    Apocynum Tablet Protects against Cardiac Hypertrophy via Inhibiting AKT and ERK1/2 Phosphorylation after Pressure Overload

    Background. Cardiac hypertrophy occurs in many cardiovascular diseases. Apocynum tablet (AT), a traditional Chinese medicine, has been widely used in China to treat patients with hypertension. However, the underlying molecular mechanisms of AT on the hypertension-induced cardiac hypertrophy remain elusive. The current study evaluated the effect and mechanisms of AT on cardiac hypertrophy. Methods. We created a mouse model of cardiac hypertrophy by inducing pressure overload with surgery of transverse aortic constriction (TAC) and then explored the effect of AT on the development of cardiac hypertrophy using 46 mice in 4 study groups (combinations of AT and TAC). In addition, we evaluated the signaling pathway of phosphorylation of ERK1/2, AKT, and protein expression of GATA4 in the cardioprotective effects of AT using Western blot. Results. AT inhibited the phosphorylation of Thr202/Tyr204 sites of ERK1/2, Ser473 site of AKT, and protein expression of GATA4 and significantly inhibited cardiac hypertrophy and cardiac fibrosis at 2 weeks after TAC surgery (P<0.05). Conclusions. We experimentally demonstrated that AT inhibits cardiac hypertrophy via suppressing phosphorylation of ERK1/2 and AKT

    Triggered star formation surrounding Wolf-Rayet star HD 211853

    The environment surrounding Wolf-Rayet star HD 211853 is studied in molecular emission, infrared emission, as well as radio and HI emission. The molecular ring consists of well-separated cores, which have a volume density of 103^{3} cm3^{-3} and kinematic temperature \sim20 K. Most of the cores are under gravitational collapse due to external pressure from the surrounding ionized gas. From SED modeling towards the young stellar objects (YSOs), sequential star formation is revealed on a large scale in space spreading from the Wolf-Rayet star to the molecular ring. A small scale sequential star formation is revealed towards core A, which harbors a very young star cluster. Triggered star formations is thus suggested. The presence of PDR, the fragmentation of the molecular ring, the collapse of the cores, the large scale sequential star formation indicate the "Collect and Collapse" process functions in this region. The star forming activities in core A seem to be affected by the "Radiation-Driven Implosion" (RDI) process

    KMT2A promotes melanoma cell growth by targeting hTERT signaling pathway.

    Melanoma is an aggressive cutaneous malignancy, illuminating the exact mechanisms and finding novel therapeutic targets are urgently needed. In this study, we identified KMT2A as a potential target, which promoted the growth of human melanoma cells. KMT2A knockdown significantly inhibited cell viability and cell migration and induced apoptosis, whereas KMT2A overexpression effectively promoted cell proliferation in various melanoma cell lines. Further study showed that KMT2A regulated melanoma cell growth by targeting the hTERT-dependent signal pathway. Knockdown of KMT2A markedly inhibited the promoter activity and expression of hTERT, and hTERT overexpression rescued the viability inhibition caused by KMT2A knockdown. Moreover, KMT2A knockdown suppressed tumorsphere formation and the expression of cancer stem cell markers, which was also reversed by hTERT overexpression. In addition, the results from a xenograft mouse model confirmed that KMT2A promoted melanoma growth via hTERT signaling. Finally, analyses of clinical samples demonstrated that the expression of KMT2A and hTERT were positively correlated in melanoma tumor tissues, and KMT2A high expression predicted poor prognosis in melanoma patients. Collectively, our results indicate that KMT2A promotes melanoma growth by activating the hTERT signaling, suggesting that the KMT2A/hTERT signaling pathway may be a potential therapeutic target for melanoma

