RĂŽle de lâazote dans la structure et la fonction des communautĂ©s de cyanobactĂ©ries toxiques
Dans cette étude de trois lacs sujets aux efflorescences de cyanobactéries, nous
avons examiné la diversité des bactéries diazotrophes et des cyanobactéries toxiques. Nous avons tenté de définir les facteurs environnementaux influençant la composition des communautés phytoplanctoniques, la concentration ainsi que la composition des
microcystines (MCs). Nous avons Ă©mis lâhypothĂšse que lâazote jouerait un rĂŽle majeur dans le façonnement des communautĂ©s cyanobactĂ©riennes et influencerait la concentration et composition des MCs. Des concentrations de cette toxine ainsi que le gĂšne mcyE codant pour lâenzyme microcystine synthĂ©tase ont Ă©tĂ© dĂ©tectĂ©s Ă chaque Ă©chantillonnage dans tous les lacs. Lâazote, particuliĂšrement sous sa forme organique dissoute (AOD) ainsi que la tempĂ©rature de lâeau Ă©taient les facteurs environnementaux expliquant le mieux les concentrations des MCs, tandis que la biomasse de Microcystis spp. Ă©tait globalement le meilleur prĂ©dicteur. Le gĂšne nifH codant pour lâenzyme nitrogĂ©nase (fixation dâazote) a
aussi été détecté dans chaque échantillon. Malgré les concentrations faibles en azote
inorganique dissous (AID) et les densitĂ©s importantes dâhĂ©tĂ©rocystes, aucun transcrits du gĂšne nâa Ă©tĂ© dĂ©tectĂ© par rĂ©verse-transcription (RT-PCR), indiquant que la fixation dâazote nâavait pas lieu Ă des niveaux dĂ©tectables au moment de lâĂ©chantillonnage. De plus, le pyrosĂ©quençage rĂ©vĂšle que les sĂ©quences des gĂšnes nifH et mcyE correspondaient Ă
diffĂ©rents taxons, donc que les cyanobactĂ©ries nâavaient pas la capacitĂ© dâeffectuer les deux fonctions simultanĂ©ment.In this study of three eutrophic lakes prone to cyanobacterial blooms, we examined
the diversity of diazotrophic bacteria and toxic cyanobacteria. We evaluated the
environmental factors effects on the community composition, the concentration and
composition of the family of toxins microcystins (MCs). Since the assimilation of nitrogen and the synthesis of MCs in cyanobacteria are thought to be under the same control of the NtcA transcriptor, we hypothesised that nitrogen played a major role in shaping cyanobacterial communities and influenced indirectly the concentration and composition of MCs. The mcyE gene coding for the microcystin synthetase enzyme and MCs
concentrations were detected at each sampling date in all lakes. Nitrogen, particularly under its organic dissolved form (DON) as well as water temperature were the environmental factors explaining the most variation in MC concentration although Microcystis spp. biomass was overall the best predictor. The nifH gene coding for the nitrogenase enzyme (N2-fixation) was also detected at all times. Even though the concentrations of dissolved inorganic nitrogen were relatively low, and that heterocysts were present in high densities, no nifH transcripts were detected by RT-PCR, indicating that no N2-fixation was going on
at detectable levels at the time of sampling. Moreover, pyrosequencing revealed that sequences of the genes nifH and mcyE corresponded to different taxa, thus cyanobacteria did not have the capacity to perform both functions simultaneously

A propos de l'Ă©tude de la synthĂšse des protĂ©ines, un graphique, classique pour les biologistes, qui rassemble des donnĂ©es obtenues par les techniques de marquage radioactif et par la microscopie Ă©lectronique a Ă©tĂ© donnĂ© Ă analyser Ă des Ă©lĂšves de classe Terminale. Les rĂ©ponses des Ă©lĂšves sont classĂ©es et commentĂ©es. Beaucoup ne semblent pas pertinentes par rapport Ă ce qu'on attend de la rĂ©citation d'un savoir. Ces erreurs sont-elles condamnables? Long Term Diversity and Distribution of Non-photosynthetic Cyanobacteria in Peri-Alpine Lakes
The phylum Cyanobacteria comprises a non-photosynthetic lineage. The diversity and distribution of non-photosynthetic cyanobacteria (NCY) across aquatic environments are currently unknown, including their ecology. Here, we report about composition and phylogenetic diversity of two clades of NCY in ten lakes of the European peri-Alpine region, over the past âŒ100 years. Using 16S rDNA sequences obtained from dated sediment cores, we found almost equal proportion of taxa assigned to Melainabacteria and the deepest-branching group Sericytochromatia (ML635J-21) (63 total detected taxa). The topology of our reconstructed phylogenies reflected evolutionary relationships expected from previous work, that is, a clear separation between the deepest branching Sericytochromatia, the Melainabacteria, and the photosynthetic cyanobacteria clades. While different lakes harbored distinct NCY communities, the diversity of NCY assemblages within and between lakes (alpha and beta diversity) did not significantly change over the last century. This is in contrast with what was previously reported for photosynthetic cyanobacteria. Unchanged community phylogenetic similarity over geographic distance indicated no dispersal limitation of NCY at the regional scale. Our results solicit studies linking in-lake environmental factors to the composition of these microorganismsâ communities, whose assembly appeared not to have been influenced by large-scale anthropogenic environmental changes. This is the first attempt to study the diversity and distribution of NCY taxa across temperate lakes. It provides a first step towards understanding their distribution and ecological function in pelagic aquatic habitats, where these organisms seem to be prevalent
