The language of internernet media focused on skateboarding
The aim of this study is to analyse language of internet media focused on skateboarding. An essential element of the introduction is a brief history of skateboarding globaly and here in the Czech Republic. The basis for the analysis of contemporary audiovisual clips also touches on the development of the distribution of skateboard videos. In this study I also mention the role of YouTube as a distributing tool and phenomenom of instant fame, which is not strong only in skateboarding, but it is a phenomenon across the full spectrum of society. The principal basis of this study is the definition of two print and later Internet traditions in skateboard journalism - Thrasher and Transworld Skateboarding magazine. The study also concerns itself with maping out the Czech media environment, where skateboarding has its own place. The genesis of skateboard media in the Czech Republic follows the formation of Thrasher and Transworld, which is a very interesting fact that is important for the general understanding of the topic. For deeper insight into skateboard subculture, I also mention the journalistic style of Czech skateboarding media. The Internet media has a special emphasis on visual content, more so than print media, due to traditional text being replaced by moving pictures, and therefore with help of..

Printed DIY media in information age: how Web 2.0 influenced culture of fanzines
The thesis aims to explore changes in the production of music fanzines brought about by the advent of new media. Fanzines are defined as nonofficial magazines published independently in compliance with the code of DIY ethics. The theoretical part of this work focuses on the history of fanzine production as well as on concepts of alternative media, which is necessary in order to examine the current state of the aforementioned alternative media sphere. Furthermore, the theoretical part describes fans' behaviour, as they are the most prominent representatives of active audiences. This behaviour is a key to understanding the motivation behind fanzine production as well as the nature of media communication in the cyberspace, where boundaries between producers and consumers of media messages disappear. The main part of this work analyses interviews with ten figures from the Czech community of both pre-internet authors and those of post-internet era, the latter being used to internet communication. Described discursive patterns reveal the existence of two separate worlds: traditional fanzine community and online environment, coexisting in mutual awareness. Samizdat in the post-digital era
Tato disertační práce zkoumá roli materiálu v šesti vybraných zinech ze subkulturních, uměleckých a aktivistických kruhů. Práce se zabývá tištěn konceptů alternativních médií, podává stručný přehled teoretických perspektiv na ziny od subkulturních studií přes fan studies a participativní kulturu. Kriticky se vůči konceptům h pohledů, převážně ukotvených ve starém (post marxistickém) materialismu: konkrétně o reflexi post které dovolují přehodnotit roli materiálu v produkci a distribuci zinů. Případová studie kombinuje důrazem na materiál s etnografií prostorů, kde se ziny tvoří a předávají. S produkce a snaží se dojít k radikální kontextualizaci provozu mezi těly materiály, stroji a nakonec kapitály (ekonomickými i subkulturními). Aparát umožňuje zkoumat proměňující se roli zinů v digitální době, kdy se z rozsévačů informací mění v emocí a doteku. Dále umožňuje argumentovat, že materi zinové kultuře všudypřítomný hlubší průzkum materiality odhalí nové souvislosti zinové tvorby.This dissertation examines the role of material in six selected zines from subcultural, artistic and activist circles. The work deals with printed micro media with a small reach and is based on the concepts of alternative media, gives a brief overview of theoretical perspectives on zines from subcultural studies, through fan studies and participatory culture. The thesis is critical of the concepts and calls for the enrichment of existing perspectives, mostly consolidated in old (post- marxist) materialism: specifically, for a reflection on post-digital culture and new materialism, which allows us to reconsider the role of material in the production and distribution of zines. The case study combines personal interviews and content analysis with an emphasis on material and ethnography of the spaces where zines are formed and transmitted. Based on Bourdieu's field theory, the study conceptualises the zine scene as a field of zine production and seeks to radically contextualise the traffic between bodies, spaces, paper and other materials, machines, and finally capital (economic and subcultural). The theoretical apparatus allows to examine the changing role of zines in the post-digital age when information shifts from sowers to platforms of emotion and touch. Title of Thesis Development and validation of HPLC methods for transport and metabolic studies The presented PhD thesis deals with the development and validation of chromatographic methods in bioanalysis of drugs and their metabolites. The work provides a theoretical introduction to the field of bioanalysis, using methods of liquid chromatography. Attention was paid to recent trends in the area, particularly with respect to the new types of stationary phases. In the experimental part of the thesis, reader is introduced to a development and validation of several bio-analytical methods using high performance liquid chromatography. The first method concerns with a determination of 5-fluorouracil and its metabolites by HPLC with gradient elution and UV/VIS detection on a column Luna PFP (2). Discussed are mainly specifics for the assay of this group of polar and poorly detectable compounds. The developed and validated method is being used for TDM of patients with rectal cancer. Nezbytnou součástí je úvod do skateboardového diskurzu zahrnující stručný nástin dějin skateboardingu jak ve světě, tak u nás. Nezbytným základem pro analýzu současných audiovizuálních klipů je část o vývoji distribuce skateboardových klipů. V práci se také zmíním o roli YouTube jakožto mediačního nástroje a jevu instantní slávy, což není jev týkající se pouze skateboardingu, ale jedná se o celospolečenský fenomén. Nosným základem práce je vymezení dvou tradic ve skateboardové žurnalistice - Thrasher magazine a Transworld Skateboarding magazine. Práce se zabývá také zmapováním české mediální krajiny, v níž má skateboarding své místo. Geneze skateboardových médií v České republice kopíruje vznik Thrasheru a Transworldu, což je velice zajímavý fakt pro celkové pochopení problematiky. Pro hlubší vhled do skateboardové subkultury se zmíním o publicistickém stylu českých médií zabývajících se skateboardingem. Skateboardová média kladou důraz na vizuální stránku mediálního obsahu a u síťových médií to platí nadvakrát, proto se dále pomocí kvalitativní sémiotické analýzy zaměřím na dekódování dvou audiovizuálních klipů skateboardové internetové žurnalistiky.
