    Inhibitory Effect of Berberine on Zeste Homolog 2 (Ezh2) Enhancement in Human Esophageal Cell Lines

    Purpose: To investigate the inhibitory effect of berberine treatment on enhancement of zeste of homolog 2 (Ezh2) expressions in KYSE450 human esophageal cancer cells.Methods: Transwell motility chambers were used to analyze cell migration and invasion. Bio-Rad protein assay was used for the determination of protein concentration. Chemiluminescence with ECL system was employed for the detection of protein bands as per the manufacturer’s protocol. Staining was carried out with Alexa-Fluor 647 mouse anti-BrdU antibody. Flow cytometry was performed after adding DAPI. Annexin-V/DAPI staining and flow cytometry were used for the quantification of apoptotic cell death. Total RNA was isolated from KYSE450 cells using an RNA isolation kit.Results: Berberine-induced inhibition of Ezh2 expression led to inhibition of cell proliferation by G1 phase cell cycle arrest and induced anti-invasive properties of KYSE450 cells in Boyden chamber assays. There was 92 % reduction in invasive tendency of KYSE450 cells following treatment with berberine. Histone  methylation inhibitor, 3-deazaneoplanocin A (DZNep), also led to a similar effect on cell proliferation of KYSE450 cells. Berberine treatment also resulted in strong transcriptional reduction of the AXL receptor kinase. The results of qRT-PCR and FACS analyses showed significant inhibition of AXL mRNA and protein expression in KYSE450 carcinoma cells after treatment with berberine.Conclusion: Berberine may be an effective therapeutic agent in the treatment of esophageal carcinoma.Keywords: Berberine, Histone methylation inhibitor, Anti-invasive, Cell proliferation, Human Esophageal cance

    Anti-HIV Effect of Liposomes Bearing CXCR4 Receptor Antagonist N15P

    Purpose: To evaluate lymphatic system targeting and inhibitory ability of N15P nano-liposomal preparation (naLipo-N15P) of CXCR4 receptor antagonist in HIV infection.Methods: Chemotactic and chemotaxic inhibition activity assays were used to analyze the biological activity of naLipo-N15P. The anti-HIV potential of NaLipo-N15P in vitro was evaluated when NaLipo- N15P combined with the peripheral blood mononuclear cells of Macaca fascicularis which carry the Simian immunodeficiency virus. Furthermore, the anti-HIV potential in vivo of NaLipo-N15P was evaluated by the plasma concentration and tissue distribution (Ki)Results: The half-maximal inhibitory concentration of naLipo-N15P binding to CXCR4 as an antagonist in competition with SDF-1α was 1.89 pM while Ki was 2.4 pM. Viral load was 289 ± 45. NaLipo- N15P majorly accumulated in liver and spleen.Conclusion: When N15P is encapsulated into nano-liposomes, it not only retains specific binding to CXCR4 and facilitates cell-type-specific targeting of nano-liposomes to PBMCs with high CXCR4 expression, but also shows enhanced anti-HIV effect. Therefore, we propose that naLipo-N15P as a CXCR4 antagonist will play an important role in inflammation and blocking of HIV infection.Keywords: Antagonist, CXCR4, Liposomes, Receptor, Inflammation, HI

    Quasi-16-day periodic meridional movement of the equatorial ionization anomaly

    Based on the daytime location of the equatorial ionization anomaly (EIA) crest derived from GPS observations at low latitude over China during the 2005–2006 stratospheric sudden warming (SSW), a quasi-16-day periodic meridional movement of EIA crest with the maximum amplitude of about 2 degrees relative to the average location of EIA crest has been revealed. In addition, periodic variations that are in phase with the meridional EIA movement are also revealed in the equatorial electrojet (EEJ) and F2 layer peak height (<i>hm</i>F2) over Chinese ionosonde stations Haikou and Chongqing. The quasi-16-day periodic component in Dst index is weak, and the 16-day periodic component does not exist in F10.7 index. Such large-scale periodic meridional movement of EIA crest is likely related to the globally enhanced stratospheric planetary waves coupled with anomalous stratospheric zonal wind connected with SSW. In addition, such large-scale periodic movement of EIA should be global, and can affect the ionospheric morphology around the low-latitude belt near the EIA region. Further case analysis, simulation and theoretical studies must proceed in order to understand the periodic movements of EIA connected with the different periodic atmospheric variations

