A Nonpolymorphic Class I Gene in the Murine Major Histocompatibility Complex
DNA sequence analysis of a class I gene (QlO), which maps to the Qa2,3 locus in the C57BL/lO (H-
2b haplotype) mouse, reveals that it is almost identical
to a cDNA clone (pH16) isolated from a SWR/J
(H-2q haplotype) mouse liver cDNA library. Exon 5,
in particular, has an unusual structure such that a
polypeptide product is unlikely to be anchored in the
cell membrane. Our findings suggest that the two
sequences are derived from allelic class I genes,
which are nonpolymorphic, in contrast to H-2K allelic
sequences from the same mice, and they may encode
liver-specific polypeptides of unknown function.
Our previous studies indicate that the QlO gene
is a potential donor gene for the generation of mutations
at the H-2K locus by inter-gene transfer of
genetic information. Thus the lack of polymorphism
in class I genes at the QlO locus implies either that
they are not recipients for such exchanges or that
selective pressure prevents the accumulation of mutations
in genes at this locus
Optical power transmission in a polygon mirror-based swept source optical coherence tomography system
Swept Source Optical Coherence Tomography (SS-OCT) relies on the rapid tuning of a broadband light source to produce narrow laser linewidths. Imaging speed is governed by the sweeping frequency of the source and the axial resolution is given by the total bandwidth generated. Mechanical, free space methods, employing rotating polygonal mirrors with a pair of telescopically arranged lenses, can achieve tuning speeds in excess of 100 kHz. Their success relies upon maximising the light throughput of the swept spectrum by reducing the effects of aberration and vignetting caused by the lens design and the geometrical properties of the polygon respectively. However, these properties impose constrictions on the spectral filter’s design and care must be taken when building the filter to avoid unnecessarily limiting the performance of the system. This paper presents some of the initial stages of a much larger study into the optimisation of such systems.
Theoretical work has been confirmed by experimental observations and compared with ideal simulations for a spectral filter consisting of a dispersive element, a double lens telescope, arranged in a Littman configuration, and a 72-facet, off-axis polygon mirror with end reflector. A non-linear relationship between the linewidth’s location on the telescope in time with the rotation of the polygon was observed and a first approximation for the tuned wavelength with respect to the polygon rotation angle was found. These observations, coupled with ongoing research, will lead to a complete description of polygon based scanners and how their performance can be optimised in future design
Substantiating a political public sphere in the Scottish press : a comparative analysis
This article uses content analysis to characterize the performance of the media in a national public sphere, by setting apart those qualities that typify internal press coverage of a political event. The article looks at the coverage of the 1999 devolved Scottish election from the day before the election until the day after. It uses a word count to measure the election material in Scottish newspapers the Herald, the Press and Journal and the Scotsman, and United Kingdom newspapers the Guardian, the Independent and The Times, and categorizes that material according to discourse type, day and page selection. The article finds a number of qualities that typify the Scottish sample in particular, and might be broadly indicative of a political public sphere in action. Firstly, and not unexpectedly, it finds that the Scottish newspapers carry significantly more election coverage. Just as tellingly, though, the article finds that the Scottish papers offer a greater proportion of advice and background information, in the form of opinion columns and feature articles. It also finds that the Scottish papers place a greater concentration of both informative and evaluative material in the period before the vote, consistent with their making a contribution to informed political action. Lastly, the article finds that the Scottish sample situates coverage nearer the front of the paper and places a greater proportion on recto pages. The article therefore argues that the Scottish papers display features that distinguish them from the UK papers, and are broadly consistent with their forming part of a deliberative public sphere, and suggests that these qualities might be explored as a means of judging future media performance
ART Suppresses Plasma HIV-1 RNA to a Stable Set Point Predicted by Pretherapy Viremia
