    Data preprocessing for artificial neural network applications in prioritizing railroad projects â a practical experience in Taiwan

    [[abstract]]Financial constraints necessitate the tradeoff among proposed railroad projects, so that the project priorities for implementation and budget allocation need to be determined by the ranking mechanisms in the government. At present, the Taiwan central government prioritizes funding allocations primarily using the analytic hierarchy process (AHP), a methodology that permits the synthesizing of subjective judgments systematically and logically into objective consensus. However, due to the coopetition and heterogeneity of railway projects, the proper priorities of railroad projects could not be always evaluated by the AHP. The decision makers prefer subjective judgments to referring to the AHP evaluation re- sults. This circumstance not only decreased the AHP advantages, but also raised the risk of the policies. A method to con- sider both objective measures and subjective judgments of project attributes can help reduce this problem. Accordingly, combining the AHP with the artificial neural network (ANN) methodologies would theoretically be a proper solution to bring a ranking predication model by creating the obscure relations between objective measures by the AHP and subjec- tive judgments. However, the inconsistency between the AHP evaluation and subjective judgments resulted in the inferior soundness of the AHP/ANN ranking forecast model. To overcome this problem, this study proposes the data prepro- cessing method (DPM) to calculate the correlation coefficient value using the subjective and objective ranking incidence matrixes; according to the correlation coefficient value, the consistency between the AHP rankings and subjective judg- ments of railroad projects can be evaluated and improved, so that the forecast accuracy of the AHP/ANN ranking forecast model can also be enhanced.     Aerosolized amphotericin B as prophylaxis for invasive pulmonary aspergillosis: a meta-analysis

    SummaryObjectivesInvasive pulmonary aspergillosis (IPA) is associated with high mortality in high-risk (immunosuppressed) patients. Many studies have investigated whether prophylactic inhalation of amphotericin B (AMB) reduces the incidence of IPA, but no definitive conclusions have been reached. The present meta-analysis was performed to evaluate the efficacy of prophylactic inhalation of AMB for the prevention of IPA.MethodsMEDLINE and other databases were searched for relevant articles published until December 2013. Randomized controlled trials that compared aerosolized AMB with placebo were included. Two reviewers independently assessed and extracted the data of all trials.ResultsSix animal studies and two clinical trials involving 768 high-risk patients were eligible. The animal studies showed lower overall mortality rate among animals that underwent aerosolized AMB prophylaxis (odds ratio (OR) 0.13, 95% confidence interval (CI) 0.08–0.21). Similarly, the clinical trials showed a lower incidence of IPA among patients who underwent aerosolized AMB prophylaxis (OR 0.42, 95% CI 0.22–0.79).ConclusionsThis analysis provides evidence supporting the notion that the prophylactic use of aerosolized AMB effectively reduces the incidence of IPA among high-risk patients

    Research on a Pulmonary Nodule Segmentation Method Combining Fast Self-Adaptive FCM and Classification

    The key problem of computer-aided diagnosis (CAD) of lung cancer is to segment pathologically changed tissues fast and accurately. As pulmonary nodules are potential manifestation of lung cancer, we propose a fast and self-adaptive pulmonary nodules segmentation method based on a combination of FCM clustering and classification learning. The enhanced spatial function considers contributions to fuzzy membership from both the grayscale similarity between central pixels and single neighboring pixels and the spatial similarity between central pixels and neighborhood and improves effectively the convergence rate and self-adaptivity of the algorithm. Experimental results show that the proposed method can achieve more accurate segmentation of vascular adhesion, pleural adhesion, and ground glass opacity (GGO) pulmonary nodules than other typical algorithms

    Electronic reading is a process of digitalizing not only the content and reading devices but also the publication and distribution of books. With the development of e-books, more attention has been paid to use of e-books in education. Through online communities and innovative learning models, digital content providers can offer plenty of resources to help learners at all levels and promote the effectiveness of their self-regulated learning. The objective of this study is to build a cloud-based learning platform that integrates designs of electronic picture books and the social functions of Facebook. Students of multimedia design can upload their works to this platform and learn from each other through idea sharing and discussion. This platform is expected to help students create works with better quality and more creativity. Besides, this study also investigates the learning behavior and use intentions of students on this platform based on “Social Interaction Theory” and “Uses and Gratifications Theory”. This study attempts to capture students’ needs and how they are related to their learning outcomes. Results of this study contribute to the development of cloud-based learning and can be a reference for design of learning materials and improvement of teaching models

    FIP200 Claw Domain Binding to p62 Promotes Autophagosome Formation at Ubiquitin Condensates

