    The Urumchi Unrest Revisited

    The violence in Xinjiang took place almost a month ago, but it continues to generate interesting commentary (see, for example, this thoughtful essay by Pallavi Aiyar). The early July events have also recently had two reverberations in Australia, as Jia Zhangke and two other Chinese filmmakers pulled out of a Melbourne film festival where a documentary expected to present a sympathetic view of one of the people Beijing blames for the unrest was to be shown, and then hackers attacked the festival’s website to protest that film’s inclusion in the line-up. In light of this, we asked James Millward, a leading specialist in the history of Xinjiang who has written about related issues for us before, to share with the readers of China Beat his take on what happened in early July and how it should be understood. The ugly mob violence that roiled the western Chinese city of Urumchi in Xinjiang on July 5th was rather quickly suppressed, and Urumchi is now quiet. Thanks to an unprecedented degree of openness to the international press, moreover, we have a better idea specifically what happened than we have for other such incidents in China. Students who are members of the Uyghur minority—a largely Muslim, Turkic-language speaking group who are natives of the Xinjiang region in far northwestern China—demonstrated on Sunday, July 5 to call for a more thorough investigation into a deadly brawl among Uyghur and Han workers that had occurred at a factory in Guangdong province the previous week. The demonstration turned violent, possibly while it was being repressed by security forces, and thousands of Uyghurs went on a rampage, attacking Hans and destroying property. By Monday, July 6, mobs of Han—the majority ethnic group in China—took to the streets armed with clubs, meat-cleavers and other makeshift weapons, seeking revenge. Police eventually calmed the situation with batons, tear-gas, firearms with live ammunition, curfews and mass arrests. At least 192 people died, and some 1000 were injured. Though we know the broad outlines of what happened, why it happened remains in dispute. The official story from the Xinjiang regional and Chinese authorities is that the riot was instigated by Rebiya Kadeer and the World Uyghur Congress, an umbrella group made up of overseas Uyghur organizations in Europe, America and Central Asia that claims to represent Uyghur interests internationally. (A Uyghur economist and outspoken blogger, Ilham Tohti, has also been blamed by Xinjiang authorities for inciting the riot, and has apparently been detained.) The PRC routinely claims that the WUC and Kadeer—a charismatic spokeswoman for the Uyghur cause who enjoys sympathy in the US Congress and EU parliament—is surreptitiously engaged in separatist and even terrorist activity. Some of the commentary in Western media has harkened back to the issue of alleged Uyghur jihadism, involvement with Al Qaeda, and terrorist plots—issues much discussed with regard to the Uyghurs who wound up in Guantanamo

    Developing Performance Indicators for Nature-Based Solution Projects in Urban Areas: The Case of Trees in Revitalized Commercial Spaces

    It is becoming increasingly important to audit nature-based solutions (NBS) projects to understand their utility in addressing urban sustainability goals. However, the ecological and social complexity of such projects makes it difficult to develop performance indicators. Focusing on specific case studies and specific natural elements could advance this area of research. Urban trees are a vital component of many NBS initiatives. Cities with ambitious tree-planting goals rely on urban revitalization to provide the conditions necessary to grow trees in highly urbanized areas, and in this way deploy NBS projects. We present a conceptual and methodological framework of case-specific performance indicators in the context of NBS projects. This framework addresses the type of parameters, measures, and data that could be considered when assessing small-scale, NBS-inspired, revitalization projects, taking the natural elements of these projects, in this case the trees, as the unit of assessment. Our framework integrates ecological, environmental, and social indicators of tree performance and was developed with the experience gained from on-going, multi-year research projects at two revitalization sites in Toronto, Canada, where street trees grew in engineered sub-surface habitats. The framework includes indicators related to: urban tree ecology; tree characteristics; soils; climate and atmosphere; built environment; tree planting, care, and maintenance; social characteristics of the urban space; and human decisions and governance. This study frames the need for interdisciplinarity and case specificity in the development of performance indicators for NBS projects

    A method for mapping the turbulence intensity and excess energy available to building mounted wind turbines over a UK City

    Assessing the potential of proposed urban wind installations is further hindered by insufficient assessments of both urban wind resource, and the effectiveness of commercial gust control solutions within built up areas. Evaluating the potential performance of wind turbines within the urban environment requires an estimation of the total energy that would be available to them were effective control systems to be used. This paper presents a methodology for estimating the excess energy content (EEC) present in the gusty urban wind, which is usually under represented when using assessments based only on mean wind speeds. The method is developed using high temporal resolution wind measurements from eight potential turbine sites within the urban and suburban environment. By assessing the relationship between turbulence intensities and the EEC, an analytical methodology for predicting the total wind energy available at a potential turbine site is proposed. Sensitivity analysis with respect to temporal data resolution on the predicted EEC is also demonstrated. The methodology is then integrated with an analytical methodology that was initially developed to predict mean wind speeds at different heights within a UK city based on detailed mapping of its aerodynamic characteristics. Additional estimates of turbulence intensities and EEC based on the current methodology allow a more complete assessment of the wind resource available. The methodology is applied to the UK city of Leeds as a case study and the potential to map turbulence intensities and the total kinetic energy available at different heights within a typical urban city is demonstrated

