Streszczenie:
Rozwój genetyki molekularnej umożliwił wytypowanie genów odpowiedzialnych
za regulacje wzrostu i rozwoju organizmu. Genem kandydującym, którego produkt białkowy uczestniczy w licznych procesach zachodzących w komórkach ciała, oddziałując
na wzrost i metabolizm mięśni szkieletowych, kości, chrząstek i tkanki tłuszczowej jest gen hormonu wzrostu (GH). Wielu badaczy podejmowało i nadal podejmuje trud ustalenia jego wpływu na cechy budowy ciała i użytkowości mięsnej. Liczne badania wskazują na istnienie zależności między polimorfizmem genu GH a produkcją mięsa oraz budową ciała m. in. u świń, bydła, owiec i kóz.

Summary:
The development of molecular genetics has allowed predicting genes responsible for regulating growth and development. Candidate gene whose protein product is involved
in many processes occurring in the cells of the body, affecting on the growth and metabolism
of the skeletal muscle, bone, cartilage and fat cells is a gene of growth hormone (GH). Many researchers undertook and continue to take the trouble to determine its impact on the features of body growth and meat production. Many study suggest a relationship between GH gene polymorphism and the features of meat production and growth traits in pigs, cattle, sheep and goats.

Streszczenie
Bydło mleczne jest hodowane głównie z uwagi na duże ilości pozyskiwanego mleka, które jest bogate w białko. Produkowane mleko jest ważne w diecie dzieci, młodzieży i osób starszych. Posiada ono niezbędne składniki do budowy kości i regeneracji komórek. Wydajność mleczna, razem z zawartością białka w mleku są najważniejszym parametrem klasyfikującym jego przydatność. Jednym z gatunków bydła mlecznego jest rasa Holsztyńsko – Fryzyjska. Obejmuje ono bydło czarno – białe i czerwono – białe pochodzenia krajowego i zagranicznego oraz potomstwo pochodzące z kojarzenia pomiędzy odmianami barwnymi. Badanie obejmowało grupę o odmianie HO – czyli bydło tylko odmiany czarno – białej. Ta grupa obejmowała dwa stada: jedno wolnowybiegowe, gdzie badaniami objęto 149 krów i drugie – alkierzowe o liczebności 150 sztuk. Średnia wydajność mleczna w pierwszej grupie to 8988 kg mleka w 305-dniowej laktacji, natomiast w drugiej grupie wynik ten kształtował się na poziomie 8421,74 kg mleka. Celem badania było wykazanie wpływu wybranych czynników na wydajność mleczną i zawartość białka w mleku. W tym doświadczeniu był to system utrzymania oraz kolejność laktacji.

Summary
Dairy cattle are breeding for the ability to produce large quantities of milk which is plentiful of protein. Produced milk is an important in the diet of children, youth and the elderly. It has necessary factors to build bones and regenerate cells. Milk yield, together with a protein content in milk is the most important parameter determining the profitability of farming. One of the species of dairy cattle are Holstein – Friesian cows. They are include black-and-white and red-and-white domestic and foreign origin and offspring from the mating between varieties of colored cattle. The research was conducted on herd HO variety of Holstein – Friesian breed – it means the only black-and-white cattle domestic and foreign origin and their offspring. This group consisted of two herds. One of it - 149 cows in a free range housing system and the second – 150 cows kept indoor. The average milk yield in first group is 8988 kg of milk per 305-day lactation and in the second group it’s 8421,74 kg of milk. The aim of this study is to demonstrate the influence of selected factors on milk yield and protein content in milk of cows. In this research as selected factors were housing system and lactation sequence.

Streszczenie
Obecnie konsumenci poszukują mleka o wysokiej zawartości białka oraz niskim poziomie tłuszczu. Stworzenie takiego produktu stawia przed hodowca nie lada wyzwanie. Opłacalna hodowla jest również związana z wysoka wydajnością mleczną. Jednakże pozyskanie znacznych ilości mleka o wysokiej jakości jest celem trudnym w realizacji. Badania dowodzą iż czynniki środowiskowe takiej jak system utrzymania, żywienie czy kolejna laktacja mogą w znacznym stopniu wpływać na ilość oraz jakość pozyskiwanego mleka. W przypadku odpowiedniego żywienia mleko może być wzbogacone o substancje korzystne dla zdrowia. Wraz z rozwojem nauki możliwości oceny zwierząt pod względem ich produkcyjności również rosną. Cechy związane z mlecznością są uwarunkowane genetycznie. Badania nad polimorfizmem określonych genów pozwala na określenie tych genotypów które warunkują najkorzystniejszą wartość cechy.