Current antiretroviral therapy is effective in suppressing but not eliminating HIV-1 infection. Understanding the source of viral persistence is essential for developing strategies to eradicate HIV-1 infection. We therefore investigated the level of plasma HIV-1 RNA in patients with viremia suppressed to less than 50–75 copies/ml on standard protease inhibitor- or non-nucleoside reverse transcriptase inhibitor-containing antiretroviral therapy using a new, real-time PCR-based assay for HIV-1 RNA with a limit of detection of one copy of HIV-1 RNA. Single copy assay results revealed that >80% of patients on initial antiretroviral therapy for 60 wk had persistent viremia of one copy/ml or more with an overall median of 3.1 copies/ml. The level of viremia correlated with pretherapy plasma HIV-1 RNA but not with the specific treatment regimen. Longitudinal studies revealed no significant decline in the level of viremia between 60 and 110 wk of suppressive antiretroviral therapy. These data suggest that the persistent viremia on current antiretroviral therapy is derived, at least in part, from long-lived cells that are infected prior to initiation of therapy

An introduction to video methods in organizational research
Video has become a methodological tool of choice for many researchers in social science, but video methods are relatively new to the field of organization studies. This article is an introduction to video methods. First, we situate video methods relative to other kinds of research, suggesting that video recordings and analyses can be used to replace or supplement other approaches, not only observational studies but also retrospective methods such as interviews and surveys. Second, we describe and discuss various features of video data in relation to ontological assumptions that researchers may bring to their research design. Video involves both opportunities and pitfalls for researchers, who ought to use video methods in ways that are consistent with their assumptions about the world and human activity. Third, we take a critical look at video methods by reporting progress that has been made while acknowledging gaps and work that remains to be done. Our critical considerations point repeatedly at articles in this special issue, which represent recent and important advances in video method
Control and Manipulation of Pathogens with an Optical Trap for Live Cell Imaging of Intercellular Interactions
The application of live cell imaging allows direct visualization of the dynamic interactions between cells of the immune system. Some preliminary observations challenge long-held beliefs about immune responses to microorganisms; however, the lack of spatial and temporal control between the phagocytic cell and microbe has rendered focused observations into
the initial interactions of host response to pathogens difficult. This paper outlines a method that advances live cell imaging by integrating a spinning disk confocal microscope with an optical trap, also known as an optical tweezer, in order to provide exquisite spatial and temporal control of pathogenic organisms and place them in proximity to host cells, as
determined by the operator. Polymeric beads and live, pathogenic organisms (Candida albicans and Aspergillus fumigatus) were optically trapped using non-destructive forces and moved adjacent to living cells, which subsequently phagocytosed the trapped particle. High resolution, transmitted light and fluorescence-based movies established the ability to observe early events of phagocytosis in living cells. To demonstrate the broad applicability of this method to immunological studies, anti-CD3 polymeric beads were also trapped and manipulated to form synapses with T cells in vivo, and time-lapse imaging of synapse formation was also obtained. By providing a method to exert fine control of live pathogens with respect to immune cells, cellular interactions can be captured by fluorescence microscopy with minimal perturbation to cells and can

Bose-Einstein Correlations of Three Charged Pions in Hadronic Z^0 Decays
Bose-Einstein Correlations (BEC) of three identical charged pions were
studied in 4 x 10^6 hadronic Z^0 decays recorded with the OPAL detector at LEP.
The genuine three-pion correlations, corrected for the Coulomb effect, were
separated from the known two-pion correlations by a new subtraction procedure.
A significant genuine three-pion BEC enhancement near threshold was observed
having an emitter source radius of r_3 = 0.580 +/- 0.004 (stat.) +/- 0.029
(syst.) fm and a strength of \lambda_3 = 0.504 +/- 0.010 (stat.) +/- 0.041
(syst.). The Coulomb correction was found to increase the \lambda_3 value by
\~9% and to reduce r_3 by ~6%. The measured \lambda_3 corresponds to a value of
0.707 +/- 0.014 (stat.) +/- 0.078 (syst.) when one takes into account the
three-pion sample purity. A relation between the two-pion and the three-pion
