In this paper, we present the development of an impedance-based method for detecting T. pallidum antigens and antibodies as an auxiliary tool for syphilis laboratory diagnosis. We evaluate the voltammetric signal obtained after incubation in carbon or gold nanoparticle-modified carbon electrodes in the presence or absence of Poly-L-Lysine. Our results indicate that the signal obtained from the electrodes was sufficient to distinguish between infected and non-infected samples immediately (T0′) or 15 min (T15′) after incubation, indicating its potential use as a point-of-care method as a screening strategy.publishersversionpublishe

    Structural Changes of the Paraflagellar Rod during Flagellar Beating in Trypanosoma cruzi

    , the agent of Chagas disease, is a protozoan member of the Kinetoplastidae family characterized for the presence of specific and unique structures that are involved in different cell activities. One of them is the paraflagellar rod (PFR), a complex array of filaments connected to the flagellar axoneme. Although the function played by the PFR is not well established, it has been shown that silencing of the synthesis of its major proteins by either knockout of RNAi impairs and/or modifies the flagellar motility.Here, we present results obtained by atomic force microscopy (AFM) and transmission electron microscopy (TEM) of replicas of quick-frozen, freeze-fractured, deep-etched and rotary-replicated cells to obtain detailed information of the PFR structures in regions of the flagellum in straight and in bent state. The images obtained show that the PFR is not a fixed and static structure. The pattern of organization of the PFR filament network differs between regions of the flagellum in a straight state and those in a bent state. Measurements of the distances between the PFR filaments and the filaments that connect the PFR to the axoneme as well as of the angles between the intercrossed filaments supported this idea.Graphic computation based on the information obtained allowed the proposal of an animated model for the PFR structure during flagellar beating and provided a new way of observing PFR filaments during flagellar beating

    Investigating the seismic response of URM walls with irregular opening layout through different modeling approaches

    TThe façade and internal walls of unreinforced masonry (URM) buildings often present an irregular opening layout, due to architectural reasons or modifications to the structure, which make the expected seismic damage pattern less predictable a priori. Therefore, the discretization of the walls in structural components is not standardized, conversely to cases with a regular opening layout for which the available modeling methods are corroborated by seismic damage surveys reporting recurrent failure patterns. The structural component discretization is a relevant step for the code-conforming seismic assessment, typically based on comparing the internal forces and drifts of each component to strength criteria and drift thresholds. Therefore, the lack of well-established approaches can significantly influence the assessment. The issue is even more evident when the structural components must be identified a priori in the modeling stage, namely for equivalent frame models. The applicability of available methods for discretization of URM walls with irregular opening layout has been already investigated in literature, but a conclusive judgment requires further studies. In this context, this paper presents an overview of the preliminary results addressing the numerical modeling of this type of walls within the framework of the DPC-ReLUIS 2022-2024 project (Subtask 10.3), funded by the Italian Department of Civil Protection. The Subtask aims to propose consensus-based recommendations for researchers and practitioners which can contribute to harmonize the use of different modeling approaches. Seven research groups are involved in the research, adopting different modeling approaches and computer codes, but similar assumptions and the same analysis method (pushover) are used. The benchmark URM structure illustrated in the paper is a two-story wall from which four configurations with increasing irregularity of opening layout were derived. The results of four modeling approached are presented. Three of them reproduce the mechanical response of masonry at the material scale by means of FE models implemented in OpenSees, DIANA and Abaqus software, while the remaining approach describes the mechanical response of masonry at the macro-element scale in 3DMacro software. Results were compared in terms of capacity curves, predicted failure mechanisms and evolution of internal forces in piers. The adoption of consistent assumptions among the different approaches led to an overall agreement of predictions at both wall and pier scales, particularly in terms of damage pattern with higher concentration of damage at the ground story. Despite that, differences on the pushover curves have been highlighted.     Antitumor activity of photodynamic therapy performed with nanospheres containing zinc-phthalocyanine

