    Impact of natural killer cells on outcomes after allogeneic hematopoietic stem cell transplantation: A systematic review and meta-analysis.

    Background: Natural killer (NK) cells play a vital role in early immune reconstitution following allogeneic hematopoietic stem cell transplantation (HSCT). Methods: A literature search was performed on PubMed, Cochrane, and Clinical trials.gov through April 20, 2022. We included 21 studies reporting data on the impact of NK cells on outcomes after HSCT. Data was extracted following the PRISMA guidelines. Pooled analysis was done using the meta-package (Schwarzer et al.). Proportions with 95% confidence intervals (CI) were computed. Results: We included 1785 patients from 21 studies investigating the impact of NK cell reconstitution post-HSCT (8 studies/1455 patients), stem cell graft NK cell content (4 studies/185 patients), therapeutic NK cell infusions post-HSCT (5 studies/74 patients), and pre-emptive/prophylactic NK cell infusions post-HSCT (4 studies/77 patients). Higher NK cell reconstitution was associated with a better 2-year overall survival (OS) (high: 77%, 95%CI 0.73-0.82 vs low: 55%, 95%CI 0.37-0.72; n=899), however, pooled analysis for relapse rate (RR) or graft versus host disease (GVHD) could not be performed due to insufficient data. Higher graft NK cell content demonstrated a trend towards a better pooled OS (high: 65.2%, 95%CI 0.47-0.81 vs low: 46.5%, 95%CI 0.24-0.70; n=157), lower RR (high: 16.9%, 95%CI 0.10-0.25 vs low: 33%, 95%CI 0.04-0.72; n=157), and lower acute GVHD incidence (high: 27.6%, 95%CI 0.20-0.36 vs low: 49.7%, 95%CI 0.26-0.74; n=157). Therapeutic NK or cytokine-induced killer (CIK) cell infusions for hematologic relapse post-HSCT reported an overall response rate (ORR) and complete response (CR) of 48.9% and 11% with CIK cell infusions and 82.8% and 44.8% with NK cell infusions, respectively. RR, acute GVHD, and chronic GVHD were observed in 55.6% and 51.7%, 34.5% and 20%, and 20.7% and 11.1% of patients with CIK and NK cell infusions, respectively. Pre-emptive donor-derived NK cell infusions to prevent relapse post-HSCT had promising outcomes with 1-year OS of 69%, CR rate of 42%, ORR of 77%, RR of 28%, and acute and chronic GVHD rates of 24.9% and 3.7%, respectively. Conclusion: NK cells have a favorable impact on outcomes after HSCT. The optimal use of NK cell infusions post-HSCT may be in a pre-emptive fashion to prevent disease relapse

    Outcomes with chimeric antigen receptor t-cell therapy in relapsed or refractory acute myeloid leukemia: a systematic review and meta-analysis

    BackgroundWe conducted a systematic review and meta-analysis to evaluate outcomes following chimeric antigen receptor T cell (CAR-T) therapy in relapsed/refractory acute myeloid leukemia (RR-AML).MethodsWe performed a literature search on PubMed, Cochrane Library, and Clinicaltrials.gov. After screening 677 manuscripts, 13 studies were included. Data was extracted following PRISMA guidelines. Pooled analysis was done using the meta-package by Schwarzer et al. Proportions with 95% confidence intervals (CI) were computed.ResultsWe analyzed 57 patients from 10 clinical trials and 3 case reports. The pooled complete and overall response rates were 49.5% (95% CI 0.18-0.81, I2 =65%) and 65.2% (95% CI 0.36-0.91, I2 =57%). The pooled incidence of cytokine release syndrome, immune-effector cell associated neurotoxicity syndrome, and graft-versus-host disease was estimated as 54.4% (95% CI 0.17-0.90, I2 =77%), 3.9% (95% CI 0.00-0.19, I2 =22%), and 1.6% (95%CI 0.00-0.21, I2 =33%), respectively.ConclusionCAR-T therapy has demonstrated modest efficacy in RR-AML. Major challenges include heterogeneous disease biology, lack of a unique targetable antigen, and immune exhaustion

One approach to solve this problem is to measure the global velocity field. Therefore Particle Image Velocimetry (PIV) has to be arranged for use in high speed flows. In this project PIV setup was installed for the use in hypersonic Ludwieg tube Braunschweig (HLB).     Profiling of anti-oxidative enzymes and lipid peroxidation in leaves of salt tolerant and salt sensitive maize hybrids under NaC1 and Cd stress