Summary
Currently, consumers are looking for milk with a high protein content and a low level of fat. The creation of such a product is great challenge for a breeder. Profitable animal husbandry is also associated with high milk yield. However, obtaining significant quantities of high quality milk is a difficult goal to achieve. Research shows that environmental factors such as system maintenance, nutrition or another lactation can significantly affect the quantity and quality of the acquired milk. In the case of adequate nutrition milk it may be enriched with substances beneficial to health. With the development of science the possibility to assess the animals for their productivity also increase. Features associated with milk production are genetically determined. Research on polymorphism of certain genes allows for identification of those genotypes that determine the most favorable value characteristics.

Wpływ wybranych czynników środowiskowych oraz polimorfizmu wybranych genów na wydajność mleczną oraz skład chemiczny mleka krów rasy polskiej Holsztyńsko-Fryzyjskiej
Doskonalenie cech produkcyjnych bydła mlecznego stanowi jeden z głównych
trendów nowoczesnej hodowli. Kierunkiem badań stają się coraz częściej
genetyczne uwarunkowania takich cech jak wydajność mleczna, poziom
białka czy tłuszczu w mleku. Nieustannie rosnące wymagania konsumentów,
dotyczących jakości spożywanego mleka i jego przetworów stawiają wysoko
poprzeczkę hodowcom oraz producentom. Najważniejszymi doskonalonymi
cechami są: wydajność mleczna, poziom białka oraz tłuszczu. Wpływ na nie
mają nie tylko czynniki środowiskowe ale również uwarunkowania genetyczne.
Badania prowadzone nad polimorfizmem genów hormonu wzrostu, IGF-
1 czy PIT-1 mają znaczący wpływ na kształtowanie się cech produkcyjnych
u bydła. Ponadto ważnym aspektem są również czynniki środowiska takie jak:
sezon utrzymania, kolejna laktacja, żywienie czy sezon produkcyjny również
w znacznym stopniu decydują o ilości oraz jakości otrzymywanego surowca
Współczesne trendy konsumenckie a jakość mleka krów rasy Holsztyńsko–Fryzyjskiej
Współcześnie pożądane jest mleko wysokiej jakości, zarówno do bezpośredniego
spożycia, jak i do produkcji wysokogatunkowych przetworów
mlecznych. Przez wysoką jakość rozumie się mleko o dużej zawartości
białka przy zmniejszonej zawartości tłuszczu. Istnieje wiele metod hodowlanych,
które sprzyjają produkcji mleka dobrej jakości. Oprócz metod
hodowlanych, wpływ na skład mleka może mieć także system utrzymania,
czy kolejność laktacji
Duplicate divergence of two bacterial small heat shock proteins reduces the demand for Hsp70 in refolding of substrates.
Small heat shock proteins (sHsps) are a conserved class of ATP-independent chaperones that bind to aggregation-prone polypeptides at stress conditions. sHsps encage these polypeptides in assemblies, shielding them from further aggregation. To facilitate their subsequent solubilization and refolding by Hsp70 (DnaK) and Hsp100 (ClpB) chaperones, first, sHsps need to dissociate from the assemblies. In most γ-proteobacteria, these functions are fulfilled by a single sHsp (IbpA), but in a subset of Enterobacterales, a two-protein sHsp (IbpA and IbpB) system has evolved. To gain insight into the emergence of complexity within this chaperone system, we reconstructed the phylogeny of γ-proteobacteria and their sHsps. We selected proteins representative of systems comprising either one or two sHsps and analysed their ability to form sHsps-substrate assemblies. All the tested IbpA proteins, but not IbpBs, stably interact with an aggregating substrate. Moreover, in Escherichia coli cells, ibpA but not ibpB suppress the growth defect associated with low DnaK level, which points to the major protective role of IbpA during the breakdown of protein quality control. We also examined how sHsps affect the association of Hsp70 with the assemblies at the initial phase of disaggregation and how they affect protein recovery after stress. Our results suggest that a single gene duplication event has given rise to the sHsp system consisting of a strong canonical binder, IbpA, and its non-canonical paralog IbpB that enhances sHsps dissociation from the assemblies. The cooperation between the sHsps reduces the demand for Hsp70 needed to outcompete them from the assemblies by promoting sHsps dissociation without compromising assembly formation at heat shock. This potentially increases the robustness and elasticity of sHsps protection against irreversible aggregation