    Abstract\ud \ud Background\ud The increasing incidence of cancer and the search for more effective therapies with minimal collateral effects have prompted studies to find alternative new treatments. Among these, photodynamic therapy (PDT) has been proposed as a very promising new modality in cancer treatment with the lowest rates of side effects, revealing itself to be particularly successful when the photosensitizer is associated with nanoscaled carriers. This study aimed to design and develop a new formulation based on albumin nanospheres containing zinc-phthalocyanine tetrasulfonate (ZnPcS4-AN) for use in the PDT protocol and to investigate its antitumor activity in Swiss albino mice using the Ehrlich solid tumor as an experimental model for breast cancer.\ud \ud \ud Methods\ud Ehrlich tumor’s volume, histopathology and morphometry were used to assess the efficacy of intratumoral injection of ZnPcS4-AN in containing tumor aggressiveness and promoting its regression, while the toxicity of possible treatments was assessed by animal weight, morphological analysis of the liver and kidneys, hemogram, and serum levels of total bilirubin, direct bilirubin, indirect bilirubin, aspartate aminotransferase (AST), alanine aminotransferase (ALT), gamma glutamyl transferase (GGT), alkaline phosphatase, creatinine and urea. In order to evaluate the efficacy of PDT, groups of animals treated with intratumoral injection of doxorubicin (Dox) were also investigated.\ud \ud \ud Results\ud Intratumoral injection of ZnPcS4-AN was found to be efficient in mediating PDT to refrain tumor aggressiveness and to induce its regression. Although tumor volume reduction was not significant, PDT induced a remarkable increase in the necrosis area seen in the tumor’s central region, as in other experimental groups, including tumor and Dox treated groups, but also in the tumor’s peripheral region. Further, PDT showed minimal adverse effects.     Euphol, a tetracyclic triterpene, from Euphorbia tirucalli induces autophagy and sensitizes temozolomide cytotoxicity on glioblastoma cells

    Glioblastoma (GBM) is the most frequent and aggressive type of brain tumor. There are limited therapeutic options for GBM so that new and effective agents are urgently needed. Euphol is a tetracyclic triterpene alcohol, and it is the main constituent of the sap of the medicinal plant Euphorbia tirucalli. We previously identified anti-cancer activity in euphol based on the cytotoxicity screening of 73 human cancer cells. We now expand the toxicological screening of the inhibitory effect and bioactivity of euphol using two additional glioma primary cultures. Euphol exposure showed similar cytotoxicity against primary glioma cultures compared to commercial glioma cells. Euphol has concentration-dependent cytotoxic effects on cancer cell lines, with more than a five-fold difference in the IC50 values in some cell lines. Euphol treatment had a higher selective cytotoxicity index (0.64-3.36) than temozolomide (0.11-1.13) and reduced both proliferation and cell motility. However, no effect was found on cell cycle distribution, invasion and colony formation. Importantly, the expression of the autophagy-associated protein LC3-II and acidic vesicular organelle formation were markedly increased, with Bafilomycin A1 potentiating cytotoxicity. Finally, euphol also exhibited antitumoral and antiangiogenic activity in vivo, using the chicken chorioallantoic membrane assay, with synergistic temozolomide interactions in most cell lines. In conclusion, euphol exerted in vitro and in vivo cytotoxicity against glioma cells, through several cancer pathways, including the activation of autophagy-associated cell death.     The national inventory of geological heritage: methodological approach and results

    A existência de um inventário nacional de património geológico é fundamental para se poderem implementar estratégias de geoconservação. Este trabalho apresenta a metodologia usada no desenvolvimento do mais completo inventário de geossítios, realizado até ao momento em Portugal, assim como os principais resultados obtidos. O inventário vai integrar o Sistema de Informação do Património Natural e o Cadastro Nacional dos Valores Naturais Classificados, ambos geridos pelo Instituto de Conservação da Natureza e da Biodiversidade.The existence of a national inventory of the geological heritage is of paramount importance for the implementation of a geoconservation strategy. This paper presents the methodological approach used to produce the most complete geosites inventory in Portugal, so far, and the obtained results.
