    Molecular evidence for the clonal origin of blast crisis in chronic myeloid leukaemia.

    Cytogenetic and enzymatic studies have shown that chronic myeloid leukemia (CML) represents the clonal proliferation of a pluripotent stem cell. The Philadelphia chromosome (Ph') is the characteristic karyotypic abnormality seen in this disease, although the exact role of this clonal marker in the pathogenesis of CML is uncertain. At a molecular level, the Ph' has recently been shown to represent the translocation of c-abl to a limited (breakpoint cluster region, bcr) on chromosome 22. We have used probes for the bcr gene to obtain molecular evidence for the clonal origin of blast crisis in 2 patient with CML. In both cases, the first with myeloid and the second with lymphoid blast crisis, there was rearrangement of the bcr gene. The patterns of rearrangement varied between patients but were identical when comparing acute and chronic phases within the same individual. As the Ph' translocation is thought to represent a random recombination event these data not only provide further evidence for the clonal origin of blast crisis in CML, but also suggest that in the second patient this translocation event had already occurred at the pluripotent stem cell

    Population adaptation for genetic algorithm-based cognitive radios

    Abstract — Genetic algorithms are best suited for optimization problems involving large search spaces. The problem space encountered when optimizing the transmission parameters of an agile or cognitive radio for a given wireless environment and set of performance objectives can become prohibitively large due to the high number of parameters and their many possible values. Recent research has demonstrated that genetic algorithms are a viable implementation technique for cognitive radio engines. However, the time required for the genetic algorithms to come to a solution substantionally increases as the system complexity grows. In this paper, we present a population adaptation technique for genetic algorithms that takes advantage of the information from previous cognition cycles in order to reduce the time required to reach an optimal decision. Our simulation results demonstrate that the amount of information from the previous cognition cycle can be determined from the environmental variation factor (EVF), which represents the amount of change in the environment parameters since the previous cognition cycle.     Suppression of Raf-1 kinase activity and MAP kinase signalling by RKIP

    Raf-1 phosphorylates and activates MEK-1, a kinase that activates the extracellular signal regulated kinases (ERK). This kinase cascade controls the proliferation and differentiation of different cell types. Here we describe a Raf-1-interacting protein, isolated using a yeast two-hybrid screen. This protein inhibits the phosphorylation and activation of MEK by Raf-1 and is designated RKIP (Raf kinase inhibitor protein). In vitro, RKIP binds to Raf-1, MEK and ERK, but not to Ras. RKIP co-immunoprecipitates with Raf-1 and MEK from cell lysates and colocalizes with Raf-1 when examined by confocal microscopy. RKIP is not a substrate for Raf-1 or MEK, but competitively disrupts the interaction between these kinases. RKIP overexpression interferes with the activation of MEK and ERK, induction of AP-1-dependent reporter genes and transformation elicited by an oncogenically activated Raf-1 kinase. Downregulation of endogenous RKIP by expression of antisense RNA or antibody microinjection induces the activation of MEK-, ERK- and AP-1-dependent transcription.     Design and Architectural Implications of a Spatial Information System

    Image analysis, at the higher levels, works with extracted regions and line segments and their properties, not with the original raster data. Thus a spatial information system must be able to store points, lines, and areas as well as their properties and interrelationships. In a previous paper (Shapiro and Haralick [22]), we proposed for this purpose an entity-oriented relational database system. In this paper we describe our first experimental spatial information system, which employs these concepts to store and retrieve watershed data for a portion of the state of Virginia. We describe the logical and physical design of the system and discuss the architectural implications