Wpływ polimorfizmu genu kalpastatyny na wybrane parametry użytkowości mięsnej owiec
Ze względu na fakt, iż dla gospodarki duże znaczenie zaczyna odgrywać
mięsny kierunek użytkowania owiec, niezbędna jest wiedza dotycząca
uwarunkowań genetycznych cech wpływających na jakość mięsa jagnięcego.
Gen kodujący kalpastatynę został wytypowany jako gen kandydujący
dla cech użytkowości mięsnej owiec. Badania nad polimorfizmem tego
genu prowadzone były na wielu rasach owiec w Europie, Azji i Ameryce
Północnej. Powiązano zaobserwowany polimorfizm z masą urodzeniową,
dziennymi przyrostami masy ciała, masą jagniąt po odsadzeniu oraz masą
mięśni zwierząt. Równocześnie zaobserwowano związek tego genu z takimi
cechami mięsa jak: zawartość tłuszczu śródmięśniowego, kruchość oraz
barwa. Świadomość i struktura związku między masą ciała oraz cechami
mięsa owczego a polimorficznymi postaciami genu kalpastatyny może
prowadzić do wyznaczenia markerów genetycznych, które będą mogły być
wykorzystywane jako dodatkowe kryterium selekcji owiec ras mięsnych
Two-step mechanism of J-domain action in driving Hsp70 function.
J-domain proteins (JDPs), obligatory Hsp70 cochaperones, play critical roles in protein homeostasis. They promote key allosteric transitions that stabilize Hsp70 interaction with substrate polypeptides upon hydrolysis of its bound ATP. Although a recent crystal structure revealed the physical mode of interaction between a J-domain and an Hsp70, the structural and dynamic consequences of J-domain action once bound and how Hsp70s discriminate among its multiple JDP partners remain enigmatic. We combined free energy simulations, biochemical assays and evolutionary analyses to address these issues. Our results indicate that the invariant aspartate of the J-domain perturbs a conserved intramolecular Hsp70 network of contacts that crosses domains. This perturbation leads to destabilization of the domain-domain interface-thereby promoting the allosteric transition that triggers ATP hydrolysis. While this mechanistic step is driven by conserved residues, evolutionarily variable residues are key to initial JDP/Hsp70 recognition-via electrostatic interactions between oppositely charged surfaces. We speculate that these variable residues allow an Hsp70 to discriminate amongst JDP partners, as many of them have coevolved. Together, our data points to a two-step mode of J-domain action, a recognition stage followed by a mechanistic stage
Structure and evolution of the 4-helix bundle domain of Zuotin, a J-domain protein co-chaperone of Hsp70.
The J-domain protein Zuotin is a multi-domain eukaryotic Hsp70 co-chaperone. Though it is primarily ribosome-associated, positioned at the exit of the 60S subunit tunnel where it promotes folding of nascent polypeptide chains, Zuotin also has off-ribosome functions. Domains of Zuotin needed for 60S association and interaction with Hsp70 are conserved in eukaryotes. However, whether the 4-helix bundle (4HB) domain is conserved remains an open question. We undertook evolutionary and structural approaches to clarify this issue. We found that the 4HB segment of human Zuotin also forms a bundle of 4 helices. The positive charge of Helix I, which in Saccharomyces cerevisiae is responsible for interaction with the 40S subunit, is particularly conserved. However, the C-termini of fungal and human 4HBs are not similar. In fungi the C-terminal segment forms a plug that folds back into the bundle; in S. cerevisiae it plays an important role in bundle stability and, off the ribosome, in transcriptional activation. In human, C-terminal helix IV of the 4HB is extended, protruding from the bundle. This extension serves as a linker to the regulatory SANT domains, which are present in animals, plants and protists, but not fungi. Further analysis of Zuotin sequences revealed that the plug likely arose as a result of genomic rearrangement upon SANT domain loss early in the fungal lineage. In the lineage leading to S. cerevisiae, the 4HB was subjected to positive selection with the plug becoming increasingly hydrophobic. Eventually, these hydrophobic plug residues were coopted for a novel regulatory function-activation of a recently emerged transcription factor, Pdr1. Our data suggests that Zuotin evolved off-ribosome functions twice-once involving SANT domains, then later in fungi, after SANT domain loss, by coopting the hydrophobic plug.
