Chondrolipoma in the Pelvic Cavity: a Case Report
A chondrolipoma is an extremely rare form of a benign mesenchymal tumor containing mature cartilage and fatty tissue. Chondrolipomas may be found in almost any part of the body, particularly in the connective tissue of the breast, head and neck area, as well as in the skeletal muscle. However, to the best of our knowledge, chondrolipomas located in the pelvic cavity have not been reported. In this case report, we describe a case of a chondrolipoma in the pelvis, and show that it has its own characteristic imaging findings, which included the composition of fatty tissue and calcification in most parts, as well as some focal areas of chondroid tissue based on the CT and MR findings
Three new chondrosarcoma cell lines: one grade III conventional central chondrosarcoma and two dedifferentiated chondrosarcomas of bone
BackgroundChondrosarcoma is the second most common primary sarcoma of bone. High-grade conventional chondrosarcoma and dedifferentiated chondrosarcoma have a poor outcome. In pre-clinical research aiming at the identification of novel treatment targets, the need for representative cell lines and model systems is high, but availability is scarce.MethodsWe developed and characterized three cell lines, derived from conventional grade III chondrosarcoma (L835), and dedifferentiated chondrosarcoma (L2975 and L3252) of bone. Proliferation and migration were studied and we used COBRA-FISH and array-CGH for karyotyping and genotyping. Immunohistochemistry for p16 and p53 was performed as well as TP53 and IDH mutation analysis. Cells were injected into nude mice to establish their tumorigenic potential.ResultsWe show that the three cell lines have distinct migrative properties, L2975 had the highest migration rate and showed tumorigenic potential in mice. All cell lines showed chromosomal rearrangements with complex karyotypes and genotypic aberrations were conserved throughout late passaging of the cell lines. All cell lines showed loss of CDKN2A, while TP53 was wild type for exons 5â8. L835 has an IDH1 R132C mutation, L2975 an IDH2 R172W mutation and L3252 is IDH wild type.ConclusionsBased on the stable culturing properties of these cell lines and their genotypic profile resembling the original tumors, these cell lines should provide useful functional models to further characterize chondrosarcoma and to evaluate new treatment strategies
Principal Curves in High Dimensions
Thesis (Ph.D.)--University of Washington, 2021Principal curves are curves passing through the "center" of a dataset and generalize the notion of principal components to non-linear curves, thus providing more meaningful insights to the structure of the data. SCMS is an elegant and robust method for recovering the principal curves of a dataset. However, its dependence on evaluating the Hessian of the density function at every step and the need for an abundance of probes make it prohibitively computationally intensive when dealing with large and high-dimensional data. In this paper we present L-SCMS, an approximation algorithm for SCMS which reduces the computational complexity from n^2 to n, where n is the dimensionality of the data. We also present MorsePCS, an extension to SCMS that provides with additional information on the structure of the data, while reducing the number of steps required. Finally, we provide empirical evaluation of our MorsePCS, indicating that the runtime guarantees do realize in practice, while maintaining the quality of the curves produce
STLS: Cycle-Cutset-Driven Local Search For MPE
In this paper we present Stochastic Tree-based Local Search or STLS, a local search algorithm combining the notion of cycle-cutsets with the well-known Belief Propagation to approximatethe optimum of sums of unary and binary potentials. This is done by the previously unexplored concept oftraversal from one cutset to another and updating the induced forest, thus creating a local search algorithm, whose updatephase spans over all the forest variables. Intraductal Carcinoma of the Pancreas
Four multicentric intraductal papillary carcinomas arising in the main pancreatic duct are presented. Three of the neoplasms showed stromal invasion and metastasized to regional lymph nodes. Three patients had a long history of epigastric pain, confirming the progressive slow growth and less aggressive nature of this clinicoâpathologic entity. Histologically, all tumors were papillary, and three also showed a pseudocribriform pattern. Individual cells exhibited a range of atypia from mild to overt malignant change. Focal intestinalâdifferentiation was recognized in two tumors. Despite the wellâdifferentiated appearance of these tumors, two patients died within 1 year of surgery. One patient with an entirely intraductal carcinoma is alive and well 3 years after surgical treatment. The fourth patient who had lymph node metastasis is alive 6 months after a Whipple's procedure
