    Assimilation Between Two Generations of Chicano Immigrants in the United States in a Better Life Movie

    Dalam masyarakat majemuk pasti ada keberagaman budaya. Namun untuk hidup dan bertahan di dalamnya bukanlah hal yang mudah. Mereka yang termasuk dalam kelompok minoritas dituntut untuk mengadopsi budaya-budaya yang dimiliki kelompok mayoritas. Proses ini disebut dengan asimilasi. Hal ini dilakukandengan tujuan agar mereka dapat diterima dan mendapat perlakuan dan kesempatan yang sama dengan kelompok mayoritas. Sebagian dari mereka dengan sukarela mengasimilasi budaya-budaya baru dan sebagian lagi sebaliknya. Mereka tetap berpegang pada budaya mereka dan enggan untuk mengadopsi budaya dari kelompok mayoritas seperti yang terjadi pada sebagian besar etnik Chicano. Namun, setiap orang memiliki tingkatan yang berbeda dalam mengadopsi budaya tersebut. Proses asimilasi tersebutdapat direfleksikan dalam karya sastra, salah satunya melalui sebuah film. A Better Life adalah sebuah film yang mencoba untuk mendeskripsikan tentang hidup seorang ilegal imigran asal Meksikodan seorang anaknya di tengah- tengah masyarakat Amerika Serikat. Di dalam film ini,digambarkan bagaimana perbedaan asimilasi yang dialami oleh dua generasi yang berbeda. Tujuan penulisan skripsi ini adalah untuk mengetahui adanya perbedaan tingkatan dalam proses asimilasi yang dialami oleh dua generasiChicano yang berbeda. Metode yang digunakan adalah penelitian pustaka dengan pendekatan sosilogi. Penulis mengaplikasikan teori asimilasi dalam menganalisis film. Hasil analisis menunjukkan bahwa secara umum, generasi kedua lebih terasimilasi dari pada generasi pertamaseperti yang digambarkan dalam film A Better Life. Terdapat perbedaan tingkatan yang tipis di dalam beberapa sub proses asimilasi yang dialami generasi pertama dan kedua. Perbedaan tersebut terjadi pada sub proses asimilasi budaya dan asimilasi struktural sekunder yang mengacu pada pencapaian dalam pekerjaan dan pendidikan. Selain itu, terdapat persamaan dalam sub proses asimilasi struktural primer, yaitu asimilasi yang mengacu pada kontak sosial dalam masyarakat. Hal ini disebabkan oleh tingginya kekeluargaan yang dimiliki etnik Chicano yang menjagakeeratan hubungan sosial diantara mereka

    Taxing the Disposition Effect: The Impact of Tax Awareness on Investor Behavior

    Standard portfolio choice models predict that investors consider the tax implications of trading. However, individuals are disposed toward realizing gains and holding losing investments, behaviors that worsen their performance. We show, in an experimental market, that increasing tax salience reduces the disposition effect between 22% and 47%, leading to higher portfolio balances without increasing total trading activity. Using field data, we find that investors\u27 disposition is sensitive to taxes around tax rate changes, when taxes are likely salient. Our analysis demonstrates that increasing tax awareness can affect households\u27 portfolio choices, which suggests policy implications for improving financial decision-making

    Intracytoplasmic lipid droplets in high grade glioma: metabolism and target for therapy

    Glioblastoma is a highly malignant and aggressive high grade glioma with a poor prognosis. The low survival rates stem from tumour progression, late intervention, ineffective therapies and drug resistance, requiring new therapeutic and diagnostic approaches. Lipid droplets are dynamic organelles suggested to be influential facets of cancer metabolism and biology in many tumours. In glioblastoma lipid droplets have been associated with hypoxia higher clinical grades and poor survival however the cellular pathways underlying lipid droplet metabolism remain unclear. Using a publically available database of grade 2 ta 4 glioma gene expression, we observed that genes associated with lipid droplet metabolism were important prognostic survival and tumour progression indicators. Moreover, through confocal microscopy, flow cytometry and NMR-based methods, we observed that uptake of exogenous lipids and adipose triglyceride lipase-mediated lipid shuttling produced lipid droplets whilst autophagy was vital to lipid droplet breakdown. ATGL-mediated lipid shuttling was further observed to prevent unsaturated fatty acid oxidative damage. Finally, we investigated the effect of pharmacological lipid droplet manipulation and observed that autophagy inhibition can improve temozolomide and irradiation cytotoxicity. Taken together our data suggests that understanding lipid droplet metabolic pathways may generate prognostic bio-markers of survival and progression and improve current therapies

    Ecohydrology of a Floodplain Forest: Relationships Between Evapotranspiration, Vegetation, and Topography at Congaree National Park, South Carolina

    Congaree National Park supports high biodiversity and provides ecosystem services for the surrounding area in the floodplain wetland system, especially in the bottomland hardwood forests which contain some of the last remaining old-growth stands in the eastern U.S. Maintaining the hydraulic functions of this ecosystem is essential not only for the conservation of biodiversity, but also for the ecosystem services it provides, such as nitrification, denitrification, decomposition, removal of organic carbon, and phosphorous uptake and sorption. Because management practices of the park depend on understanding the area‟s hydrology, past research has been performed to analyze the flooding of Congaree River. However, not much has been done to better understand groundwater movement through the floodplain sediments in the Congaree River Valley. The goal of this project is to quantify interactions between the shallow unconfined aquifer and local vegetation surrounding eight piezometers in the Congaree Observation Well Network at Congaree National Park through calculating and comparing evapotranspiration rates, specific yield, vegetation diversity and basal area, and microtopography. Data on groundwater response to storm events, diurnal signals caused by evaporation and transpiration in the forest, vegetation community structure, and local topography were compared to better understand the role of these factors on vegetation water demand in this wetland-dominated system

    Determination of Triapine, A Ribonucleotide Reductase Inhibitor, In Human Plasma by Liquid Chromatography Tandem Mass Spectrometry

    Triapine is an inhibitor of ribonucleotide reductase (RNR). Studies have shown that triapine significantly decreases the activity of RNR and enhanced the radiation-mediated cytotoxicity in cervical and colon cancer. In this work, we have developed and validated a selective and sensitive LC-MS/MS method for the determination of triapine in human plasma. In this method, 2-[(3-fluoro-2-pyridinyl)methylene] hydrazinecarbothioamide (NSC 266749) was used as the internal standard (IS); plasma samples were prepared by deproteinization with acetonitrile; tripaine and the IS were separated on a Waters Xbridge Shield RP18 column (3.5 µm; 2.1 × 50 mm) using a mobile phase containing 25.0% methanol and 75.0% ammonium bicarbonate buffer (10.0 mm, pH 8.50; v/v); column eluate was monitored by positive turbo-ionspray tandem mass spectrometry; and quantitation of triapine was carried out in multiple-reaction-monitoring mode. The method developed had a linear calibration range of 0.250–50.0 ng/mL with correlation coefficient of 0.999 for triapine in human plasma. The IS-normalized recovery and the IS-normalized matrix factor of triapine were 101–104% and 0.89–1.05, respectively. The accuracy expressed as percentage error and precision expressed as coefficient of variation were ≤±6 and ≤8%, respectively. The validated LC-MS/MS method was applied to the measurement of triapine in patient samples from a phase I clinical trial
