X-33 Metallic TPS Tests in NASA-LaRC High Temperature Tunnel
Conclusions The first series of metallic TPS tests in the NASA-LaRC Mach 7 High Temperature Tunnel has been completed. Additional testing is 'in progress and shall provide data for off-design configurations for the metallic TPS. The available data are being analyzed and being used to correlate analytical models to be used for X-33 flight design analysis. The final paper shan present additional data from these tests and comparisons between the data and analytical predictions
Comparative Analysis of Pollution in Farmington Bay and the Great Salt Lake, Utah
Farmington Bay covers 94 mi2 (260 km2) in the SW comer of the Great Salt Lake, and is essentially a separate lake because it is enclosed by Antelope Island and a causeway leading to the island from the mainland. The bay has received wastes from the adjoining Salt Lake City metropolitan area for decades. Because of water quality concerns for Farmington 8ay, the Aquatic Ecology Laboratory class at Utah State University studied the bay and a nearby control site (Bridger Bay) in the Great Salt Lake during the fall of 2001. Field sampling and laboratory experiments, as well as other data sources, demonstrated the bay is severely eutrophic and is one of the most polluted water bodies in the state of Utah. A preliminary nutrient loading estimate for the bay indicates that total phosphorus coming into the system is a-times higher than necessary for the bay to be classed as eutrophic. Sewage treatment plants discharging directly to the bay contribute approximately 500/0 of the nutrients. Metrics of eutrophication (chlorophyll, Secchi depth and total phosphorus) all indicated that the bay was hypereutrophic and the combined Trophic State Index was 91, higher than any other lake or reservoir in the state. Oxygen was supersaturated in the surface waters of Farmington Bay during the day, but the bottom water was anoxic. During the night, nearly the entire water column became anoxic due to respiratory demand of the biota. The anoxic conditions allowed high concentrations \u27Of foul-smelling hydrogen sulfide to be produced. Brine shrimp were not abundant in Farmington Bay and the community was dominated by rotifers. In contrast, water quality in Bridger 8ay located on the main lake, was good and brine shrimp were abundant there. Our results, although restricted in scope, corroborate existing monitoring data from this bay. Water quality characteristics in Farmington Bay do not meet those mandated for the protection of aquatic life. Odor problems from the bay likely impact more people than are affected by any other polluted water body in the state. The impact of eutrophication and anoxia on the biota in Farmington Bay may also be substantial, although inadequate data exists to determine these impacts. There are substantial technical challenges to be overcome if water quality in the bay is to be improved to meet its designated use. However, before these technical issues can be solved, the responsible agencies will need to address the problem, and begin studies that may eventually lead to a solution to this serious water quality issue
QFMatch: multidimensional flow and mass cytometry samples alignment
Part of the flow/mass cytometry data analysis process is aligning (matching) cell subsets between relevant samples. Current methods address this cluster-matching problem in ways that are either computationally expensive, affected by the curse of dimensionality, or fail when population patterns significantly vary between samples. Here, we introduce a quadratic form (QF)-based cluster matching algorithm (QFMatch) that is computationally efficient and accommodates cases where population locations differ significantly (or even disappear or appear) from sample to sample. We demonstrate the effectiveness of QFMatch by evaluating sample datasets from immunology studies. The algorithm is based on a novel multivariate extension of the quadratic form distance for the comparison of flow cytometry data sets. We show that this QF distance has attractive computational and statistical properties that make it well suited for analysis tasks that involve the comparison of flow/mass cytometry samples

were hunted close to extinction in the 20th century (Clapham et al. 1999). Their
recovery is now a key international conservation goal, and they are important in marine
ecosystems as massive consumers that can promote primary production through
nutrient cycling (Roman et al. 2014). However, although abundance has been
assessed to monitor the recovery of some large whale populations (e.g., Barlow et al.
2011, Laake et al. 2012) many populations are wide-ranging and pelagic, and this
inaccessibility has generally impeded quantitative assessments of recovery (Peel et al.
2015). To augment traditional abundance monitoring, we suggest that photogrammetric
measures of individual growth and body condition can also inform about population
status, enabling assessment of individual health as well as population numbers. Photogrammetry
from manned aircraft has used photographs taken from directly above
whales to estimate individual lengths (Gilpatrick and Perryman 2008) and monitor growth trends (Fearnbach et al. 2011), and shape profiles can be measured to assess
body condition to infer reproductive and nutritional status (e.g., Perryman and Lynn
2002, Miller et al. 2012). Recently, Durban et al. (2015) demonstrated the utility of
an unmanned hexacopter for collecting aerial photogrammetry images of killer
whales (Orcinus orca); this provided a noninvasive, cost-effective, and safe platform
that could be deployed from a boat to obtain vertical images of whales. Here we
describe the use of this small, unmanned aerial system (UAS) to measure length and

Adolescents’ responses to the promotion and flavouring of e-cigarettes
Objectives
The purpose of the study is to examine adolescents’ awareness of e-cigarette marketing and investigate the impact of e-cigarette flavour descriptors on perceptions of product harm and user image.
Methods
Data come from the 2014 Youth Tobacco Policy Survey, a cross-sectional in-home survey conducted with 11–16 year olds across the UK (n = 1205). Adolescents’ awareness of e-cigarette promotion, brands, and flavours was assessed. Perceptions of product harm, and likely user of four examples of e-cigarette flavours was also examined.