    Data taking strategy for the phase study in ψ′→K+K−\psi^{\prime} \to K^+K^-

    The study of the relative phase between strong and electromagnetic amplitudes is of great importance for understanding the dynamics of charmonium decays. The information of the phase can be obtained model-independently by fitting the scan data of some special decay channels, one of which is ψ′→K+K−\psi^{\prime} \to K^{+}K^{-}. To find out the optimal data taking strategy for a scan experiment in the measurement of the phase in ψ′→K+K−\psi^{\prime} \to K^{+} K^{-}, the minimization process is analyzed from a theoretical point of view. The result indicates that for one parameter fit, only one data taking point in the vicinity of a resonance peak is sufficient to acquire the optimal precision. Numerical results are obtained by fitting simulated scan data. Besides the results related to the relative phase between strong and electromagnetic amplitudes, the method is extended to analyze the fits of other resonant parameters, such as the mass and the total decay width of ψ′\psi^{\prime}.Comment: 13 pages, 7 figure

    The epithelial and epidermal innate cytokines IL-25, IL-33, and thymic stromal lymphopoietin (TSLP) have pivotal roles in the initiation of allergic inflammation in asthma and atopic dermatitis (AD). However, the mechanism by which the expression of these innate cytokines is regulated remains unclear. Intelectin (ITLN) is expressed in airway epithelial cells and promotes allergic airway inflammation. We hypothesized that ITLN is required for allergen-induced IL-25, IL-33, and TSLP expression. In two asthma models, Itln knockdown reduced allergen-induced increases in Il-25, Il-33, and Tslp and development of type 2 response, eosinophilic inflammation, mucus overproduction, and airway hyperresponsiveness. Itln knockdown also inhibited house dust mite (HDM)-induced early upregulation of Il-25, Il-33, and Tslp in a model solely inducing airway sensitization. Using human airway epithelial cells, we demonstrated that HDM-induced increases in ITLN led to phosphorylation of epidermal growth factor receptor and extracellular-signal regulated kinase, which were required for induction of IL-25, IL-33, and TSLP expression. In two AD models, Itln knockdown suppressed expression of Il-33, Tslp, and Th2 cytokines and eosinophilic inflammation. In humans, ITLN1 expression was significantly increased in asthmatic airways and in lesional skin of AD.     The Alignment between Satellites and Central Galaxies: Theory vs. Observations

    Recent studies have shown that the distribution of satellite galaxies is preferentially aligned with the major axis of their central galaxy. The strength of this alignment has been found to depend strongly on the colours of the satellite and central galaxies, and only weakly on the mass of the halo in which the galaxies reside. In this paper we study whether these alignment signals, and their dependence on galaxy and halo properties, can be reproduced in a hierarchical structure formation model of a Λ\LambdaCDM concordance cosmology. To that extent we use a large NN-body simulation which we populate with galaxies following a semi-analytical model for galaxy formation. We find that if the orientation of the central galaxy is perfectly aligned with that of its dark matter halo, then the predicted central-satellite alignment signal is much stronger than observed. If, however, the minor axis of a central galaxy is perfectly aligned with the angular momentum vector of its dark matter halo, we can accurately reproduce the observed alignment strength as function of halo mass and galaxy color. Although this suggests that the orientation of central galaxies is governed by the angular momentum of their dark matter haloes, we emphasize that any other scenario in which the minor axes of central galaxy and halo are misaligned by ∼40∘\sim 40^{\circ} (on average) will match the data equally well. Finally, we show that dependence of the alignment strength on the color of the central galaxy is most likely an artefact due to interlopers in the group catalogue. The dependence on the color of the satellite galaxies, on the other hand, is real and owes to the fact that red satellites are associated with subhaloes that were more massive at their time of accretion.Comment: 13 Pages, 10 Figures, one figure replaced. added in discussion about comparison with others results, Updated version to match accepted version to MNRA

    Improved preservation effects of litchi fruit by combining chitosan coating with ascorbic acid treatment during postharvest storage

    Pericarp browning and aril decay of litchi fruits shorten post-harvest storage and thus reduce market value. Effects of chitosan coating and ascorbic acid (AsA) on litchi fruits storage were investigated in this paper. Based on the fact that AsA increases anti-oxidation capacity and chitosan inhibitsdehydration and microbial attack, the novel strategies of combining chitosan with AsA treatment were developed to improve litchi storability. By treating harvest fruits with 40 mmol/l AsA and 1.0% (w/v) chitosan solution, parameters of browning index and relative leakage rate and activities of polyphenol oxidase (PPO) and peroxidase (POD) in pericarp were markedly lowered in treated fruits. Moreover, increased activities of super oxide dismutase (SOD) and catalase (CAT) and contents of AsA andglutathione were observed in pulp of treated fruit, thus leading to lowered contents of hydrogen peroxide (H2O2) and malondialdehyde (MDA). Correspondingly, total soluble solids, soluble sugar and titratable acidity were significantly increased and thus lowered decay rate was achieved. It was suggested that chitosan and AsA play active roles in inhibiting pericarp browning, dehydration and microbial attack and maintaining membrane integrity, thus improved litchi storability being achieved