    Gender Specific Effects of Mood on Alcohol Seeking Behaviors: Preliminary Findings using Intravenous Alcohol Self-Administration

    Background Although negative mood has long been implicated in differences in alcohol seeking by men and women, little research has used precise, well-controlled laboratory experiments to examine how negative mood affects alcohol-seeking behaviors. Methods A total of 34 (19 women) community-dwelling, alcohol-using adults aged 21 to 32 (mean age = 24.86, SD = 3.40, 74.3% Caucasian; Alcohol Use Disorders Identification Test [AUDIT] = 10.1, SD = 3.4) completed 2 counterbalanced intravenous alcohol self-administration sessions: one under negative mood and one under neutral mood. Fourteen individuals (9 women; mean age = 25.00, SD = 2.77) participated in an alcohol “liking” experiment (i.e., free access [FA] drinking) and 20 individuals (10 women; mean age = 24.77, SD = 3.73) participated in an alcohol “wanting” experiment, in which gaining access to alcohol required progressively effortful work. There was no significant difference between men and women on the AUDIT, t(32) = −0.38, p = 0.71. Results Priming with negative mood induction caused a significant decrease in self-reported mood (mean change = −1.85, t(32) = −6.81, p < 0.001), as intended. In FA, negative mood was associated with a significantly increased peak breath alcohol concentration (BrAC; F = 9.41, p = 0.01), with a trend toward a greater effect in men than in women (F = 2.67, p = 0.13). Negative mood also had a significant effect on peak BrAC achieved in the progressive work paradigm (F = 5.28, p = 0.04), with a significantly stronger effect in men (F = 5.35, p = 0.03) than women; men also trended toward more consistent work for alcohol across both neutral and negative sessions. Conclusions These preliminary findings demonstrate a gender-specific response on how mood affects alcohol seeking and suggest gender-specific interventions to prevent mood-based alcohol consumption

    Monitoring changes in circulating tumour cells as a prognostic indicator of overall survival and treatment response in patients with metastatic melanoma

    Background: New effective treatments for metastatic melanoma greatly improve survival in a proportion of patients. However biomarkers to identify patients that are more likely to benefit from a particular treatment are needed. We previously reported on a multimarker approach for the detection of heterogenous melanoma circulating tumour cells (CTCs). Here we evaluated the prognostic value of this multimarker quantification of CTCs and investigated whether changes in CTC levels during therapy can be used as a biomarker of treatment response and survival outcomes.Methods: CTCs were captured by targeting the melanoma associated markers MCSP and MCAM as well as the melanoma stem cell markers ABCB5 and CD271. CTCs were quantified in 27 metastatic melanoma patients treated by surgery or with vemurafenib, ipilimumab or dacarbazine. Patients were enrolled prospectively and CTC counts performed at baseline (prior to treatment), during and after treatment.Results: Baseline CTC numbers were not found to be prognostic of overall survival nor of progression free survival. However, a low baseline CTC number was associated with a rapid response to vemurafenib therapy. A decrease in CTCs after treatment initiation was associated with response to treatment and prolonged overall survival in vemurafenib treated patients.Conclusions: Measuring changes in CTC numbers during treatment is useful for monitoring therapy response in melanoma patients and for providing prognostic information relating to overall survival. Further studies with larger sample sizes are required to confirm the utility of CTC quantification as a companion diagnostic for metastatic melanoma treatment

    Iterative in Situ Click Chemistry Assembles a Branched Capture Agent and Allosteric Inhibitor for Akt1

    We describe the use of iterative in situ click chemistry to design an Akt-specific branched peptide triligand that is a drop-in replacement for monoclonal antibodies in multiple biochemical assays. Each peptide module in the branched structure makes unique contributions to affinity and/or specificity resulting in a 200 nM affinity ligand that efficiently immunoprecipitates Akt from cancer cell lysates and labels Akt in fixed cells. Our use of a small molecule to preinhibit Akt prior to screening resulted in low micromolar inhibitory potency and an allosteric mode of inhibition, which is evidenced through a series of competitive enzyme kinetic assays. To demonstrate the efficiency and selectivity of the protein-templated in situ click reaction, we developed a novel QPCR-based methodology that enabled a quantitative assessment of its yield. These results point to the potential for iterative in situ click chemistry to generate potent, synthetically accessible antibody replacements with novel inhibitory properties