    Effects of NaCl salinity and cadmium on the anti-oxidative activity of enzymes like superoxide dismutase (SOD), catalase (CAT), peroxidase (POD), ascorbate peroxidase (APX), glutathione reductase (GR) and lipid peroxidation contents; malondialdehyde (MDA) were studied in two maize hybrids of different salt tolerance characteristics. An increase in the amount of lipid peroxidation indicated the oxidative stress induced by NaCl and Cd. The results also depicted that NaCl stress caused an increase in the activities of POD, SOD, CAT, APX and GR while cadmium stress increased the activities of POD, SOD and APX but showed no significant effect on CAT and GR in both the studied hybrids. The combined effect of salinity and cadmium on these parameters was higher than that of sole effect of either NaCl or Cd. It was also found that maize hybrid 26204 had better tolerance against both stresses with strong antioxidant system as compared to that of maize hybrid 8441. A comparison of the antioxidants and lipid peroxidation in two maize hybrids having varying level of NaCl and Cd stress tolerance corroborated the importance of reactive oxygen species (ROS) in defense against abiotic stresses

    Date palm counts among the oldest fruit crops of the world and is mainly cultivated for its highly nutritious fruits consumed as a staple food in many countries, especially in the Gulf region. Dates are enriched with numerous therapeutic bioactives and functional compounds such as phenolics, flavonols, carotenoids, minerals, and vitamins that not only provide an appreciable amount of energy required for the human body but also act as an effective therapeutic agent against several diseases. This review aimed to provide a deep insight into the nutritional as well as phytochemicals profile of date fruit and its seeds in order to explore their biological (anti-cancer, anti-diabetic, cardio-protective, anti-inflammatory properties), functional food, and nutra-pharmaceutical attributes. PRACTICAL APPLICATIONS: This review provides updated information regarding the date fruits and seeds phytochemicals composition together with highlighting dates potential as a natural therapeutic agent against several diseases. The study also urges the importance of consuming dates as a great package to live a healthy life due to the functional food and nutraceutical properties of this valuable fruit. The study also provides information first time as recommending dates to cope with the hidden hunger or micronutrient deficiency faced by the third world inhabitants. Hence, the review may further help the industry and researchers to explore the potential of dates for future medicinal and nutra-pharmaceutical applications

    Study of Comparative Morphology of Eight Cultivated Genotypes of <i>Olea europaea</i> L

