    Enhanced relativistic-electron beam collimation using two consecutive laser pulses

    The double laser pulse approach to relativistic electron beam (REB) collimation has been investigated at the LULI-ELFIE facility. In this scheme, the magnetic field generated by the first laser-driven REB is used to guide a second delayed REB. We show how electron beam collimation can be controlled by properly adjusting laser parameters. By changing the ratio of focus size and the delay time between the two pulses we found a maximum of electron beam collimation clearly dependent on the focal spot size ratio of the two laser pulses and related to the magnetic field dynamics. Cu-K alpha and CTR imaging diagnostics were implemented to evaluate the collimation effects on the respectively low energy ( MeV) components of the REB

    Antenatal Microbial Colonization of Mammalian Gut

    The widely accepted dogma of intrauterine sterility and initial colonization of the newborn during birth has been blurred by recent observations of microbial presence in meconium, placenta, and amniotic fluid. Given the importance of a maternal-derived in utero infant seeding, it is crucial to exclude potential environmental or procedural contaminations and to assess fetal colonization before parturition. To this end, we analyzed sterilely collected intestinal tissues, placenta, and amniotic fluid from rodent fetuses and tissues from autoptic human fetuses. Total bacterial DNA was extracted from collected samples and analyzed by Next Generation Sequencing (NGS) techniques using hypervariable 16S ribosomal RNA (rRNA) regions (V3-V4). Colonizing microbes were visualized in situ, using labeled probes targeting 16S ribosomal DNA by fluorescent in situ hybridization. The NGS analysis showed the presence of pioneer microbes in both rat and human intestines as well as in rodent placentas and amniotic fluids. Microbial communities showed fetus- and dam-dependent clustering, confirming the high interindividual variability of commensal microbiota even in the antenatal period. Fluorescent in situ hybridization analysis confirmed the microbes' presence in the lumen of the developing gut. These findings suggest a possible antenatal colonization of the developing mammalian gut

    Fashion brands strategies and contemporaneous consumer behavior on social media in Portugal

    This study presents two main analysis focused on Portuguese fashion brands strategies and contemporaneous consumer behavior. The first analysis consisted on the case studied and observation of three Portuguese brands (Patrick de Pádua, Luís Carvalho and Nair Xavier) aiming to understand their behavior on social media. During 4 months, Facebook and Instagram were monitored aiming to understand the strategy of each brand and the consequent results. The second analysis was a online survey with 436 valid answers focused on the understanding of Portuguese fashion consumer behavior. The case studies analysis allowed to understand that to have more followers and thus produce more interactivity on a continuous basis with their public, the brands have to create contents consistent with their markets and publics, with the brand language and also with the brand image. The results obtained on the survey allow to conclude that the number participants that share opinions about fashion products and services are still limited.     GUT microbiota change and time of restore in intensive care therapy : a case report

    Exposure of gut microbiota to antibiotics can promote antibiotic resistance and development of diseases caused by multi-drug-resistant organisms. Here we present a case of a 54-year-old male patient with a diagnosis of acute cholecystitis with significant biliary duct leakage, admitted to Intensive Care Unit (ICU) due to a septic syndrome after surgery. Fecal microbial population was analyzed by DGGE and Real Time PCR during and after a combined antibiotic therapy and enteral nutritional feeding. Gut microbiota dysbiosis was demonstrated during ICU recovery. After antibiotic therapy discontinuation and the switch to normal diet, microbial gut population gradually increased up to values comparable with those of a healthy subject. Bifidobacterium spp. took longer to re-stabilize, reaching normal value after two weeks. Our case report corroborates the resilient nature of gut microbiota, but points out the long time needed to recover after antibiotic treatment, paving the way to supplementation with key probiotic species

    Stop-event-related potentials from intracranial electrodes reveal a key role of premotor and motor cortices in stopping ongoing movements

    In humans, the ability to withhold manual motor responses seems to rely on a right-lateralized frontal–basal ganglia–thalamic network, including the pre-supplementary motor area and the inferior frontal gyrus (IFG). These areas should drive subthalamic nuclei to implement movement inhibition via the hyperdirect pathway. The output of this network is expected to influence those cortical areas underlying limb movement preparation and initiation, i.e., premotor (PMA) and primary motor (M1) cortices. Electroencephalographic (EEG) studies have shown an enhancement of the N200/P300 complex in the event-related potentials (ERPs) when a planned reaching movement is successfully stopped after the presentation of an infrequent stop-signal. PMA and M1 have been suggested as possible neural sources of this ERP complex but, due to the limited spatial resolution of scalp EEG, it is not yet clear which cortical areas contribute to its generation. To elucidate the role of motor cortices, we recorded epicortical ERPs from the lateral surface of the fronto-temporal lobes of five pharmacoresistant epileptic patients performing a reaching version of the countermanding task while undergoing presurgical monitoring. We consistently found a stereotyped ERP complex on a single-trial level when a movement was successfully cancelled. These ERPs were selectively expressed in M1, PMA, and Brodmann's area (BA) 9 and their onsets preceded the end of the stop process, suggesting a causal involvement in this executive function. Such ERPs also occurred in unsuccessful-stop (US) trials, that is, when subjects moved despite the occurrence of a stop-signal, mostly when they had long reaction times (RTs). These findings support the hypothesis that motor cortices are the final target of the inhibitory command elaborated by the frontal–basal ganglia–thalamic network