Results
Some participants had tried e-cigarettes (12 %) but regular use was low (2 %) and confined to adolescents who had also smoked tobacco. Most were aware of at least one promotional channel (82 %) and that e-cigarettes came in different flavours (69 %). Brand awareness was low. E-cigarettes were perceived as harmful (M = 3.54, SD = 1.19) but this was moderated by product flavours. Fruit and sweet flavours were perceived as more likely to be tried by young never smokers than adult smokers trying to quit (p < 0.001).
Conclusions
There is a need to monitor the impact of future market and regulatory change on youth uptake and perceptions of e-cigarettes
Multimodality assessment of the coronary microvasculature with TIMI frame count versus perfusion PET highlights coronary changes characteristic of coronary microvascular disease
BackgroundThe diagnosis of coronary microvascular disease (CMVD) remains challenging. Perfusion PET-derived myocardial blood flow (MBF) reserve (MBFR) can quantify CMVD but is not widely available. Thrombolysis in Myocardial Infarction (TIMI) frame count (TFC) is an angiography-based method that has been proposed as a measure of CMVD. Here, we compare TFC and PET-derived MBF measurements to establish the role of TFC in assessing for CMVD. We use coronary modeling to elucidate the relationship between MBFR and TFC and propose TFC thresholds for identifying CMVD.MethodsIn a cohort of 123 individuals (age 58 ± 12.1, 63% women, 41% Caucasian) without obstructive coronary artery disease who had undergone perfusion PET and coronary angiography for clinical indications, we compared TFC and perfusion PET parameters using Pearson correlation (PCC) and linear regression modeling. We used mathematical modeling of the coronary circulation to understand the relationship between these parameters and performed Receiver Operating Curve (ROC) analysis.ResultsWe found a significant negative correlation between TFC and MBFR. Sex, race and ethnicity, and nitroglycerin administration impact this relationship. Coronary modeling showed an uncoupling between TFC and flow in epicardial vessels. In ROC analysis, TFC performed well in women (AUC 0.84–0.89) and a moderately in men (AUC 0.68–0.78).ConclusionsWe established an inverse relationship between TFC and PET-derived MBFR, which is affected by patient selection and procedural factors. TFC represents a measure of the volume of the epicardial coronary compartment, which is increased in patients with CMVD, and performs well in identifying women with CMVD
-Tetrachlorodibenzo-p-dioxin Exposure: Effects on the Prostate and Its Response to Castration in Senescent C57BL/6J Mice
In utero and lactational 2,3,7,8-tetrachlorodibenzo-p-dioxin (TCDD) exposure inhibits ventral, dorsolateral, and anterior prostate development in C57BL/6 mice. To determine if prostatic abnormalities persist into senescence, mice born to dams given TCDD (5 mg/kg, po) or vehicle on gestation day 13 were examined at 100 and 510 days of age. Half the mice were castrated ten days prior to necropsy in order to assess androgen dependence, while the remaining mice were sham castrated. Effects of TCDD on the dorsolateral and anterior prostate of senescent sham-castrated mice were relatively subtle, whereas the ventral prostate was rudimentary or absent. Castration of vehicle-exposed mice caused far greater reductions in prostate lobe weights, epithelial cell height, and androgen-dependent gene expression (MP25 and probasin) in young mice than in senescent ones, while cell proliferation was decreased by castration in young mice and increased in senescence. Responses to castration were similar at 100 days of age in vehicle-and TCDD-exposed mice. At 510 days, however, TCDD-exposed mice were substantially more responsive to castration by most indices than vehicle-exposed mice. These results demonstrate that prostatic androgen dependence in mice declines substantially with age in several key ways, and that in utero and lactational TCDD exposure protects against this decline. Surprisingly, TCDD increased the incidence of cribriform structures in dorsolateral prostate ducts, from 2-3% in vehicleexposed senescent mice to 16% in sham-castrated and to 7% in castrated senescent mice. Collectively, these results demonstrate that effects of in utero and lactational TCDD exposure on the prostate persist into senescence, and suggest that in utero and lactational TCDD exposure retards the aging process in the prostate. However, because cribriform structures are often considered to be associated with prostate carcinogenesis, these results also suggest that TCDD exposure early in development may increase susceptibility to prostate cancer
Earth Mover’s Distance (EMD): A True Metric for Comparing Biomarker Expression Levels in Cell Populations
Changes in the frequencies of cell subsets that (co)express characteristic biomarkers, or levels of the biomarkers on the subsets, are widely used as indices of drug response, disease prognosis, stem cell reconstitution, etc. However, although the currently available computational “gating” tools accurately reveal subset frequencies and marker expression levels, they fail to enable statistically reliable judgements as to whether these frequencies and expression levels differ significantly between/among subject groups. Here we introduce flow cytometry data analysis pipeline which includes the Earth Mover’s Distance (EMD) metric as solution to this problem. Well known as an informative quantitative measure of differences between distributions, we present three exemplary studies showing that EMD 1) reveals clinically-relevant shifts in two markers on blood basophils responding to an offending allergen; 2) shows that ablative tumor radiation induces significant changes in the murine colon cancer tumor microenvironment; and, 3) ranks immunological differences in mouse peritoneal cavity cells harvested from three genetically distinct mouse strains
