    Energy and carbon performance of housing : upgrade analysis, energy labelling and national policy development

    The area of policy formulation for the energy/carbon performance of housing is coming under increasing focus. A major challenge is to account for the large variation within national housing stocks relative to factors such as location, climate, age, construction, previous upgrades, appliance use and heating/cooling system types. Existing policy oriented tools rely on static calculation models that have limited ability to represent building behaviour and the impact of future changes in climate and technology. The switch to detailed simulation tools to address these limitations in the context of policy development has hitherto been focussed on the modelling of a small number of representative designs rather than dealing with the spread inherent in large housing stocks. To address these challenges, the ESRU Domestic Energy Model (EDEM) has been developed as a Web based tool built on detailed simulation models that have been aligned with the outcomes of national house condition surveys. On the basis of pragmatic inputs, EDEM is able to determine energy use and carbon emissions at any scale – from an individual dwelling to national housing stocks. The model was used at the behest of the Scottish Building Standards Agency and South Ayrshire Council to determine the impact of upgrades and the deployment of new and renewable energy systems. EDEM was also used to rate the energy/carbon performance of individual dwellings as required by the EU Directive on the Energy Performance of Buildings (EU, 2002). This paper describes the EDEM methodology and presents the findings from applications at different scales

    Tv-RIO1 – an atypical protein kinase from the parasitic nematode Trichostrongylus vitrinus

    Background: Protein kinases are key enzymes that regulate a wide range of cellular processes, including cell-cycle progression, transcription, DNA replication and metabolic functions. These enzymes catalyse the transfer of phosphates to serine, threonine and tyrosine residues, thus playing functional roles in reversible protein phosphorylation. There are two main groups, namely eukaryotic protein kinases (ePKs) and atypical protein kinases (aPKs); RIO kinases belong to the latter group. While there is some information about RIO kinases and their roles in animals, nothing is known about them in parasites. This is the first study to characterise a RIO1 kinase from any parasite. Results: A full-length cDNA (Tv-rio-1) encoding a RIO1 protein kinase (Tv-RIO1) was isolated from the economically important parasitic nematode Trichostrongylus vitrinus (Order Strongylida). The uninterrupted open reading frame (ORF) of 1476 nucleotides encoded a protein of 491 amino acids, containing the characteristic RIO1 motif LVHADLSEYNTL. Tv-rio-1 was transcribed at the highest level in the third-stage larva (L3), and a higher level in adult females than in males. Comparison with homologues from other organisms showed that protein Tv-RIO1 had significant homology to related proteins from a range of metazoans and plants. Amino acid sequence identity was most pronounced in the ATP-binding motif, active site and metal binding loop. Phylogenetic analyses of selected amino acid sequence data revealed Tv-RIO1 to be most closely related to the proteins in the species of Caenorhabditis. A structural model of Tv-RIO1 was constructed and compared with the published crystal structure of RIO1 of Archaeoglobus fulgidus (Af-Rio1). Conclusion: This study provides the first insights into the RIO1 protein kinases of nematodes, and a foundation for further investigations into the biochemical and functional roles of this molecule in biological processes in parasitic nematodes

    Observing transiting planets with JWST -- Prime targets and their synthetic spectral observations

    The James Webb Space Telescope will enable astronomers to obtain exoplanet spectra of unprecedented precision. Especially the MIRI instrument may shed light on the nature of the cloud particles obscuring planetary transmission spectra in the optical and near-infrared. We provide self-consistent atmospheric models and synthetic JWST observations for prime exoplanet targets in order to identify spectral regions of interest and estimate the number of transits needed to distinguish between model setups. We select targets which span a wide range in planetary temperature and surface gravity, ranging from super-Earths to giant planets, and have a high expected SNR. For all targets we vary the enrichment, C/O ratio, presence of optical absorbers (TiO/VO) and cloud treatment. We calculate atmospheric structures and emission and transmission spectra for all targets and use a radiometric model to obtain simulated observations. We analyze JWST's ability to distinguish between various scenarios. We find that in very cloudy planets such as GJ 1214b less than 10 transits with NIRSpec may be enough to reveal molecular features. Further, the presence of small silicate grains in atmospheres of hot Jupiters may be detectable with a single JWST MIRI transit. For a more detailed characterization of such particles less than 10 transits are necessary. Finally, we find that some of the hottest hot Jupiters are well fitted by models which neglect the redistribution of the insolation and harbor inversions, and that 1-4 eclipse measurements with NIRSpec are needed to distinguish between the inversion models. Wet thus demonstrate the capabilities of JWST for solving some of the most intriguing puzzles in current exoplanet atmospheric research.     Analysis of dominant-negative mutations of the Caenorhabditis elegans let-60 ras gene

    The let-60 gene of Caenorhabditis elegans controls the choice between vulval and hypodermal differentiation in response to an inductive signal from the gonad. let-60 encodes a ras protein that acts downstream of the let-23 receptor tyrosine kinase in a signal transduction pathway. Dominant-negative mutations of let-60 [let-60(dn)] cause a reduction of the gene activity in let-60(dn)/+ heterozygotes and a vulva-less mutant phenotype. We have found that nine let-60(dn) mutations cause replacements of conserved residues. Four are in two novel positions; others are in positions known previously to cause dominant-negative mutations in mammalian cells. The locations of these lesions suggest that they disrupt the ability of the ras protein to bind guanine nucleotides. Four let-60(dn) mutant genes were introduced into wild-type animals in the form of extrachromosomal arrays and were found to generate three dominant phenotypes--lethality, vulva-less, or multivulva--depending on gene dose and alleles. The dominant lethality caused by high-dose transgenic let-60(dn) genes suggests a toxic effect of these mutant genes in early development. The dominant-negative effects of these mutations in heterozygotes are likely to be caused by competition between let-60(dn) and let-60(+) protein for a positive regulator. All let-60(dn) mutations interfere with let-60(+) activity, but some alleles have partial constitutive activity, suggesting that the ability to interact with the activator is separable from the ability to exert a physiological effect (stimulation of vulval differentiation). These dn mutations might be useful for interfering with ras-mediated signal transduction pathways in other multicellular organisms
