MODEL SELECTION TECHNIQUES FOR REPEATED MEASURES COVARIANCE STRUCTURES

A parsimonious covariance structure of repeated measures is often sought for purposes of increased power for testing hypotheses about the means, and for insight into the stochastic processes governing the repeated measures. For normal data, model selection is often based upon likelihood ratio tests or information criteria derived from the likelihood, sometimes supplemented with graphical plots of correlations and partial correlations. We exploit the ordered nature of repeated measures to decompose the likelihood ratio goodness-of-fit test statistic, and display graphical fingerprints associated with the covariance structures to help detect covariance structure misspecification, in order to provide guidance in choosing an appropriate structure for the data. The proposed methodology is illustrated with simulated repeated measures data and then applied to an experiment to compare tillage methods of pasture establishment

Soluble RAGE: a hot new biomarker for the hot joint?

The receptor for advanced glycation endproducts (RAGE) interacts with distinct ligand families linked to the inflammatory response. Studies in animal models suggest that RAGE is upregulated in the inflamed joint and that blockade of the receptor, using a ligand decoy soluble form of RAGE (sRAGE), attenuates joint inflammation and expression of inflammatory and tissue-destructive mediators. In this issue of Arthritis Research & Therapy, Rille Pullerits and colleagues reported that plasma levels of sRAGE were reduced in subjects with rheumatoid arthritis compared with healthy controls or subjects with non-inflammatory joint disease. These findings suggest the possibility that levels of sRAGE might be a biomarker of inflammation. Not resolved by these studies, however, is the intriguing possibility that endogenously higher levels of sRAGE might be linked to a lower incidence of arthritis or to the extent of inflammation. Nevertheless, although 'cause or effect' relationships may not be established in this report, fascinating insights into RAGE, inflammation and human arthritis emerge from these studies

A COMPARISON OF SPATIAL PREDICTION METHODS USING INTENSE SPATIALLY-ACQUIRED WATER QUALITY DATA

Water quality information obtained through intensive spatial sampling using automated devices provides opportunities to monitor and forecast the spatial distribution of nutrients and phytoplankton concentrations, and help establish water circulation patterns in estuarine and coastal waters. To be cost effective, efficient sampling designs and estimation methodologies must first be developed. As a starting basis, we applied an original transect sampling design that was used to estimate the spatial distribution of chlorophyll a, salinity, and temperature in the Cienaga Grande de Santa Marta, a coastal lagoon in Colombia. We superimposed the transects over satellite images of the lagoon obtained in the period 1993-2001 to evaluate the efficiency and accuracy of using such transects to estimate the distribution of water quality variables. The satellite images were taken in 1993 (SPOT-3), 1995 (Landsat-6), and 1999 (Landsat-6), and water reflectance values were used as a “proxy” for the water quality variables. Spatial prediction using kriging and thin-plate smoothing splines were used to predict reflectance for a grid network of points taken from the images, and predictions were compared with observed values to compare methods and transect routes. Rapid changes in reflectance in short distances (for example , caused by phytoplankton blooms), complicated the analysis, and neither method proved superior over all transect routes and images, although the kriging predictor remained relatively consistent in performance over the various selected sampling routes

Glycation and diabetes: The RAGE connection

The hyperglycaemic state seen in diabetes mellitus is associated with the development of diabetes-specific microvascular complications and accelerated macrovascular disease. Evidence implicates the formation and subsequent effects of advanced glycation endproducts (AGEs) as a contributing cause. AGEs exert their effects through interaction with the Receptor for AGE (RAGE) which upregulates expression of the receptor and induces a cascade of cytotoxic pathways. Accumulation of AGE/RAGE can be seen at sites of vascular disease in both animal models of diabetes and human diabetic subjects. Blockade of RAGE in animal models of diabetes suppresses development of dysfunction in the vasculature and atherosclerosis development. Genetic studies of RAGE reveal that a number of allelic variants of RAGE occur in key protein and regulatory domains. A Gly to Ser change at position 82 and two 5¢¢ flanking polymorphisms at position –374 and –429 lead to altered function and expression of RAGE which may impact on diabetic vascular disease development. Therapy aimed to block RAGE upregulation may prove to be useful in treating individuals with diabetic vascular disease

For patients with cancer who are married or in an intimate relationship, their relationships with their partners play a critical role in their adaptation to their illness. However, cancer patients and their partners often have difficulty in talking with each other about their cancer-related concerns. Difficulties in communication may ultimately compromise both the patient-partner relationship and the patient's psychological adjustment.
