    An insight to HTLV-1-associated myelopathy/tropical spastic paraparesis (HAM/TSP) pathogenesis; evidence from high-throughput data integration and meta-analysis

    Background Human T-lymphotropic virus 1-associated myelopathy/tropical spastic paraparesis (HAM/TSP) is a progressive disease of the central nervous system that significantly affected spinal cord, nevertheless, the pathogenesis pathway and reliable biomarkers have not been well determined. This study aimed to employ high throughput meta-analysis to find major genes that are possibly involved in the pathogenesis of HAM/TSP. Results High-throughput statistical analyses identified 832, 49, and 22 differentially expressed genes for normal vs. ACs, normal vs. HAM/TSP, and ACs vs. HAM/TSP groups, respectively. The protein-protein interactions between DEGs were identified in STRING and further network analyses highlighted 24 and 6 hub genes for normal vs. HAM/TSP and ACs vs. HAM/TSP groups, respectively. Moreover, four biologically meaningful modules including 251 genes were identified for normal vs. ACs. Biological network analyses indicated the involvement of hub genes in many vital pathways like JAK-STAT signaling pathway, interferon, Interleukins, and immune pathways in the normal vs. HAM/TSP group and Metabolism of RNA, Viral mRNA Translation, Human T cell leukemia virus 1 infection, and Cell cycle in the normal vs. ACs group. Moreover, three major genes including STAT1, TAP1, and PSMB8 were identified by network analysis. Real-time PCR revealed the meaningful down-regulation of STAT1 in HAM/TSP samples than AC and normal samples (P = 0.01 and P = 0.02, respectively), up-regulation of PSMB8 in HAM/TSP samples than AC and normal samples (P = 0.04 and P = 0.01, respectively), and down-regulation of TAP1 in HAM/TSP samples than those in AC and normal samples (P = 0.008 and P = 0.02, respectively). No significant difference was found among three groups in terms of the percentage of T helper and cytotoxic T lymphocytes (P = 0.55 and P = 0.12). Conclusions High-throughput data integration disclosed novel hub genes involved in important pathways in virus infection and immune systems. The comprehensive studies are needed to improve our knowledge about the pathogenesis pathways and also biomarkers of complex diseases.Peer reviewe

    NEOTROPICAL XENARTHRANS: a data set of occurrence of xenarthran species in the Neotropics

    Xenarthrans – anteaters, sloths, and armadillos – have essential functions for ecosystem maintenance, such as insect control and nutrient cycling, playing key roles as ecosystem engineers. Because of habitat loss and fragmentation, hunting pressure, and conflicts with 24 domestic dogs, these species have been threatened locally, regionally, or even across their full distribution ranges. The Neotropics harbor 21 species of armadillos, ten anteaters, and six sloths. Our dataset includes the families Chlamyphoridae (13), Dasypodidae (7), Myrmecophagidae (3), Bradypodidae (4), and Megalonychidae (2). We have no occurrence data on Dasypus pilosus (Dasypodidae). Regarding Cyclopedidae, until recently, only one species was recognized, but new genetic studies have revealed that the group is represented by seven species. In this data-paper, we compiled a total of 42,528 records of 31 species, represented by occurrence and quantitative data, totaling 24,847 unique georeferenced records. The geographic range is from the south of the USA, Mexico, and Caribbean countries at the northern portion of the Neotropics, to its austral distribution in Argentina, Paraguay, Chile, and Uruguay. Regarding anteaters, Myrmecophaga tridactyla has the most records (n=5,941), and Cyclopes sp. has the fewest (n=240). The armadillo species with the most data is Dasypus novemcinctus (n=11,588), and the least recorded for Calyptophractus retusus (n=33). With regards to sloth species, Bradypus variegatus has the most records (n=962), and Bradypus pygmaeus has the fewest (n=12). Our main objective with Neotropical Xenarthrans is to make occurrence and quantitative data available to facilitate more ecological research, particularly if we integrate the xenarthran data with other datasets of Neotropical Series which will become available very soon (i.e. Neotropical Carnivores, Neotropical Invasive Mammals, and Neotropical Hunters and Dogs). Therefore, studies on trophic cascades, hunting pressure, habitat loss, fragmentation effects, species invasion, and climate change effects will be possible with the Neotropical Xenarthrans dataset

    Buscando compreender as relações entre emoção e imaginação numa pratica de leitura

