    Utility of accelerometers to measure physical activity in children attending an obesity treatment intervention

    Objectives. To investigate the use of accelerometers to monitor change in physical activity in a childhood obesity treatment intervention. Methods. 28 children aged 7–13 taking part in “Families for Health” were asked to wear an accelerometer (Actigraph) for 7-days, and complete an accompanying activity diary, at baseline, 3-months and 9-months. Interviews with 12 parents asked about research measurements. Results. Over 90% of children provided 4 days of accelerometer data, and around half of children provided 7 days. Adequately completed diaries were collected from 60% of children. Children partake in a wide range of physical activity which uniaxial monitors may undermonitor (cycling, nonmotorised scootering) or overmonitor (trampolining). Two different cutoffs (4 METS or 3200 counts⋅min-1) for minutes spent in moderate and vigorous physical activity (MVPA) yielded very different results, although reached the same conclusion regarding a lack of change in MVPA after the intervention. Some children were unwilling to wear accelerometers at school and during sport because they felt they put them at risk of stigma and bullying. Conclusion. Accelerometers are acceptable to a majority of children, although their use at school is problematic for some, but they may underestimate children's physical activity

    Behavioral Couple Therapy: Partner-Involved Treatment for Substance-Abusing Women

    Among the various psychosocial interventions presently available to treat alcohol and drug abuse, it could be argued that partner-involved treatments are the most broadly efficacious. There is not only substantial empirical support for the use of couple-based treatments in terms of improvements in primary targeted outcomes, such as substance use and relationship adjustment, but also in other areas that are of clear public health significance, including intimate partner violence (IPV), children\u27s adjustment, and cost-benefit ratio and cost-effectiveness. During the last few decades, programmatic research on the application of partner-involved therapies for substance abuse has been among the most active and fruitful. Although marital and family therapies for substance abuse have been used with a wide variety of patient populations, the purpose of this chapter is to focus on the application of partner-involved interventions with women who abuse substances and are in intimate relationships. More specifically, we (1) provide a conceptual rationale as to why couple therapy for female patients with substance abuse problems may be particularly appealing, compared to more traditional individual-based approaches; (2) describe theoretical and practical considerations involved when implementing couple therapy with these patients; (3) examine available evidence for the efficacy of couple therapy with female patients who abuse alcohol and drug; and (4) discuss future directions with respect to partner-involved therapies with these patients

    Work and Family Variables as Related to Paternal Engagement, Responsibility, and Accessibility in Dual-Earner Couples with Young Children

    Fathers and mothers (N = 75 dual-earner couples) of preschool-aged children completed questionnaires that examined work and family variables as related to paternal involvement in three areas: engagement (i.e., directly interacting with the child), responsibility (i.e., scheduling activities and being accountable for the child\u27s well-being), and accessibility (i.e., being available to the child but not in direct interaction). Fathers\u27 reports of responsibility and accessibility were significantly predicted by structural variables and beliefs; however, fathers\u27 reports of engagement were not predicted by work and family variables. Mothers\u27 reports of work and family variables did not predict their reports of father involvement. These findings suggest that for fathers of young children, parental involvement appears mainly self-determined

    Adolescent Opiate Exposure in the Female Rat Induces Subtle Alterations in Maternal Care and Transgenerational Effects on Play Behavior

    The non-medical use of prescription opiates, such as Vicodin® and MSContin®, has increased dramatically over the past decade. Of particular concern is the rising popularity of these drugs in adolescent female populations. Use during this critical developmental period could have significant long-term consequences for both the female user as well as potential effects on her future offspring. To address this issue, we have begun modeling adolescent opiate exposure in female rats and have observed significant transgenerational effects despite the fact that all drugs are withdrawn several weeks prior to pregnancy. The purpose of the current set of studies was to determine whether adolescent morphine exposure modifies postpartum care. In addition, we also examined juvenile play behavior in both male and female offspring. The choice of the social play paradigm was based on previous findings demonstrating effects of both postpartum care and opioid activity on play behavior. The findings revealed subtle modifications in the maternal behavior of adolescent morphine-exposed females, primarily related to the amount of time females’ spend nursing and in non-nursing contact with their young. In addition, male offspring of adolescent morphine-exposed mothers (MOR-F1) demonstrate decreased rough and tumble play behaviors, with no significant differences in general social behaviors (i.e., social grooming and social exploration). Moreover, there was a tendency toward increased rough and tumble play in MOR-F1 females, demonstrating the sex-specific nature of these effects. Given the importance of the postpartum environment on neurodevelopment, it is possible that modifications in maternal–offspring interactions, related to a history of adolescent opiate exposure, plays a role in the observed transgenerational effects. Overall, these studies indicate that the long-term consequences of adolescent opiate exposure can impact both the female and her future offspring

    Cytokine-facilitated transduction leads to low-level engraftment in nonablated hosts