    Methicillin-Resistant Staphylococcus aureus in Raw Milk : Prevalence, SCCmec Typing, Enterotoxin Characterization, and Antimicrobial Resistance Patterns

    Staphylococcus aureus is a known major cause of foodborne illnesses, and raw milk and dairy products are often contaminated by enterotoxigenic and antimicrobial-resistant S. aureus strains. In the present study, 35 S. aureus strains were isolated from 383 raw milk samples collected from various dairy herds in the province of Milan (northern Italy). The isolates were characterized based on their antimicrobial susceptibility patterns and the presence of genes encoding staphylococcal enterotoxins (sea, seb, sec, sed, and see). About half (45.7%) of the strains were enterotoxigenic, and 37.1% were resistant to at least one of the antimicrobial drugs tested. Seven (20%) of 35 isolates were identified as methicillin-resistant S. aureus (MRSA), and SCCmec typing performed with a multiplex PCR assay revealed the presence of gene cassettes IV and V, typical of community acquired MRSA, and I and II, characteristic of health care\u2013associated MRSA. The MRSA strains were evaluated for the presence of the Panton-Valentine leukocidin gene, but this gene was not found. The results of the present study revealed the presence of toxin-producing S. aureus and MRSA strains in raw milk. MRSA and enterotoxigenic S. aureus in dairy farms are an important risk factor for the spread of staphylococcal infections; therefore, further studies are needed to find strategies for monitoring and controlling the presence of S. aureus, especially MRSA, in dairy products

    Time-of-flight methodologies with large-area diamond detectors for the effectively characterization of tens MeV protons

    A novel detector based on a polycrystalline diamond sensor is here employed in an advanced time-of-flight scheme for the characterization of energetic ions accelerated during laser-matter interactions. The optimization of the detector and of the advanced TOF methodology allow to obtain signals characterized by high signal-to-noise ratio and high dynamic range even in the most challenging experimental environments, where the interaction of high-intensity laser pulses with matter leads to effective ion acceleration, but also to the generation of strong Electromagnetic Pulses (EMPs) with intensities up to the MV/m order. These are known to be a serious threat for the fielded diagnostic systems. In this paper we report on the measurement performed with the PW-class laser system Vega 3 at CLPU (30 J energy, 1021 W/cm2 intensity, 30 fs pulses) irradiating solid targets, where both tens of MeV ions and intense EMP fields were generated. The data were analyzed to retrieve a calibrated proton spectrum and in particular we focus on the analysis of the most energetic portion (E > 5.8 MeV) of the spectrum showing a procedure to deal with the intrinsic lower sensitivity of the detector in the mentioned spectral-range

    Candidemia in intensive care unit : a nationwide prospective observational survey (GISIA-3 study) and review of the European literature from 2000 through 2013

    BACKGROUND: Candida bloodstream infections (BSI) represent an important problem in Intensive Care Units (ICUs). The epidemiology of candidemia is changing with an increase in the proportion of Candida (C.) non-albicans. OBJECTIVES: An Italian 2-year observational survey on ICU was conducted to evaluate the species distribution and possible differences between BSI caused by C. albicans and C. non-albicans. For comparative purposes, we performed a European literature-based review to evaluate distribution and frequency of Candida spp. causing ICU candidemia, during the period 2000-2013. MATERIALS AND METHODS: This laboratory-based survey involved 15 microbiology centers (GISIA-3 study). All candidemia episodes in adult patients were considered. Data were prospectively collected from 2007 to 2008. PubMed was searched for peer-reviewed articles. RESULTS: In total, 462 candidemia episodes were collected. C. albicans accounted for 49.4% of the isolates, followed by C. parapsilosis (26.2%) and C. glabrata (10.4%). Mortality was higher in patients with C. non-albicans than C. albicans (47.3% vs. 32.4 %, p > 0.05). Among risk factors, parenteral nutrition was more common (p = 0.02) in non-albicans candidemia, while surgery was more frequent (p = 0.02) in C. albicans candidemia. Twenty-four relevant articles were identified. C. albicans was the predominant species in almost all studies (range 37.9% -76.3%). C. glabrata was commonly isolated in the German-speaking countries, France, UK and North Europe; C. parapsilosis in Turkey, Greece and Spain. CONCLUSIONS: Although C. non-albicans BSI is increasing, our study shows that C. albicans is still the predominant species in ICU candidemia. There are differences in the epidemiology of Candida BSI among European countries, with a prevalence of C. glabrata and C. parapsilosis in Northern and Southern countries, respectively