    As relações existentes entre emoção e imaginação configuram um problema, que ainda e pouco estudado no ambito educacional. Isso devido ao fato da importância exagerada que se dá aos aspectos cognitivos, ênfase essa que näo é dada apenas no âmbito escolar, mas também na sociedade como um todo, relegando, assim, as relações existentes entre os aspectos afetivos, motor, social e cognitivo, a segundo plano. Faz-se necessário olhar a sala de aula, ou melhor a escola e a Educação, de uma maneira diferenciada, ou seja, não apenas privilegiando o cognitivo, como se esse fosse o fator decisivo na conquista do conhecimento. É indiscutível que a instituição escolar tem um papel muito importante na formação do individuo, as experiências, os conhecimentos e tudo que e vivenciado dentro da escola, e por meio da escola, possuem um grande significado para o desenvolvimento social e afetivo da criançaSendo assim, a escola, e todos os sujeitos envolvidos nela, assumem um papel relevante no desenvolvimento infantil. A ausencia de uma educação que não reconheça a importância da emoção na sala de aula, segundo Almeida (1999), traz prejuízos para a ação pedagogica, pois suas conseqüências atingem não somente o professor, mas também, e principalmente, o aluno. Tornando-se assim necessário que nos cursos de formação de professores haja uma maior preocupação com o estudo das emoções e também da imaginação, assim como hã uma preocupação constante com a cognição, com a aquisição do conhecimento, para que nos, futuros pedagogos, sejamos capazes de saber enfrentar as diversas situações, que envolvem o aspecto emocional, que ocorrem na sala de aula. A partir de uma experiência vivenciada por sujeitos envolvidos em um projeto de incentivo a leitura foi possível vivenciar e perceber a importãncia da imaginação, e da emoção e a sua relação com o conhecimento no contexto escolar. Situações que serão aqui narradas, discutidas e analisadas a luz da perspectiva Historico Social, de Wallon e Vvgostsky pretendem trazer a tona a importância da emoção e da imaginação no desenvolvimento da crianç

    Water Uptake in PHBV/Wollastonite Scaffolds: A Kinetics Study

    Poly(3-hydroxybutyrate-co-3-hydroxyvalerate) (PHBV) is a widely studied polymer and it has been found that porous PHBV materials are suitable for substrates for cell cultures. A crucial factor for scaffolds designed for tissue engineering is the water uptake. This property influences the transport of water and nutrients into the scaffold, which promotes cell growth. PHBV has significant hydrophobicity, which can harm the production of cells. Thus, the addition of α-wollastonite (WOL) can modify the PHBV scaffold’s water uptake. To our knowledge, a kinetics study of water uptake of α-wollastonite phase powder and the PHBV matrix has not been reported. In this work, PHBV and WOL, (PHBV/WOL) films were produced with 0, 5, 10, and 20 wt % of WOL. Films were characterized, and the best concentrations were chosen to produce PHBV/WOL scaffolds. The addition of WOL in concentrations up to 10 wt % increased the cell viability of the films. MTT analysis showed that PHBV/5%WOL and PHBV/10%WOL obtained cell viability of 80% and 98%, respectively. Therefore, scaffolds with 0, 5 and 10 wt % of WOL were fabricated by thermally induced phase separation (TIPS). Scaffolds were characterized with respect to morphology and water uptake in assay for 65 days. The scaffold with 10 wt % of WOL absorbed 44.1% more water than neat PHBV scaffold, and also presented a different kinetic mechanism when compared to other samples. Accordingly, PHBV/WOL scaffolds were shown to be potential candidates for biological applications

    Human T-lymphotropic virus type 1 (HTLV-1) is the agent of the HTLV-1-associated myelopathy/tropical spastic paraparesis (HAM/TSP), which may Occur in > 5% of patients during their lifetime. HTLV-1-infection causes disturbances in the immune system, and the viral load may also play an important role in the pathogenesis of HAM/TSP. Some cytokines are involved in the pathogenesis of this disorder. We have determined IL-2, IL-4, IL-10, IL-12 p70, IFN-gamma and TNF-alpha production among HTLV-1-infected subjects from our HTLV-out Clinic in Institute of Infectious `Emilio Ribas` in Sao Paulo city, Brazil. PBMC obtained from healthy controls (n = 32), asymptomatic HTLV-1 carriers (n = 68) and HAM/TSP patients (n = 44) were grown in the absence and in the presence of phytohaemagglutinin (PHA), and the supernatants` fluids were measured for cytokines production. IL-2 levels were increased in the a-symptomatic HTLV-1 carriers, and IFN-gamma was increased in both groups of patients (asymptomatic HTLV-1 carriers and more significantly among HAM/TSP patients). IL-4, IL-10, TNF-alpha and IL-12 p70 levels were not significantly increased on both groups of patients, as compared with controls. The major finding Of this Study is that IFN-gamma was an important cytokine for the HAM/TSP pathogenesis.