    Using a murine bone marrow transplantation model, we evaluated the long-term engraftment of retrovirally transduced bone marrow cells in nonmyeloablated hosts. Male bone marrow was stimulated in a cocktail of interleukin-3 (IL-3), IL-6, IL-11, and stem cell factor (SCF) for 48 hours, then cocultured on the retroviral producer line MDR18.1 for an additional 24 hours. Functional transduction of hematopoietic progenitors was detected in vitro by reverse transcriptase-polymerase chain reaction (RT-PCR) amplification of multiple drug resistance 1 (MDR1) mRNA from high proliferative potential-colony forming cell (HPP-CFC) colonies. After retroviral transduction, male bone marrow cells were injected into nonablated female mice. Transplant recipients received three TAXOL (Bristol-Myers, Princeton, NJ) injections (10 mg/kg) over a 14-month period. Transplant recipient tissues were analyzed by Southern blot and fluorescence in situ hybridization for Y-chromosome-specific sequences and showed donor cell engraftment of approximately 9%. However, polymerase chain reaction amplification of DNAs from bone marrow, spleen, and peripheral blood showed no evidence of the transduced MDR1 gene. RT-PCR analysis of total bone marrow RNA showed that transcripts from the MDR1 gene were present in a fraction of the engrafted donor cells. These data show functional transfer of the MDR1 gene into nonmyeloablated murine hosts. However, the high rates of in vitro transduction into HPP-CFC, coupled with the low in vivo engraftment rate of donor cells containing the MDR1 gene, suggest that the majority of stem cells that incorporated the retroviral construct did not stably engraft in the host. Based on additional studies that indicate that ex vivo culture of bone marrow induces an engraftment defect concomitantly with progression of cells through S phase, we propose that the cell cycle transit required for proviral integration reduces or impairs the ability of transduced cells to stably engraft

    Development and characterization of a high-throughput in vitro cord formation model insensitive to VEGF inhibition

    BACKGROUND: Anti-VEGF therapy reduces tumor blood vessels, however, some vessels always remain. These VEGF insensitive vessels may help support continued tumor growth and metastases. Many in vitro assays examining multiple steps of the angiogenic process have been described, but the majority of these assays are sensitive to VEGF inhibition. There has been little focus on the development of high-throughput, in vitro assays to model the vessels that are insensitive to VEGF inhibition. METHODS: Here, we describe a fixed end-point and kinetic, high-throughput stem cell co-culture model of cord formation. RESULTS: In this system, cords develop within 24 hours, at which point they begin to lose sensitivity to VEGF inhibitors, bevacizumab, and ramucirumab. Consistent with the hypothesis that other angiogenic factors maintain VEGF-independent vessels, pharmacologic intervention with a broad spectrum anti-angiogenic antagonist (suramin), a vascular disrupting agent (combretastatin), or a combination of VEGF and Notch pathway inhibitors reduced the established networks. In addition, we used our in vitro approach to develop an in vivo co-implant vasculogenesis model that connects with the endogenous vasculature to form functional blood vessels. Similar to the in vitro system, over time these vessels become insensitive to VEGF inhibition. CONCLUSION: Together, these models may be used to identify novel drugs targeting tumor vessels that are not sensitive to VEGF inhibition

    Biodiversity baselines : tracking insects in Kruger National Park with DNA barcodes

    Reflecting their species richness and ecological diversification, insects play a central role in terrestrial ecosystems but difficulties in species-level assignments have restricted large-scale analysis of their community structure. Employing South Africa’s largest national park as a model system, we demonstrate that DNA barcoding can break this barrier. A year-long deployment of Malaise traps at 25 sites in Kruger National Park (KNP) generated 1000+ weekly collections containing about 800,000 specimens. Insect biomass averaged 1.05 g per trap-day but varied by up to 2-fold between months, being lower in the dry than wet season. Nearly 370,000 specimens were individually analyzed to reveal 19,730 Barcode Index Numbers (BINs; species proxy), a count equal to 43% of the known insect fauna of southern Africa. There was clear differentiation in insect richness and composition between KNP’s two ecoregions, but little among the vegetation types comprising them. The spatial gradient in annual rainfall explained more than half of the variation in compositional similarity among sites with less similarity among samples in the wet season, particularly among those in high rainfall areas. These results suggest that the factors organising insect communities in KNP are not fine-scale vegetation differences, but larger-scale processes associated with ecoregions and rainfall. Estimates of sample coverage indicate that the species not collected are rare, comprising only 4% of the individuals in the community. With a well-parameterized DNA barcode reference library in place, metabarcoding can be used to assess future shifts in the insect fauna of KNP rapidly and inexpensively.http://www.elsevier.com/locate/bioconpm2021Paraclinical Science

    Multiword Units Predict Non-inversion Errors in Children's Wh-questions: "What Corpus Data Can Tell Us?".

    Subject-auxiliary inversion in interrogatives has been a topic of great interest in language acquisition research, and has often been held up as evidence for the structure-dependence of grammar. Usage-based and nativist approaches posit different representations and processes underlying children’s question formation and therefore predict different causes for these errors. Here, we explore the question of whether input statistics predict children’s spontaneous non-inversion errors with wh- questions. In contrast to previous studies, we look at properties of the non-inverted, errorful forms of questions. Through a series of corpus analyses, we show that the frequency of uninverted subsequences (e.g., “she is going” in “what she is going to do?*”) is a good predictor of children’s errors, consistent with recent evidence for multiword units in children’s comprehension and production. This finding has implications for the types of mental representations and cognitive processes researchers ascribe to children acquiring a first language