    The current study was designed to assess the comparative morphology of eight olive cultivars with different geographical origins and diverse genetic backgrounds, introduced to a new climatic zone. The morphological parameters of eight (five exotic and three domestic) olive cultivars (Bari Zaitoon-1, Bari Zaitoon-2, Favolosa (FS-17), Koroneiki, Balkasar, Ottobratica, Leccino, and Arbequina) were compared at the experimental area of the Department of Botany, The Islamia University of Bahawalpur, Pakistan (29°24′0″ North, 71°41′0″ East, 401–421 feet above sea level). Plant height, number of leaves/15 cm shoot, leaf size characteristics (leaf length, leaf width, leaf area, and length/width ratio), leaf shape characteristics (margin, leaf axil, base, and apex angles), leaf pigments (Chlorophyll a, Chlorophyll b, total chlorophyll contents, and carotenoids), phyllotaxy, and leaf color and venation were recorded. The highest plant height (28 cm) was obtained by Bari Zaitoon-2 followed by Bari Zaitoon-1 (24 cm), both of which are domestic cultivar of Pakistan, while the shortest height (5 cm) was obtained by Koroneiki. Leccino displayed the highest average number of leaves (17.8) on main shoot, followed by BARI-2 (16.4) and the lowest score was from Balkasar (10.4). Leaf area ranged from 5.66 cm2 (Bari Zaitoon-1) to 3.08 cm2 (Koroneiki). The longest leaf length (5.74 cm) was found in Bari Zaitoon-1 and the shortest (4.04 cm) in Koroneiki, while the broadest leaves were found in Leccino (1.54 cm) and the narrowest (1.12 cm) in Koroneiki. Bari Zaitoon-2 led in leaf length to width ratio (4.058) followed by Bari Zaitoon-1 (3.772) with small lanceolate leaves hardly reaching the value of 4, with the lowest value illustrated by Leccino. The total chloroplast pigments were highest in FS-17 followed by Bari Zaitoon-1 and Bari Zaitoon-2, while the lowest was in Arbequina. Chlorophyll a was highest in Bari Zaitoon-1 followed by FS-17 and Balkasar, with the lowest rate in Arbequina. Chlorophyll b content of FS-17 was the highest whereas the Chlorophyll b and total chlorophyll contents in Arbequina were the lowest of all the cultivars. The highest value of total carotenoids was found in Balkasar followed by FS-17 with the lowest value in Arbequina. The phyllotaxy was categorized into three types, i.e., alternate, opposite, and whorled. The combination of two or more types was usually observed on the same branch. The whorl of four leaves was also present in rare cases. Leaf venation was both pinnate and reticulate. The leaf base of most (four) of the olive cultivars, i.e., Arbequina, Balkasar, Leccino and FS-17, were cuneate having acute, rounded, apiculate, and cuspidate leaf tips, respectively. The findings revealed remarkable variations in olive morphology, especially in the leaves and a successful record of the preliminary data of olive cultivars from the study area was made. The present research demonstrated that local olive cultivars have unique characteristics that differentiate them from imported cultivars.     Alveolar bone loss is associated with infections and its augmentation is a pre-requisite for the success of dental implants. In present study, we aim to develop and evaluate novel freeze dried doxycycline loaded chitosan (CS)/hydroxyapatite (HA) spongy scaffolds where hydroxypropylmethyl cellulose (HPMC) was added as a crosslinker. Scaffolds displayed compressive strength of 14MPa/cm3 and 0.34 as elastic response. The interconnected pore diameter was 41-273μm, favorably provided the template supporting cells and transport. An overall 10% degradation was seen after 14day's studies at pH 7.4 in PBS. Doxycycline hyclate, a frequently used drug to counter oral infections, demonstrated an initial burst release (6-8h), followed by a sustain release profile for the remaining 64h. CS/HA/HPMC scaffolds were nontoxic and promoted pre-osteoblast cell viability as seen with live/dead calcein staining after 24h where scaffolds with 10% and 25% HPMC by weight of scaffold had more viable cells. Scaffolds with 10%, 20% and 25% HPMC by weight of scaffold showed efficient cellular adhesion as seen in scanning electron microscopy images (day 8) indicating that pre-osteoblast cells were able to adhere well on the surface and into the porous structure via cytoplasmic extensions. Hoechst 33258 nuclear staining at day 2 and 8 indicated cell proliferation which was further supported byMTT assay at day 2, 4 and 8. Although all scaffolds supported pre-osteoblast cell viability, alkaline phosphatase (ALP) staining demonstrated that upon induction, differentiation was pronounced in case of scaffolds with 10% HMPC scaffolds. Conclusively, these materials having all the required mechanical and biological properties are potential candidates for alveolar bone regeneration

    Impact of COVID-19 in hematopoietic stem cell transplant recipients: A systematic review and meta-analysis.

    BACKGROUND: Hematopoietic stem cell transplant (HSCT) recipients are at increased risk of mortality and morbidity with coronavirus disease 2019 (COVID-19) due to severe immune dysfunction. METHODS: A literature search was performed on PubMed, Cochrane, and Clinical trials.gov from the date of inception to 12/08/2021. We identified 19 original studies reporting data on COVID-19 in HSCT recipients after screening 292 articles. Data were extracted following preferred reporting items for systematic reviews and meta-analysis guidelines. Quality evaluation was done using the National Institutes of Health (NIH) quality assessment tool. Inter-study variance was calculated using Der Simonian-Laird Estimator. Pooled analysis was conducted using MetaXL. A random-effects model was used to estimate the proportions with 95% confidence intervals (CI). RESULTS: Of 6711 patients in 19 studies, 2031 HSCT patients with severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) infection were analyzed. The median age of patients was 56.9 (range 1-81.6) years, and 63% patients were men according to 14 studies. The median time from transplant to SARS-CoV-2 infection for autologous (auto) and allogeneic (allo) HSCT patients was 23.2 (0.33-350.5) months and 16.4 (0.2-292.7) months, respectively. The median follow-up time after COVID-19 diagnosis was 28 (0-262) days. The COVID-19 mortality rate was 19% (95% CI 0.15-0.24, I CONCLUSIONS: HSCT recipients have a high risk of mortality and clinical complications due to COVID-19. There is a need for ongoing vigilance, masks, and social distancing, vaccination, and aggressive management of SARS-CoV-2 infection in HSCT recipients
