    Molecular Identification Of Cultivable Bacteria From Infected Root Canals Associated With Acute Apical Abscess

    The objective of this study was to investigate the bacterial composition present in root canals of teeth associated with acute apical abscess by molecular identification (16S rRNA) of cultivable bacteria. Two hundred and twenty strains isolated by culture from 20 root canals were subjected to DNA extraction and amplification of the 16S rRNA gene (PCR), followed by sequencing. The resulting nucleotide sequences were compared to the GenBank database from the National Center of Biotechnology Information through BLAST. Strains not identified by sequencing were submitted to clonal analysis. The association of microbiological findings with clinical features and the association between microbial species were also investigated. Fifty-nine different cultivable bacteria were identified by 16S rRNA gene sequencing, belonging to 6 phyla, with an average number of 6 species per root canal. Molecular approaches allowed identification of 99% of isolates. The most frequently identified bacteria were Prevotella spp., Pseudoramibacter alactolyticus, Parvimonas micra, Dialister invisus, Filifactor alocis, and Peptostreptococcus stomatis. Positive association was found between Prevotella buccae and Pseudoramibacter alactolyticus and between Parvimonas micra and Prevotella nigrescens (both p<0.05). It was concluded that the microbiota of infected root canals associated with acute apical abscess is diverse and heterogeneous, composed mainly of anaerobic Gram-negative bacteria, with the great majority belonging to the phyla Firmicutes and Bacteroidetes.27331832

    Evaluation of water and sucrose diffusion coefficients during osmotic dehydration of jenipapo (Genipa americana L.).

    The apparent diffusion coefficients for sucrose and water during osmotic dehydration of jenipapos were determined. Long time experiments (up to 60 h) were carried out in order to determine equilibrium concentrations inside jenipapos, whereas short time experiments (up to 4 h) were performed to provide detailed information on kinetics of water loss and solids gain at the beginning of osmotic treatment. According to the result, mass transfer rates for water and solutes, as well as the apparent diffusion coefficients for sucrose showed to be dependent on sucrose concentration in osmotic solution. The immersion time did not have significant effect (p>0.05) over the diffusion coefficients for sucrose and water

    It is here discussed the development of a low cost analytical instrument with capacity for metals determination using atomic emission measurements in an electrothermal atomization system with a tungsten coil atomizer. The main goal was to show a new frontier for using this atomizer and to demonstrate that the simple instrumental arrangement here proposed has potential for portability and for solving analytical tasks related to metals determination. Atomic emission of calcium was selected for the adjustment of instrumental parameters and to evaluate the main characteristics of the lab-built instrument. With increasing computer power and more efficient simulational techniques, this belief turned to a stretched exponential behaviour, instead. Cepas não identificadas pelo sequenciamento foram submetidas à clonagem. Associação de achados microbiológicos e características clínicas e associação entre espécies bacterianas também foram investigadas. Cinquenta e nove bactérias cultiváveis diferentes foram identificadas pelo sequenciamento do gene 16S rRNA, pertencentes a 6 filos, numa média de 6 espécies por canal. Método molecular permitiu identificação de 99% das cepas isoladas. As bactérias mais frequentes foram Prevotella spp., Pseudoramibacter alactolyticus, Parvimonas micra, Dialister invisus, Filifactor alocis, Peptostreptococcus stomatis. Associação positiva foi encontrada entre Prevotella buccae e Pseudoramibacter alactolyticus, e entre Parvimonas micra e Prevotella nigrescens (p<0,05).     Atributos químicos de um Latossolo Amarelo sob diferentes sistemas de manejo.

    O objetivo deste trabalho foi avaliar os atributos químicos de um Latossolo Amarelo sob diferentes sistemas de manejo do solo, no cerrado piauiense. Foram avaliados quatro sistemas de manejo: plantio convencional por três anos; plantio direto por três anos (PD3) e cinco anos (PD5), com uso de milheto como cultivo de cobertura; e plantio direto por nove anos (PD9), dos quais sete com uso de milheto e dois com forrageira. Utilizou-se área de cerrado nativo como referência. As amostras do solo foram coletadas em períodos chuvosos e secos, nas camadas 0,00?0,05, 0,05?0,10, 0,10?0,20 e 0,20?0,40 m, para determinação de pH, Al3+, H+Al, Ca2+, Mg2+, K+, P disponível, carbono orgânico total (COT), soma de bases (SB), capacidade de troca de cátions efetiva (t) e potencial (T), e saturação por bases (V) e por alumínio (m%). O sistema PD9 apresentou maiores valores de pH e menores de Al3+, H+Al e m%. Observaram-se maiores valores de Ca2+, K+, SB, t, T, V e P sob PD5 e PD9, até 0,20 m. Os maiores valores de COT foram verificados sob PD5 e PD9, exceto na camada de 0,00?0,05 m. O acúmulo de material orgânico associado ao uso de forrageira em PD favorece o aumento dos teores de COT nas camadas mais profundas do solo, no período seco
