    Sample preparation for nanoanalytical electron microscopy using the FIB lift-out method and low energy ion milling

    Thinning specimens to electron transparency for electron microscopy analysis can be done by conventional (2 - 4 kV) argon ion milling or focused ion beam (FIB) lift-out techniques. Both these methods tend to leave ''mottling'' visible on thin specimen areas, and this is believed to be surface damage caused by ion implantation and amorphisation. A low energy (250 - 500 V) Argon ion polish has been shown to greatly improve specimen quality for crystalline silicon samples. Here we investigate the preparation of technologically important materials for nanoanalysis using conventional and lift-out methods followed by a low energy polish in a GentleMill™ low energy ion mill. We use a low energy, low angle (6 - 8°) ion beam to remove the surface damage from previous processing steps. We assess this method for the preparation of technologically important materials, such as steel, silicon and GaAs. For these materials the ability to create specimens from specific sites, and to be able to image and analyse these specimens with the full resolution and sensitivity of the STEM, allows a significant increase of the power and flexibility of nanoanalytical electron microscopy

    Semiotics of alterity and the cultural dimensions of Bible translation

    Translated sacred writings from various religious traditions often retain a few selected cultural terms borrowed from the incipient sign system, while other cultural dimensions are translated in ways that can broadly be construed as domestication. By contrast, many Bible translation agencies eschew translation strategies in which cultural terms are borrowed, advocating in stead for wholesale domestication. In this article, we develop a theoretical framework for representing the alterity, but not the foreignness, of the Bible in translation. Alterity involves the incipient sign system, namely the biblical languages and their cultural contexts ranging from Iron Age Israel within the context of the Ancient Near East for the Old Testament to Roman Palestine in the first century for the New Testament. Examples from African contexts, including Afrikaans (South Africa), Lokaa (Nigeria) and Tira (Sudan), illustrate multiple approaches to representing alterity and provide an important corrective to current practice in many Bible translation projects.

    Omega-3 fatty acids are essential nutrients for humans, but dietary intake of these nutrients by many Americans is inadequate due to low consumption of omega-3-rich foods such as fish, walnuts, and flaxseed. In contrast, per capita consumption of red meat is relatively high, but these products normally contain only small amounts of omega-3 fatty acids. Feeding cattle diets that contain omega-3 fatty acids has consistently increased the proportion of the desirable fats that accumulate in beef. Unfortunately, the proportion of dietary omega-3 fats that are deposited into beef tissues is relatively low, because microorganisms within the rumen biohydrogenate the unsaturated omega-3 fatty acids extensively to produce the saturated fats that are characteristic of beef fat. Encapsulation of fats has been proposed as a method for improving efficiency of transfer of omega-3 fats into beef. Encapsulation processes apply a protective barrier on the surface of fats or fat-containing feeds, which theoretically decreases fats’ susceptibility to microbial biohydrogenation. Protective coatings must remain intact to retain their functionality, and physical damage to the coatings that occurs with normal handling can result in poor efficacy because the core material is exposed to microorganisms in the rumen. Embedding feed particles within a homogeneous protective matrix constitutes a potentially useful alternative to protective surface barriers. The matrix is created by mixing feed particles that are to be protected with a suitable matrix material that is resistant to microbial digestion and subsequently forming the mixture into pills. In cases where physical damage occurs, exposure of the core material is confined to the broken surface, and the remainder of the matrix retains its ruminal stability. The objective of this study was to determine if embedding flaxseed within a matrix of hydrated dolomitic lime could be used as a method to decrease biohydrogenation of polyunsaturated omega-3 fatty acids, thus improving efficiency of omega-3 fatty acids absorption into the bloodstream

    Orally dosing steers with Lactipro (Megasphaera elsdenii) decreases the quantity of roughages fed during finishing

    The cost of roughages is relatively high in comparison to their contribution of nutrients to feedlot diets. Widespread drought has affected roughage supplies in the cattle-feeding regions of the United States, further increasing the cost of roughages for feedlots. Despite their relatively high cost, roughages are incorporated into finishing diets to maintain rumen function and to manage ruminal acidosis. The greatest proportion of roughage is utilized early in the feeding period when cattle are being transitioned from forage-based diets to concentrate-based diets, allowing ruminal microbes to adapt gradually to higher levels of starch and sugars. If cattle are not properly adapted to concentrate-based diets, undesirable lactic acid-producing microorganisms, such as Streptococcus bovis, can rapidly proliferate and produce large excesses of lactic acid in the rumen. This condition can precipitate feedlot acidosis, which can have serious consequences for health and performance of cattle. Lactipro (MS Biotec, Wamego, KS) is a probiotic drench containing Megasphaera elsdenii, which is a lactate-utilizing bacterium that prevents lactic acid accumulations in the rumen of grain-fed cattle. Lactipro has been used successfully to accelerate the adaptation of cattle from roughage-based diets to concentrate-based diets. The objective of this study was to determine if Lactipro could be utilized to eliminate the step-up period for feedlot cattle, and in so doing decrease the amount of roughage required during the finishing period