    Intrinsic Room Temperature Ferromagnetism in Boron-doped ZnO

    We report room temperature ferromagnetism in boron-doped ZnO both experimentally and theoretically. The single phase Zn1-xBxO films deposited under high oxygen pressure by pulsed-laser deposition show ferromagnetic behavior at room temperature. The saturation magnetization increases monotonously from 0 to 1.5 emu/cm3 with the increasing of B component x from 0 to 6.8%. The first-principles calculations based on density functional theory demonstrate that the ferromagnetism in B-doped ZnO originates from the induced magnetic moments of oxygen atoms in the nearest neighbor sites to the B-Zn vacancy pair. The calculated total magnetic moment increasing tendency with B component is well consistent with experiments.Comment: 16 pages, 1 table, 4 figures

    The effect of modified observation-teaching-discussion pedagogy combined with microteaching on the critical thinking ability of nursing interns in China: a quasi-experimental study

    AimThe aim of this study is to investigate the effect of modified observation-teaching-discussion pedagogy combined with microteaching on improving the critical thinking ability of nursing interns in China.MethodsTwo hundred baccalaureate nursing students practicing in the department of neurosurgery were divided into two groups chronologically, with 100 students per group. The control group received traditional clinical mentoring while their counterparts in the intervention group received mentoring by modified observation teaching discussion pedagogy combined with microteaching. Then, we compared the two groups with respect to theoretical knowledge and nursing operation skills. The Critical Thinking Ability Scale was used to evaluate the critical thinking ability of the two groups of nursing students. Data were analyzed with SPSS 22.0 software.ResultsFollowing the internship, the scores of nurses in the intervention group were significantly higher than those in the control group for theoretical knowledge, technical skills and critical thinking ability (p &lt; 0.05 for all).ConclusionThe application of modified observation-teaching-discussion pedagogy in combination with microteaching promoted the knowledge, technical skills and critical thinking ability of nursing students in clinical practice

    Design of the PMT underwater cascade implosion protection system for JUNO

    Photomultiplier tubes (PMTs) are widely used underwater in large-scale neutrino experiments. As a hollow glass spherelike structure, implosion is unavoidable during long-term operation under large water pressure. There is a possibility of cascade implosion to neighbor PMTs due to shockwave. Jiangmen Underground Neutrino Observatory designed a protection structure for each 20-inch PMT, consisting of a top cover, a bottom cover, and their connection. This paper introduces the requirement and design of the PMT protection system, including the material selection, investigation of manufacture technology, and prototyping.     Comparison of the effectiveness and safety of perampanel and oxcarbazepine as monotherapy in children and adolescents with newly diagnosed focal epilepsy

    Objective: This study aims to compare the effectiveness and safety of perampanel and oxcarbazepine as monotherapy in children with focal epilepsy (FE).Methods: This is an ambispective, single-center, non-inferiority study comparing the effectiveness and safety of perampanel (PER) monotherapy and oxcarbazepine (OXC) monotherapy in children with newly diagnosed FE. The primary endpoint was a six-month seizure freedom rate. The secondary endpoints included retention, responder, and seizure freedom rates at 3, 6, and 12 months, respectively. Adverse events (AEs) were also recorded for both groups.Results: One hundred and thirty children and adolescents aged from 4 to 18years newly diagnosed with FE between May 2020 and November 2022 in Wuhan Children’s Hospital were included. There were 71 patients in the PER group and 59 patients in the OXC group. In the per protocol set (PPS), 50 (78.1%) in the PER group and 43 (78.2%) in the OXC group completed six months of treatment without seizures. The lower 95% CI (66.0%–87.5%) limit of PER was higher than the non-inferiority margin of 62.4% (80% of the 6-month seizure freedom rate in the OXC group); PER was non-inferior to OXC. The 3-month and 12-month seizure freedom rates were 77.1% and 82.9% for the PER group, respectively, while they were 80.4% and 75.8% for the OXC group. There were no serious adverse events in both groups.Conclusion: PER showed comparable effectiveness and safety compared with OXC in children with newly diagnosed focal epilepsy, which might be an effective and safe treatment for children and adolescents with newly diagnosed FE.Clinical Trial Registration: Identifier ChiCTR230007469
