic levels at the level of B-cell terminal differentiation

    HIV-1-Transmitted Drug Resistance and Transmission Clusters in Newly Diagnosed Patients in Portugal Between 2014 and 2019

    Objective: To describe and analyze transmitted drug resistance (TDR) between 2014 and 2019 in newly infected patients with HIV-1 in Portugal and to characterize its transmission networks. Methods: Clinical, socioepidemiological, and risk behavior data were collected from 820 newly diagnosed patients in Portugal between September 2014 and December 2019. The sequences obtained from drug resistance testing were used for subtyping, TDR determination, and transmission cluster (TC) analyses. Results: In Portugal, the overall prevalence of TDR between 2014 and 2019 was 11.0%. TDR presented a decreasing trend from 16.7% in 2014 to 9.2% in 2016 (p for-trend = 0.114). Multivariate analysis indicated that TDR was significantly associated with transmission route (MSM presented a lower probability of presenting TDR when compared to heterosexual contact) and with subtype (subtype C presented significantly more TDR when compared to subtype B). TC analysis corroborated that the heterosexual risk group presented a higher proportion of TDR in TCs when compared to MSMs. Among subtype A1, TDR reached 16.6% in heterosexuals, followed by 14.2% in patients infected with subtype B and 9.4% in patients infected with subtype G. Conclusion: Our molecular epidemiology approach indicates that the HIV-1 epidemic in Portugal is changing among risk group populations, with heterosexuals showing increasing levels of HIV-1 transmission and TDR.