    Chickpea in semiarid cropping systems

    Non-Peer ReviewedRelative to other pulse crops such as dry-pea (Pisum sativum L.) and lentil (Lens culinaris L.), chickpea (Cicer arietinum L.) is still new in western Canada. In 1999, there were 350,000 acres of chickpea seeded in Saskatchewan with more than 75% of the seeded area being located in the districts of Swift Current, Shaunavon, Moose Jaw, Rosetown, and Assiniboia (Noble 2000). Approximately 93% of the seeded chickpea area was harvested in Saskatchewan in 1999 (Statistics Canada, 1999). Due to abundant rainfall and generally cooler than normal growing conditions in 1999, some late-seeded chickpea fields did not reach full maturity. The best production success came in the southwest corner of Saskatchewan where the growing season (May to August) precipitation usually is less than 8 inches. The deeper rooting habit and the tolerance to water stress makes chickpea a winner under these adverse drought conditions. The objective of this study was to develop agronomic information for the inclusion of chickpea in cropping systems for the semiarid prairie region.     Dual-energy X-ray absorptiometry scans accurately predict differing body fat content in live sheep

    Background There is considerable interest in implementing mobile scanning technology for on-farm body composition analysis on live animals. These experiments evaluated the use of dual energy X-ray absorptiometry (DXA) as an accurate method of total body fat measurement in live sheep. Results In Exp. 1, visceral and whole body fat analysis was undertaken in sheep with body condition scores (BCS) in the range 2 to 3.25 (scale 1: thin to 5: fat). The relationship of BCS was moderately correlated with visceral fat depot mass (r = 0.59, P  0.05, n = 9). There was a moderate correlation between DXA body fat and BCS (r = 0.70, P < 0.01, n = 17), and DXA body fat was highly correlated with chemical body fat (r = 0.81, P < 0.001, n = 9). In Exp. 3, a series of five DXA scans, at 8-week intervals, was performed on growing sheep over a 32-week period. The average BCS ranged from 2.39 ± 0.07 (S.E.M.) to 3.05 ± 0.11 and the DXA body fat (%) ranged from 16.8 ± 0.8 to 24.2 ± 1.2. There was a moderate correlation between DXA body fat and BCS over the 32 weeks (r = 0.61, P < 0.001, n = 24). Conclusions Overall, these experiments indicated that there was good agreement between BCS, DXA and chemical analysis for measuring total body fat in sheep, and that DXA scanning is a valid method for longitudinal measurement of total body fat in live sheep

    Detecting a stochastic background of gravitational radiation: Signal processing strategies and sensitivities

    We analyze the signal processing required for the optimal detection of a stochastic background of gravitational radiation using laser interferometric detectors. Starting with basic assumptions about the statistical properties of a stochastic gravity-wave background, we derive expressions for the optimal filter function and signal-to-noise ratio for the cross-correlation of the outputs of two gravity-wave detectors. Sensitivity levels required for detection are then calculated. Issues related to: (i) calculating the signal-to-noise ratio for arbitrarily large stochastic backgrounds, (ii) performing the data analysis in the presence of nonstationary detector noise, (iii) combining data from multiple detector pairs to increase the sensitivity of a stochastic background search, (iv) correlating the outputs of 4 or more detectors, and (v) allowing for the possibility of correlated noise in the outputs of two detectors are discussed. We briefly describe a computer simulation which mimics the generation and detection of a simulated stochastic gravity-wave signal in the presence of simulated detector noise. Numerous graphs and tables of numerical data for the five major interferometers (LIGO-WA, LIGO-LA, VIRGO, GEO-600, and TAMA-300) are also given.     Optimum plant population density for chickpea in a semiarid environment

    Non-Peer ReviewedChickpea (Cicer arietinum L.), an annual grain legume, is being rapidly included in cereal-based cropping systems throughout the semiarid Canadian prairies, but information on optimum plant population density (PPD) has not been developed for this region. This study was conducted from 1998 to 2000 in southwestern Saskatchewan to determine the impact of PPD on field emergence, seed yield and quality, and harvestability of kabuli and desi chickpea compared with dry pea (Pisum sativum L.). Seed yields of all legumes increased with increasing PPD when the crops were grown on conventional summerfallow. The PPD that produced the highest seed yields ranged from 40 to 45 plants m-2 for kabuli chickpea, 45 to 50 plants m-2 for desi chickpea, and 75 to 80 plants m-2 for dry pea. When the legumes were grown on wheat stubble, the PPD that gain optimum seed yield ranged from 35 to 40 plants m-2 for kabuli, 40 to 45 plants m-2 for desi chickpea, and 65 to 70 plants m-2 for dry pea. The proportion of large-sized (>9-mm diameter) seed in the harvested seed was >70% when the kabuli chickpea was grown on summerfallow regardless of PPD, whereas the large-seed proportion decreased with increasing PPD when the crop was grown on wheat stubble. Increases in PPD advanced plant maturity by 1.5 to 3.0 days, and also increased the height of lowest pods from the soil surface by 1.4 to 2.0 cm (or 5 to 10%), with desi type receiving the greatest benefits from increased PPD