    The autophagy cargo receptor p62 facilitates the condensation of misfolded, ubiquitin-positive proteins and their degradation by autophagy, but the molecular mechanism of p62 signaling to the core autophagy machinery is unclear. Here, we show that disordered residues 326–380 of p62 directly interact with the C-terminal region (CTR) of FIP200. Crystal structure determination shows that the FIP200 CTR contains a dimeric globular domain that we designated the “Claw” for its shape. The interaction of p62 with FIP200 is mediated by a positively charged pocket in the Claw, enhanced by p62 phosphorylation, mutually exclusive with the binding of p62 to LC3B, and it promotes degradation of ubiquitinated cargo by autophagy. Furthermore, the recruitment of the FIP200 CTR slows the phase separation of ubiquitinated proteins by p62 in a reconstituted system. Our data provide the molecular basis for a crosstalk between cargo condensation and autophagosome formation

    Induction of apoptosis by the retinoid inducible growth regulator RIG1 depends on the NC motif in HtTA cervical cancer cells

    <p>Abstract</p> <p>Background</p> <p>Retinoid-inducible gene 1 (RIG1), also known as tazarotene-induced gene 3 or retinoic-acid receptor responder 3, is a growth regulator, which induces apoptosis and differentiation. RIG1 is classified into the NC protein family. This study investigated functional domains and critical amino acids associated with RIG1-mediated cell death and apoptosis.</p> <p>Results</p> <p>Using enhanced green fluorescence protein (EGFP)-tagged RIG1 variants, RIG1 proteins with deletion at the NC domain significantly decreased cell death induced by RIG1, and fusion variants containing only the NC domain significantly induced apoptosis of HtTA cervical cancer cells. The EGFP-RIG1-induced apoptosis was significantly decreased in cells expressing N<sup>112</sup>C<sup>113 </sup>motif double- (NC→FG) or triple- (NCR→FGE) mutated RIG1 variants. Using dodecapeptides, nuclear localization and profound cell death was observed in HtTA cells expressing wild type RIG1<sub>111–123 </sub>or Leu<sup>121</sup>-mutated RIG1<sub>111–123</sub>:L→ C peptide, but peptides double- or triple-mutated at the NC motif alone, RIG1<sub>111–123</sub>:NC→FG or RIG1<sub>111–123</sub>:NCR→FGE, were cytoplasmically localized and did not induce apoptosis. The RIG1<sub>111–123 </sub>also induced apoptosis of A2058 melanoma cells but not normal human fibroblasts.</p> <p>Conclusion</p> <p>The NC domain, especially the NC motif, plays the major role in RIG1-mediated pro-apoptotic activity. The RIG1<sub>111–123 </sub>dodecapeptide exhibited strong pro-apoptotic activity and has potential as an anticancer drug.</p

    Acceleration of on-axis and ring-shaped electron beams in wakefields driven by Laguerre-Gaussian pulses

    The acceleration of electron beams with multiple transverse structures in wakefields driven by Laguerre-Gaussian pulses has been studied through three-dimensional (3D) particle-in-cell simulations. Under different laser-plasma conditions, the wakefield shows different transverse structures. In general cases, the wakefield shows a donut-like structure and it accelerates the ring-shaped hollow electron beam. When a lower plasma density or a smaller laser spot size is used, besides the donut-like wakefield, a central bell-like wakefield can also be excited. The wake sets in the center of the donut-like wake. In this case, both a central on-axis electron beam and a ring-shaped electron beam are simultaneously accelerated. Further, reducing the plasma density or laser spot size leads to an on-axis electron beam acceleration only. The research is beneficial for some potential applications requiring special pulse beam structures, such as positron acceleration and collimation

    Quercetin inhibits porcine intestinal inflammation in vitro

    Purpose: To investigate the effect of Quercetin (Que) on inflammatory reaction in intestinal porcine enterocyte cells, IPEC-J2, induced by lipopolysaccharide (LPS).Methods: IPEC-J2 cells were pretreated with Que and then incubated with LPS. Cell viability, cell morphology, nitric oxide (NO) content, gene expression levels of interleukin-6 (IL-6) and interleukin-8 (IL-8), were then evaluated.Results: LPS incubation at 10 μg/mL for 24 h showed no effect on the viability and cell morphology of IPEC-J2, while Que pre-incubation significantly enhanced the viability of IPEC-J2 and improved cell morphology (p &lt; 0.05). Que decreased the expression levels of pro-inflammatory cytokines IL-6 mRNA and IL-8 mRNA raised by LPS (p &lt; 0.05). Interestingly, both LPS and Que demonstrated no influence on the release of NO.Conclusion: Pre-treatment of Que shows a positive effect on intestinal porcine enterocyte cells and inhibited porcine intestinal inflammation in vitro induced by LPS, but the mechanism may be not associated with NO-related signaling pathway. Therefore, Que might have a potential effect as a veterinary drug or feed additive for the treatment of enteritis in pigs.Keywords: Quercetin, Inflammatory cytokines, Intestinal porcine enterocyte cells, Lipopolysaccharid
